Abstract Senescence is manifested by an increase in molecular damage and a deterioration of biological functions with age. In most organisms, body maintenance is traded-off with reproduction. This negative relationship between longevity and fecundity is also evident on the molecular level. Regulatory mechanisms of estrogen on vascular ageing

Women can be considered hemodynamically younger than men of the same age, based on epidemiological studies establishing that the incidence of vascular diseases in women is relatively lower compared to that in aged-matched men. However, after menopause, these numbers increase to values that are close to those found in men. Vascular ageing is associated with structural and functional changes of the vascular wall, including endothelial dysfunction, arterial stiffening, and remodelling, as well as impaired angiogenesis, which become major risk factors in the development of cardiovascular disease.

LRP1 mediates bidirectional transcytosis of amyloid-β across the blood-brain barrier.

According to the "amyloid hypothesis", the amyloid-β (Aβ) peptide is the toxic intermediate driving Alzheimer's disease (AD) pathogenesis. Recent evidence suggests that the low density lipoprotein receptor-related protein 1 (LRP1) transcytoses Aβ out of the brain across the blood-brain barrier (BBB). To provide genetic evidence for LRP1-mediated transcytosis of Aβ across the BBB we analyzed Aβ transcytosis across primary mouse brain capillary endothelial cells (pMBCECs) derived from wild-type and LRP1 knock-in mice. Neural stem cells (NSCs) persist in the adult mammalian brain through life. The subventricular zone (SVZ) is the largest source of stem cells in the nervous system, and continuously generates new neuronal and glial cells involved in brain regeneration. During aging, the germinal potential of the SVZ suffers a widespread decline, but the causes of this turn down are not fully understood. Telomere Shortening in Neural Stem Cells Disrupts Neuronal Differentiation and Neuritogenesis

Proliferation in the subependymal zone (SEZ) and neurogenesis in the olfactory bulb decline in the forebrain of telomerase-deficient mice. The present work reveals additional effects of telomere shortening on neuronal differentiation, as adult multipotent progenitors with critically short telomeres yield reduced numbers of neurons that, furthermore, exhibit underdeveloped neuritic arbors. Genetic data indicate that the tumor suppressor protein p53 not only mediates the adverse effects of telomere attrition on proliferation and self-renewal but it is also involved in preventing normal neuronal differentiation of adult progenitors with dysfunctional telomeres.
