The Influence of Cultivation Conditions on the Proliferation and Differentiation of Rat Bone Marrow Multipotent Mesenchymal Stromal Cells
2008
Rat bone marrow multipotent mesenchymal stromal cells (rBM MMSCs) demonstrate a potential to differentiate down the mesodermal pathway but under appropriate experimental condition differentiation into non-mesodermal lineages may occur. Knowing this fact it seems interesting whether prolonged cultivation and preservation affects BM MMSC morphology, differentiation ability into multiple lineages and proliferation capacity. Therefore this study was aimed to investigate the effects of long-term cultivation and cryopreservation on the morphology, proliferation and differentiation potential of rat bone marrow-derived MMSCs in vitro.
Reduction of Cardiac Fibrosis by Interference With YAP-Dependent Transactivation
2022
Background: Conversion of cardiac stromal cells into myofibroblasts is typically associated with hypoxia conditions, metabolic insults, and/or inflammation, all of which are predisposing factors to cardiac fibrosis and heart failure. We hypothesized that this conversion could be also mediated by response of these cells to mechanical cues through activation of the Hippo transcriptional pathway. The objective of the present study was to assess the role of cellular/nuclear straining forces acting in myofibroblast differentiation of cardiac stromal cells under the control of YAP (yes-associated protein) transcription factor and to validate this finding using a pharmacological agent that interf…
Hypoxia-inducible factor 1Α may regulate the commitment of mesenchymal stromal cells toward angio-osteogenesis by mirna-675-5P
2017
Abstract Background aims During bone formation, angiogenesis and osteogenesis are regulated by hypoxia, which is able to induce blood vessel formation, as well as recruit and differentiate human mesenchymal stromal cells (hMSCs). The molecular mechanisms involved in HIF-1α response and hMSC differentiation during bone formation are still unclear. This study aimed to investigate the synergistic role of hypoxia and hypoxia-mimetic microRNA miR-675-5p in angiogenesis response and osteo-chondroblast commitment of hMSCs. Methods By using a suitable in vitro cell model of hMSCs (maintained in hypoxia or normoxia), the role of HIF-1α and miR-675-5p in angiogenesis and osteogenesis coupling was inv…
Bone Marrow Multipotent Mesenchymal Stromal Cells Transplantation Effects after Experimental Polytrauma in Rats
2013
The aim of this study was to evaluate impact of transplantation of bone marrow mesenchymal stromal cells on recovery after polytrauma and bone fracture repair.
2005
background: Gastrointestinal stromal tumors (GIST) are rare mesenchimal tumors that are characterized by constitutive overexpression of the tyrosin-kinase receptor KIT (CD 117). The authors analyse the basis of the treatment of gastric GIS, starting form the study of a recent observed case. Experimental design: Report of one case treated with endoscopic resection. Evaluation of treatment and 2-years follow- results: The treatment of gastric GIS I must be modulated on prognostic, genetic and molecular factors. These factors are the basis of the formation and growth of GIST. conclusions: Complete surgical extirpation without rupture remains the only curative treatment of localized favourable …
Stromal and Immune Cell Dynamics in Tumor Associated Tertiary Lymphoid Structures and Anti-Tumor Immune Responses
2022
Tertiary lymphoid structures (TLS) are ectopic lymphoid organs that have been observed in chronic inflammatory conditions including cancer, where they are thought to exert a positive effect on prognosis. Both immune and non-immune cells participate in the genesis of TLS by establishing complex cross-talks requiring both soluble factors and cell-to-cell contact. Several immune cell types, including T follicular helper cells (Tfh), regulatory T cells (Tregs), and myeloid cells, may accumulate in TLS, possibly promoting or inhibiting their development. In this manuscript, we propose to review the available evidence regarding specific aspects of the TLS formation in solid cancers, including 1) …
Taukaudu mezenhimālo stromālo šūnu iegūšana un raksturošana no SigmaR1 -/- pelēm
2020
Maģistra darba mērķis bija veikt mezenhimālo cilmes (stromālo) šūnu (MCŠ) izdalīšanu no Sigma-1 receptora (SigmaR1) inaktivētā gēna peļu taukaudiem un noteikt to adipodiferenciācijas potenciālu.
2021
