Toward minimal bacterial cells: evolution vs. design.

Abstract Recent technical and conceptual advances in the biological sciences opened the possibility of the construction of newly designed cells. In this paper we review the state of the art of cell engineering in the context of genome research, paying particular attention to what we can learn on naturally reduced genomes from either symbiotic or free living bacteria. Different minimal hypothetically viable cells can be defined on the basis of several computational and experimental approaches. We investigated the immune response induced by mucosal immunization of BALB/c mice with virus-like particles (VLPs) of a genogroup II norovirus, Dijon171/96 virus, produced in the baculovirus system. VLPs administered alone by the intranasal route induced a high serum antibody response as well as fecal IgA, which were enhanced when the heat-labile Escherichia coli toxin or its non toxic mutant LT(R192G) was coadministered. In these conditions, the oral route was also efficient. Cytokine production by cells from different lymphoid tissues was then assessed after in vitro restimulation. The construction of artificial life is one of the main scientific challenges of the Synthetic Biology era. Advances in DNA synthesis and a better understanding of regulatory processes make the goal of constructing the first artificial cell a realistic possibility. The effect of light curing units and modes on cytotoxicity of resin-core systems

Objective: The aim of this study was to compare the cytotoxic effects of various resin-based core materials that were cured with three light curing units (LCUs) in different modes on L?929 mouse fibroblast cells over 24 h and 72 h periods. Study design: Eighty-four cylindrical discs (2 mm in thickness and 6 mm in diameter) of each material (Rebilda, Voco; Build-It FR, Pentron; Clearfil DC Core, Kuraray and Bis-core, Bisco) were cured by QTH LCU (soft-up and high-power modes), LED LCU (exponential and standard modes) and PAC LCU (normal and ramp-curing modes). Evaluation of different InSAR multi-baseline construction methods over a dam in southern Italy

Monitoring dam displacements using different techniques allows an evaluation of their structural behaviour over time. In this study, dam displacements (for the Castello dam, Agrigento, Italy) have been investigated using different Interferometric Synthetic Aperture Radar (InSAR) techniques exploiting a freely available dataset from the EU Copernicus Sentinel-1 SAR built by the European Space Agency (ESA). A Polyphenylene Dendrimer Drug Transporter with Precisely Positioned Amphiphilic Surface Patches

The design and synthesis of a polyphenylene dendrimer (PPD 3) with discrete binding sites for lipophilic guest molecules and characteristic surface patterns is presented. Its semi-rigidity in combination with a precise positioning of hydrophilic and hydrophobic groups at the periphery yields a refined architecture with lipophilic binding pockets that accommodate defined numbers of biologically relevant guest molecules such as fatty acids or the drug doxorubicin. The size, architecture, and surface textures allow to even penetrate brain endothelial cells that are a major component of the extremely tight blood-brain barrier. The development of synthetic antitumour vaccines from mucin glycopeptide antigens.

Based on important cell-biological and biochemical results concerning the structural difference between membrane glycoproteins of normal epithelial cells and epithelial tumour cells, tumour-associated glycopeptide antigens have been chemically synthesised and structurally confirmed.
