Abstract Ultraviolet (UV) filters are widely used in personal care products and due to their lipophilicity these chemicals tend to bioaccumulate in the aquatic biota. 4-Methylbenzylidene camphor (4-MBC) is one of the most used UV-filters, and it is commonly detected in freshwater fish tissues. This substance is suspected to be an endocrine disruptor due to its interaction with Hypothalamus-Pituitary-Gonadal (HPG) and HP-Thyroid (HPT)-axis. Diferenciācijas indukcijas izpēte pēc BRAF un mTOR inhibitoru apstrādes BRAF mutētās ādas melanomas šūnās

Darba mērķis bija padziļināti izpētīt BRAF inhibīcijas un sekojoši kombinētās BRAF un mTOR inhiibīcijas ietkemi uz metastātiskās BRAF mutēās melanomas šūnu līnijas SKMel28 šūnu ciklu, dzīvotspēju un inducēto atgriezensiko epitelilālo diferenciāciju. Ar advancētām mikroskopijas un šūnu analīzes metodēm tika apskatīti šos procesus iespējami ietkmējošie mehānismi. BRAF inhibīcija ar Vemurafenib rezultējās melanomas šūnu epiteliālā diferenciācijā un G1 arestā. THE HDAC INHIBITOR ITF2357 (GIVINOSTAT) AS A KEY PLAYER IN EPIGENETIC TARGETING OF MELANOMA AND COLON CANCER CELLS

Histone deacetylase inhibitors (HDACIs) are epigenetic compounds that have been recently considered for their promising anti-tumor activity. The aim of this PhD thesis was to elucidate and characterize the anti-tumor effect of the HDAC inhibitor ITF2357 (Givinostat) in melanoma and colon cancer cells that are characterized by oncogenic BRAF mutations. Interestingly, data reported in this thesis demonstrate that ITF2357 exerts a remarkable anti-tumor effect in melanoma cells by inducing a switch from a pro-survival autophagy to caspase-dependent apoptosis. Oncogenic BRAF and p53 Interplay in Melanoma Cells and the Effects of the HDAC Inhibitor ITF2357 (Givinostat)

Oncogenic BRAF mutations have been widely described in melanomas and promote tumour progression and chemoresistance. We previously provided evidence that the HDAC inhibitor ITF2357 (Givinostat) targets oncogenic BRAF in SK-MEL-28 and A375 melanoma cells. Here, we show that oncogenic BRAF localises to the nucleus of these cells, and the compound decreases BRAF levels in both the nuclear and cytosolic compartments. Visual Contrast Modulates Operant Learning Responses in Larval Zebrafish.

The larval zebrafish is a promising vertebrate model organism to study neural mechanisms underlying learning and memory due to its small brain and rich behavioral repertoire. Here, we report on a high-throughput operant conditioning system for zebrafish larvae, which can simultaneously train 12 fish to associate a visual conditioned pattern with electroshocks. We find that the learning responses can be enhanced by the visual contrast, not the spatial features of the conditioned patterns, highlighted by several behavioral metrics.
