Synthesis and anti-HIV activity of 2,3-diaryl-1,3-thiazolidin-4-ones

Several 1,3-thiazolidin-4-ones bearing a 2,6-dihalophenyl group at C-2 and a variously substituted phenyl ring at N-3 have been synthesized and tested as anti-HIV agents. The results of the in vitro tests showed that some of them proved to be effective inhibitors of HIV-1 replication.

Several 1,3-thiazolidin-4-ones bearing a 2,6-dihalophenyl group at C-2 and a variously substituted phenyl ring at N-3 have been synthesized and tested as anti-HIV agents. The results of the in vitro tests showed that some of them proved to be effective inhibitors of HIV-1 replication.

Synthesis of new 2,3-diaryl-1,3-thiazolidin-4-ones as anti-HIV agents

Several 2,3-diaryl-1,3-thiazolidin-4-ones were synthesized and evaluated as anti-HIV agents. The results of the in vitro tests showed that some of them were highly effective inhibitors of HIV-1 replication at 30-50 nM concentrations with minimal cytotoxicity, thereby acting as non-nucleoside HIV-1 reverse transcriptase inhibitors (NNRTIs).

Several lines of evidence now attest that lipoprotein[a] (Lp[a]) is a significant risk factor for many cardiovascular disorders. This enigmatic lipoprotein, composed of a single copy of apolipoprotein B (apoB) and apolipoprotein[a] (apo [a]), expresses peculiar metabolism, virtually independent from lifestyle interventions. Several therapeutic options have hence been proposed for lowering elevated Lp[a] values, with or without concomitant effect on low density lipoprotein (LDL) particles, mostly encompassing statins, ezetimibe, nicotinic acid, lipoprotein apheresis, and anti-PCSK9 monoclonal antibodies. Unusual redox play of Mo(V/IV) during oxidative aryl–aryl coupling

The oxidative treatment of a suitable 1,3-diarylpropene precursor by MoCl5 causes a series of redox steps yielding a dimer of dibenzo[a,c]cycloheptene. After the oxidative aryl–aryl bond formation, a C,H activation occurs providing a tropylium intermediate. Upon aqueous workup the metal waste acts as reductive media generating the dimer in an almost quantitative manner. The oxidative generation of the tropylium species as well as the subsequent redox play by the metal waste is unique and unprecedented. The dimeric compound can be oxidatively cleaved and subsequently decarboxylated providing the key intermediate of a previous synthesis of metasequirin-B derivatives.

A simple and expedient method for the synthesis of 3-methylene-isoindolin-1-ones 4 under aqueous phase-transfer conditions has been developed. Starting from 2-iodobenzamides 1 and (silyl) alkynes, the products are obtained in high yields and short reaction times (30 min) with the use of inexpensive CuCl/PPh3 catalyst system in the presence of n-Bu4NBr (TBAB) as a phase-transfer agent. Terminal alkynes are conveniently "unmasked" upon in situ desilylation under the reaction conditions. Alkynes possessing heterocyclic moieties were also found as amenable substrates. Targeting N-heterocyclic carbenes (NHCs) with increased π-acceptor character featuring N-fluorophenyl substituents, the molecular 2-chloro-1,3-bis(fluorophenyl)imidazolidine-4,5-diones (1a–c) were isolated from the condensation of the corresponding formamidine with oxalyl chloride. Microwave-assisted, highly enantioselective addition of diethylzinc to aromatic aldehydes catalyzed by chiral aminonaphthols

Abstract New optically active aminonaphthols were obtained by condensation of 2-naphthol, benzaldehyde and ( R )-(+)-1-(1-naphthyl)ethylamine or ( R )-(+)-1-(2-naphthyl)ethylamine. Their N-methylated derivatives were also synthesized. The new aminonaphthols 2 , 4 , 5 and 7 were found to catalyze the enantioselective ethylation of aryl aldehydes to 1-aryl-1-propanols (up to 92% ee). The reactions were accelerated greatly by microwave irradiation.

Aryl azides formation under mild conditions: a kinetic study in some ionic liquid solutions.

The kinetics of nucleophilic aromatic substitution of three nitrothiophene derivatives in different [1-butyl-3-methylimidazolium][N(3)]/ionic liquid binary mixtures was studied spectrophotometrically at 298 K. Ionic liquids differing for cation structure (imidazolium or pyrrolidinium) and for size, shape, and coordination ability of the anion ([BF(4)(-)], [PF(6)(-)], [SbF(6)(-)], and [NTf(2)(-)]) were used. Furthermore, in order to have a comparison with conventional organic solvents, the target reaction was also carried out in DMF solution at increasing concentration of NaN(3) or [bmim][N(3)].
