Lysosome-like particles in Geotrichum candidum : A cytochemical study

Lysosome-like particles were localized in Geotrichum candidum by means of the technique for the detection of acid phosphatase of PEARSE and staining with the vital dyes Brilliant cresyl blue and Neutral red. Pretreatments of the cells with procedures injuring the lysosome membrane resulted in shortening of the incubation time (2 min instead of 30 min). After exposure to 1% Triton X-100 the cells showed uniform strong staining due to the release of the enzyme into the cytoplasm.

Kinetics of alkaline phosphatase from pig kidney. Influence of complexing agents on stability and activity

Metal ion-complexing agents, like KCN, EDTA etc., inactivate alkaline phosphatase of pig kidney. This inactivation is reversible at low concentrations of the complexing agents and irreversible at high concentrations. The reversible inhibition is probably due to removal of Zn2+ ions from the active site, where they are necessary for catalytic action, whereas the irreversible inhibition results from the removal of Zn2+ ions necessary for preservation of the structure. The inactivation is pseudo-first order. It depends on the concentration, size and charge of the complexing agents. β-Glycerophosphate and Mg2+ ions protect the enzyme from inactivation by complexing agents. Quantitative examinat…

Zusammenfassung Mit Hilfe der vertikalen Discelektrophorese in Polyacrylamid-Gel ist es moglich, zwei Enzymbanden der sauren Spermaphosphatase nachzuweisen. Eine optimale Trennung ergibt sich bei einer Wasserstoffionenkonzentration von pH 7,5 im Trenn-Gel und pH 5,5 im Sammel-Gel und 0,1 M Acetatpuffer. Considerable attention has been given in recent years to the possibility that xenobiotics in the environment may affect reproduction in animals. In this study, the relative impact of tributyltin(IV) (TBT) chloride, one of the most toxic environmental pollutants, was investigated using Ciona intestinalis ovary as a model system. The pleiotropic effects of TBT exposure are concentration dependent and include a decrease of ATP levels, lipid content and nucleic acid content and synthesis. Adrenergic regulation of the pineal enzyme serotonin N-acetyltransferase [arylalkylamine N-acetyltransferase (AA-NAT); EC 2.3.1.87] accounts for the circadian rhythm in melatonin formation. In the present study, the role of protein phosphatases in the adrenergic regulation of rat pineal AA-NAT was investigated using specific inhibitors. Structural diversity and enzyme activity of volcanic soils at different stages of development and response to experimental disturbance

We investigated the phospholipid fatty acid (PLFA) diversity and enzyme activities in soils from the volcano, Mt. Etna (Sicily). The soils were at sites which have been developing for different periods of time and have formed in volcanic lava of differing ages that have been supplemented with volcanic ejecta from subsequent eruptions. However, the plant communities indicated a marked successional difference between the sites and we have used this as a proxy for developmental stage. Dissociation-constants of metat-ion-complexes with alkaline phosphatase from pig kidney.

Using metal-ion buffers it was possible to remove Zn2+, Mg2+ and Mn2+ ions of pig kidney alkaline phosphatase reversibly. The dissociation constants obtained are KEMg: 4 X 10(-7) M, KEMn: 4 X 10(-8) M and KEZn: 8 X 10(-13) M (22 degrees C, pH: 9.6, mu: 0.07).
