International audience; Vinblastine and vincristine are dimeric indole alkaloids derived from (formerly: ). Their monomeric precursor molecules are vindoline and catharanthine. While vinblastine and vincristine are well-known mitotic spindle poisons, not much is known about vindoline and catharanthine. Vindoline and catharanthine showed weak cytotoxicity, while vinblastine, vincristine, and the semisynthetic vindesine and vinorelbine revealed high cytotoxicity towards cancer cells. However, the multielement apparatus reconstructions of most species remain equivocal because they have been inferred from loose element collections, guided by knowledge from rare articulated ‘bedding plane assemblages’ and fused clusters, often from distantly related taxa. The surface force apparatus has been used to quantify directly the forces that govern the interactions between proteins and ligands. In this work, we describe the measured interactions between the antigen fluorescein and the Fab' fragment of the monoclonal 4-4-20 anti-fluorescyl IgG antibody. Here we first describe the use of the surface force apparatus to demonstrate directly the impact of the charge composition in the region of the antibody binding site on the antibody interactions. Publisher Summary This chapter provides an overview of what is known about carotenoid metabolism in Chlamydomonas with reference to other green algae and vascular plants. The biosynthesis of carotenoids and the subdivisions of carotenogenesis are described. With respect to subcellular distribution, the carotenoids in vegetative cells localize to the chloroplast where they either serve as photosynthetic pigments bound to the protein complexes of the two photosystems, or as components of the eyespot apparatus is outlined with its functional significance. Carotenoids can serve as precursors of a number of other molecules with important physiological functions in Chlamydomonas.
