Rapid Differentiation of a Rare Subset of Adult Human Lin−CD34−CD38− Cells Stimulated by Multiple Growth Factors In Vitro

1999

Recently, several reports of lineage-negative (lin(-)) CD34(-) cells with in vivo hematopoietic activity have focused interest on the properties and growth factor response characteristics of these cells. We have now identified a combination of 5 growth factors that are necessary and sufficient to stimulate a marked mitogenic and differentiation response by a subset of human lin(-)CD34(-)CD38(-) cells present in normal adult human marrow and granulocyte colony-stimulating factor (G-CSF)-mobilized blood. Less than 0.1% of the cells in highly purified (including doubly sorted) lin(-)CD34(-)CD38(-) cells from these 2 sources formed colonies directly in semisolid medium or generated such cells a…

Candida albicans triggers proliferation and differentiation of hematopoietic stem and progenitor cells by a MyD88-dependent signaling.

2009

As TLRs are expressed by hematopoietic stem and progenitor cells, these receptors may play a role in hematopoiesis in response to pathogens during infection. We showed here that inactivated yeasts and hyphae of Candida albicans induce in vitro the proliferation of purified murine hematopoietic stem and progenitor cells (Lin(-)c-Kit(+) Sca-1(+)) as well as their differentiation to lineage positive cells, through a MyD88-dependent pathway. These results indicate that TLR-mediated recognition of C. albicans by hematopoietic stem and progenitor cells may augment the host capability for rapidly replenishing the innate immune system during candidiasis.

CNS manifestations of Fabry's disease

2006

Summary Background Fabry's disease is a rare hereditary lysosomal storage disease with multiorgan involvement. Deficiency of α-galactosidase A activity leads to accumulation of neutral glycosphingolipids, especially in vascular endothelial and smooth-muscle cells. Along with progressive renal and cardiac dysfunction, stroke is a major and often life-threatening burden of the disease. Cerebral vasculopathy, confirmed by neuropathological, neuroradiological, and functional studies, occurs commonly and leads to ischaemic cerebrovascular events at an early age. Recent developments Fabry's disease is an X-linked disease and women have been regarded as only mildly affected carriers. However, rese…

Hyper-IL-6 (H-IL-6), a Fusion Protein of Soluble IL-6 Receptor (Sil-6R), and Interleukin-6 (IL-6), Acts Synergistic with Thrombopoietin (TPO) and Ste…

1999

It has been shown that signalings from c-kit and gpl30, the signal-transducing receptor component of the IL6 receptor, act synergistic for the ex-vivo expansion of multipotential hematopoietic progenitors. A similar synergistic effect has been demonstrated for signalings from c-kit, c-mpl, and flt3. While c-kit is activated by stem cell factor (SCF), gpl30 can be activated by the complex of soluble IL-6 receptor (sIL-6R) and interleukin-6 (IL-6). Recently, a bioactive designer cytokine, H-IL-6, a fusion protein consisting of SIL-6R and IL-6 linked by a flexible peptide chain has been shown to expand human hematopoietic colony-forming cells. We tested the activity of H-IL-6 alone and in comb…

Can Tumor Oxygenation be Improved by Reducing Cellular Oxygen Consumption?

1999

Tumor hypoxia, which can be found in many experimental and human tumors is an important factor influencing the therapeutic efficacy of standard radiotherapy, O2-dependent chemotherapy and photodynamic therapy (Hall, 1994) and might be responsible for the development of aggressive tumor cell subpopulations (Graeber et al., 1996). Since this oxygen deficiency results from a disparity between O2 supply to the tumor tissue and the oxygen demand of the cells, several attempts have been undertaken to improve tumor oxygenation primarily by increasing the arterial oxygen supply. The O2 supply to the tumor cells can be improved by (a) increasing the arterial O2 content (by breathing hyperoxic gases …

742 EFFICACY AND SAFETY OF ENTECAVIR (ETV) PROPHYLAXIS IN INACTIVE HBV CARRIERS WHO UNDERWENT CHEMOTHERAPY FOR SOLID OR HAEMATOLOGICAL CANCER: INTERI…

2013

seroconversion rate at 48 weeks were 40.7% and 37% in PEG-IFN a-2a group, respectively, which are both higher than those in ETV group (16.7% and 13.3%, P all 0.05). The mean qHBsAg level in PEG-IFN a-2a group declined over time during treatment. The qHBsAg level at 48 weeks was significantly lower than that in ETV group ((2866.0±2580.4) vs (4335.8±2650.0) IU/mL, P = 0.027). A greater HBsAg decline was observed in HBeAg seroconverters compared with nonseroconverters. The decline from baseline was significantly different between seroconverters and non-seroconverters especially at 36 weeks ((1763.4±3116.2) vs (1333.5±2483.4) IU/mL, P = 0.036), and 48 weeks (1979.6±2897.1) vs (1631.8±2395.8) IU…

Alkaline haematin D-575, a new tool for the determination of haemoglobin as an alternative to the cyanhaemiglobin method. I. description of the method

1984

A new method for the rapid and accurate measurement of haemoglobin has been developed as an alternative to the conventional cyanhaemiglobin method. This method is based on the conversion of all haeme, haemoglobin, and haemiglobin species into a stable end product by an alkaline solution of a non-ionic detergent ('AHD reagent'). The reaction product, designated as alkaline haematin D-575, is extremely stable and shows a characteristic absorption peak at 575 nm. As compared to the cyanhaemiglobin method, the determination of haemoglobin by alkaline haematin D-575 offers several advantages such as (1) extreme stability of the AHD reagent and the conversion product, (2) decreased conversion tim…

Characterisation of phenolic compounds in Algerian honeys by RP-HPLC coupled to electrospray time-of-flight mass spectrometry

2017

Abstract A total of 35 honey samples from different regions of Algeria were studied to determine their phenolic profiles. Phenolic compounds, products of the secondary metabolism of plants, were extracted with amberlite XAD-4 and analysed by liquid chromatography, with diode array detection and electrospray ionisation mass spectrometry in negative ion polarity. By using colorometric assays, Erica honeys showed the highest content of phenolic compounds and flavonoids (245 ± 54 mg GAE/100 g and 29 ± 6 mg QE/100 g, respectively). More than 30 compounds were identified in the honey samples studied including 14 phenolic acids and 16 flavonoids. In general, honey samples showed different chromato…
