Development of an Efficient In Vivo System (P-junc-TpaseIS(1223)) for Random Transposon Mutagenesis of Lactobacillus casei

ABSTRACT The random transposon mutagenesis system P junc -TpaseIS 1223 is composed of plasmids pVI129, expressing IS 1223 transposase, and pVI110, a suicide transposon plasmid carrying the P junc sequence, the substrate of the IS 1223 transposase. This system is particularly efficient in Lactobacillus casei , as more than 10,000 stable, random mutants were routinely obtained via electroporation.

ABSTRACT Cover crops may improve vineyard soil properties, grapevine nutrient status and berry composition, however, factors such as cover crop type, annual rainfall, climate and irrigation may change their effects on vineyards. From 2008 to 2011, the effects of a non-permanent cover crop and two pruning techniques on soil as well as vine nutrients and grapevine performance of two vineyards (cv. Tempranillo and cv. Little information is available about either nitrogen (N) content/concentration in table grape vines. The knowledge about N accumulated by the vine organs during the season could support sustainable fertilization programs for table grape vineyards. Aim: Based on consensus rankings from prominent rating authorities, we examined the importance of a suite of climatic variables, organized by winegrape phenological stage, in distinguishing between high- and low-ranked vintages in Burgundy.Methods and Results: Vintage ratings of Burgundy wines acquired from 12 sources were evaluated to develop consensus rankings for red and white wines from 1961–2015. The English walnut (Juglans regia L.) is the main species cultivated for the production of edible nuts. Owing to a sparse canopy and a deep rooting system, walnut is an ideal species for alley cropping, an agroforestry practice able to enhance productivity through interplant facilitative mechanisms. Walnut agroforestry requires the large scale production of seedling rootstocks selected to provide the best anchorage, vigour, and tolerance of pathogens. Due to the heterozygosity of walnut, the characteristics of agronomical interest of the chosen cultivar are not inherited via seed propagation.
