Structure of SNX9 SH3 in complex with a viral ligand reveals the molecular basis of its unique specificity for alanine-containing class I SH3 motifs

Class I SH3 domain-binding motifs generally comply with the consensus sequence [R/K]x0PxxP, the hydrophobic residue 0 being proline or leucine. We have studied the unusual 0 = Ala-specificity of SNX9 SH3 by determining its complex structure with a peptide present in eastern equine encephalitis virus (EEEV) nsP3. The structure revealed the length and composition of the n-Src loop as important factors determining specificity. We also compared the affinities of EEEV nsP3 peptide, its mutants, and cellular ligands to SNX9 SH3. The primary structural photoresponse of phytochrome proteins captured by a femtosecond X-ray laser

Phytochrome proteins control the growth, reproduction, and photosynthesis of plants, fungi, and bacteria. Light is detected by a bilin cofactor, but it remains elusive how this leads to activation of the protein through structural changes. We present serial femtosecond X-ray crystallographic data of the chromophore-binding domains of a bacterial phytochrome at delay times of 1 ps and 10 ps after photoexcitation. The data reveal a twist of the D-ring, which leads to partial detachment of the chromophore from the protein. A spontaneous mitonuclear epistasis converging on Rieske Fe-S protein exacerbates complex III deficiency in mice

We previously observed an unexpected fivefold (35 vs. 200 days) difference in the survival of respiratory chain complex III (CIII) deficient Bcs1lp.S78G mice between two congenic backgrounds. Here, we identify a spontaneous homoplasmic mtDNA variant (m.G14904A, mt-Cybp.D254N), affecting the CIII subunit cytochrome b (MT-CYB), in the background with short survival. We utilize maternal inheritance of mtDNA to confirm this as the causative variant and show that it further decreases the low CIII activity in Bcs1lp.S78G tissues to below survival threshold by 35 days of age. [EN] Why do films certain remain influential throughout film history? The purpose of this paper is to attempt to answer this question. To do so, we adopt some quantitative approaches that facilitate an objective interpretation of the data. The data source we have chosen for this study is the Internet Online Movie Database (IMDb), and in particular, one of its sections called "Connections", which lists references made to a film in subsequent movies and references made in the film itself to previous ones. We investigated the spatial and temporal variation in denitrification rates (isotopepairing technique, IPT) and in the denitrifier community (examination of gene nirK by denaturinggradient gel electrophoresis [DGGE] of microbial DNA) in the sediments of a boreal, clear-water, eutrophic lake using samples collected from shallow littoral, deep littoral and shallow profundal sediments during early summer, mid-summer, autumn and winter. The measured denitrification rates (44 to 613 μmol N m−2 d−1) are among the lowest ever reported from lacustrine sediments. This paper empirically investigates and characterizes users of recorded music, both downloaders and purchasers. To this end we analyse the role of the variables defining the different segments of music users. In doing so, we have considered two main traits influencing the use of music. First, objective variables such as demographics, music consumption habits, music genres and technology. Second, subjective variables such as motives and attitudes towards piracy.
