The development of chronic airway inflammation depends upon the recruitment and activation of inflammatory cells and the subsequent release of inflammatory mediators, including cytokines. Background: Pregnant women with asthma need to take medication during pregnancy.Objective: We sought to identify whether there is an increased risk of specific congenital anomalies after exposure to antiasthma medication in the first trimester of pregnancy.Methods: We performed a population-based case-malformed control study testing signals identified in a literature review. 1. Extracts of adrenal glands and heart tissue of the cat were prepared and chromatographed on paper. The regions corresponding to the RF values of noradrenaline, adrenaline and isoprenaline (isopropylnoradrenaline) were eluted. The eluates were assayed on the blood pressure, the isolated auricle and the isolated uterus of the rat. Differential assay on these preparations proved to be specific and extremely sensitive for isoprenaline. 2. There was no evidence for the occurrence of isoprenaline in the adrenal medulla or the heart of the cat. The method used is sufficiently sensitive to detect 0,2% of isoprenaline in the total catechol amines of the heart and 0,007% in the adrenals.

Isolated rabbit hearts were perfused with Tyrode solution containing 10 ng/ml (-)-noradrenaline for 20 min. The uptake by the heart of noradrenaline was calculated by estimating the percentage of amine removed from the perfusion fluid during a single passage through the heart. Previous experiments have shown that the amount of noradrenaline removed from the perfusion fluid corresponds well to the amount retained by the heart. In control experiments, the mean removal (elimination) of noradrenaline was 50±1.7% of the amount infused as measured biologically and 46±2.2% as measured fluorimetrically. Groups of normal rats of both sexes in moderate water diuresis received i.v. injections of 5 ml./kg, of hypertonic (5 times isoosmotic) solutions of sodium chloride, mannitol er sodium sulfate. The renal excretion of water, sodium, chloride and potassium in a 30 minute-period following this injection was measured. It was compared to control values obtained 40 minutes earlier in the same animals after an i.v. injection of isotonic glucose solution. All hypertonic solutions increased diuresis, natriuresis, chloruresis and kaliuresis. Water, sodium, potassium and chloride balances, food uptake and changes in body weight were determined in large groups of adrenalectomized or sham-operated rats having permanent access to water, 0.9 per cent saline and rat crackers. Measurements were done on the 5th to 8th day, or else on the 11th to 14th day after operation. The inhibition of 11-β-hydroxylase in the adrenal cortex with SU 4885 (metopiron) increases the ACTH-secretion. Val5-angiotensin II blocks the reaction of the anterior pituitary gland similarly as does dexamethasone. Mode and intensity of this action of angiotensin II, and the influence on rats treated as described, are studied.
