Chemometric extraction of analyte-specific chromatograms in on-line gradient LC-infrared spectrometry
2009
This work exploits the possibilities offered by the recently developed multivariate method named Science-Based Calibration (SBC), for the extraction of ‘analyte-specific’ chromatograms in on-line gradient reversed phase LC-infrared spectrometry (IR) in the presence of a high spectral and chromatographic overlapping between the analyte of interest, co-eluting sample matrix constituents and the mobile phase components. The SBC method uses an experimentally measured single response spectrum of the analyte of interest and representative noise to calculate an optimum regression vector (bopt(1)). Then, the bopt(1) vector is used to predict the concentration of the analyte of interest in the spect…
Evaluation of infrared spectroscopy as a screening tool for serum analysis
2013
Abstract The application of attenuated total reflectance Fourier transform infrared (ATR-FT-IR) spectroscopy to the determination of clinical parameters in serum using partial least squares (PLS) has been evaluated as a point-of-care diagnostic tool. In this study the effect of using an increased size of the calibration set and the influence of the origin of samples and their interyear variation on the prediction capability of the method were considered. PLS-ATR-FT-IR provides a green, fast and cheap point-of-care tool for the determination of total protein. Albumin, glucose, urea, HDL, LDL and total cholesterol were predicted with relative errors between 15 and 32%. The analytical predicti…
Infrared-based quantification of clinical parameters
2014
Abstract Infrared (IR) spectroscopy has become a promising technique for the analysis of clinical samples, and IR-based diagnosis is already a well-established technique in the academic field. This review aims to report the methodologies proposed for IR quantification of different analytes and parameters in a variety of clinical samples; including: liquids, such as whole blood, serum, plasma, urine, human milk, amniotic fluid and cerebrospinal fluid; solids, such as urinary stones, skin, hair and lips; breath; or, microscopic quantifications on tissues. For the application of IR spectroscopy to the clinical field, we also discuss: the benefits, which make IR spectroscopy ideally suited to m…
Net analyte signal as a deconvolution-oriented resolution criterion in the optimisation of chromatographic techniques
2003
The performance of two multivariate calibration measurements, multivariate selectivity (SEL(s)) and scalar net analyte signal (scalar NAS), as chromatographic objective functions (COFs), was investigated. Since both assessments are straightforwardly related to the quantification of analytes in the presence of interferents, they were expected to confer new features in the optimisation of compound resolution, not present in conventional assessments. These capabilities are especially interesting in situations of low resolution, where peak deconvolution becomes an attractive alternative. For comparison purposes, chromatographic resolution (R(s)) and peak purity (p(s)) were used as reference COF…
Enantiomeric separation of chiral phenoxy acid herbicides by electrokinetic chromatography. Application to the determination of analyte-selector appa…
2001
The enantiomeric resolution of chiral phenoxy acid herbicides was performed by electrokinetic chromatography using a cyclodextrin as chiral pseudophase (CD-EKC). A systematic evaluation of several neutral and charged cyclodextrins was made. Among the cyclodextrins tested, (2-hydroxy)propyl beta-cyclodextrin (HP-beta-CD) was found to be the most appropriate for the enantioseparation of phenoxy acids. The influence of some experimental conditions, such as nature and pH of the background electrolyte, chiral selector concentration, and temperature, on the enantiomeric separation of phenoxy acids was also studied. The use of a 50 mM electrolyte solution in ammonium formate at pH 5 and a temperat…
Fast enantiomeric separation of uniconazole and diniconazole by electrokinetic chromatography using an anionic cyclodextrin: application to the deter…
2000
The enantiomeric resolution of the fungicides uniconazole and diniconazole was performed using electrokinetic chromatography with cyclodextrins as pseudostationary phase (CD-EKC). A systematic evaluation of several chiral selectors was made. The anionic derivative carboxymethylated-gamma-cyclodextrin (CM-gamma-CD) was found to be the most appropriate for the enantioseparation of fungicides among all cyclodextrins tested. The influence of some experimental conditions such as nature and buffer pH, chiral selector concentration, and temperature on the enantiomeric separation of the compounds studied was also investigated. The use of a 50 mM phosphate buffer (pH 6.5) containing 5 mM CM-gamma-CD…
Immunoanalytical approaches for the control of xenobiotics and biotoxins in food
2020
Determination of Phenol by Preconcentration‐Direct Chemiluminescence in a FIA Assembly
2005
Abstract The determination of phenol in water samples is proposed with the aid of a flow‐injection system. The analytical procedure is based on the direct chemiluminometric emission by oxidation of the analyte with potassium permanganate in acidic medium. The flow assembly is provided with a solid‐phase reactor filled with a resin type XAD‐4 for analyte preconcentration. A large study of potential interferences, namely, amino acids reaching water through degradation of organic matter; metals and inorganic metal ions typically present in water interfering with the CL emission of the parent compound, was performed. The calibration graph was linear over the phenol concentration range 1.0–20.0 …
Analysis of drugs including illicit and new psychoactive substances in oral fluids by gas chromatography-drift tube ion mobility spectrometry
2021
Abstract In this study, a gas chromatograph (GC) has been coupled to a drift tube ion mobility spectrometer (IMS) in order to develop an analytical procedure for the determination of psychoactive substances in oral fluids. Working parameters, including the GC-IMS interface ones, were adjusted in order to obtain sensitive and robust signals. A volume of 500 μL of oral fluid was extracted with 250 μL chloroform and, after centrifugation, were injected into the GC-IMS system. Amphetamine, methylone, α-PVP, ketamine, lidocaine, MPHP, cocaine, THJ-2201, and 5F-ADB were employed as model compounds, providing limits of detection from 6 to 15 μg L−1 and recoveries from 70 to 115% for field oral flu…
Determination of tea tree oil terpenes by headspace gas chromatography mass spectrometry
2016
An alternative procedure was developed for determining quality control of terpenes in tea tree oil commercial samples. The method is based on static headspace gas chromatography mass spectrometry measurements. Absolute limits of detection ranged from 1 ng for p-cymene to 71 ng for terpinen-4-ol. The accuracy of the developed methodology was evaluated by comparing the obtained eucalyptol and terpinen-4-ol concentrations with those provided for a certified sample and by comparing the developed procedure with literature tea tree oil analysis methodology. In the same way, spiked commercial samples provided recovery values ranging between 94% and 100% for all the target analytes with relative st…
