Rethinking Cysteine Protective Groups:S-Alkylsulfonyl-l-Cysteines for Chemoselective Disulfide Formation

The ability to reversibly cross-link proteins and peptides grants the amino acid cysteine its unique role in nature as well as in peptide chemistry. We report a novel class of S-alkylsulfonyl-l-cysteines and N-carboxy anhydrides (NCA) thereof for peptide synthesis. The S-alkylsulfonyl group is stable against amines and thus enables its use under Fmoc chemistry conditions and the controlled polymerization of the corresponding NCAs yielding well-defined homo- as well as block co-polymers. Yet, thiols react immediately with the S-alkylsulfonyl group forming asymmetric disulfides. Hydrogels for potential colon drug release by thiol-ene conjugate addition of a new inulin derivative.

Inulin was chosen as a starting polymer for biocompatible, pH-sensitive and biodegradable hydrogels. Three INUDVSA-TT hydrogels were obtained by crosslinking inulin derivatives with trimethylolpropane tris(3-mercaptopropionate) under varying conditions. The resulting hydrogels were cell compatible, as demonstrated by MTS and trypan blue exclusion assays acting on Caco-2 cells, and were biodegraded by inulinase and esterase, thus suggesting their use as colonic drug delivery systems. 2-Methoxyestradiol, an anti-cancer drug, was soaked in INUDVSA-TT hydrogels and its in vitro release and apoptotic effect on Caco-2 cells were evaluated.

An efficient acetylation of dextran using in situ activated acetic anhydride with iodine

A facile, efficient, cost-effective and solvent-free acetylation method has been developed for the acetylation of dextran. Dextran acetates were successfully synthesized using different molar ratios of acetic anhydride in the presence of iodine as a catalyst without the use of any solvent. The reactions were realized at 50?C for 3 h under stirring and nitrogen. This efficient method yielded highly pure and organosoluble dextran esters. The reaction appears highly effective for obtaining higher degrees of substitution (DS) with great efficiency. Under solvent-free conditions, dextran triacetates were efficiently synthesized. The preparation and physicochemical and biological characterization of a novel polyaminoacid hydrogel have been reported. The ,-poly(N-2- hydroxyethyl)-dl-aspartamide (PHEA) has been used as a starting polymer for a derivatization reaction with methacrylic anhydride (MA) to give rise to the methacrylate derivative named PHM. Photo-Cross-Linked Hydrogels with Polysaccharide−Poly(amino acid) Structure:  New Biomaterials for Pharmaceutical Applications

The aim of this work has been the preparation and characterization of novel hydrogels with polysaccharide-poly(amino acid) structure having suitable physicochemical properties for pharmaceutical applications. In the first step, hyaluronic acid (HA) and alpha,beta-poly(N-2-hydroxyethyl)-DL-aspartamide (PHEA) have been derivatized with methacrylic anhydride (AMA), thus obtaining HA-AMA and PHM derivatives, respectively. In the second step, aqueous solutions of both these derivatives have been irradiated at 313 nm to obtain chemical hydrogels. Dynamic mechanical properties of semi-interpenetrating networks based on poly(styrene-co-maleic anhydride)

Semi-interpenetrating networks based on linear anionic polystyrene (PS) and poly(styrene-co-maleic anhydride) P(ScoMA) of low MA content are prepared by crosslinking the P(ScoMA) with 4,4′-diphenylmethanediamine (DPMDA). It is demonstrated by dynamic mechanical analysis and by DSC that immiscible semi-IPN's are obtained for MA contents greater 5 wt.-%. In miscible semi-IPN's the relaxation of free PS chains is observed as a second maximum in the loss tangent at higher temperatures. The position of this maximum is shifted to higher temperatures for a polystyrene of molecular weight 500000 compared to a polystyrene of molecular weight 150000. Gas chromatography-mass spectrometry characterization of the varnish and glue of an ancient 18th century double bass.

A GC–MS investigation is conducted on the double bass “Panormus”, property of Conservatorio di Musica “Vincenzo Bellini” in Palermo. The most important components of the varnish (fatty acids) and of the glue (proteinaceous amino acids), with which the musical instrument was treated in the past, are determined. The analyses are carried out by prior derivatization of fatty acids by acidic methanol and of amino acids by acidic methanol and trifluoroacetic anhydride (TFAA). Analytes identification is achieved by direct comparison with several reference materials and the use of a digitized library.

Efficient and Mild Ytterbium(III)-Catalyzed Tosylation of Alcohols.

Ytterbium(III) trifluoromethanesulfonate efficiently catalyzes the reaction of primary and secondary alcohols with toluenesulfonic acid anhydride to yield the alkyl tosylates in high yields. The reactions were carried outunder neutral and mild conditions and product purification was easily achieved by means of short column chromatography.
