Efficacy and safety of bempedoic acid for the treatment of hypercholesterolemia: A systematic review and meta-analysis

Background Bempedoic acid is a first-in-class lipid-lowering drug recommended by guidelines for the treatment of hypercholesterolemia. Our objective was to estimate its average effect on plasma lipids in humans and its safety profile. Methods and findings We carried out a systematic review and meta-analysis of phase II and III randomized controlled trials on bempedoic acid (PROSPERO: CRD42019129687). Apolipoprotein H is a protein of about 50 kilodaltons, structurally related to the regulators of the complement activation family. Its physiological function is poorly understood but it has been implicated in lipid metabolism and coagulative pathways. The major site of synthesis is thought to be the liver. Several reports indicate that apolipoprotein H is the antigen of the antiphospholipid antibodies and also behaves as an acute-phase reactant. Genetic polymorphisms affecting the phenotypic expression in familial hypercholesterolemia

The clinical expression of heterozygous familial hypercholesterolemia (FH) is highly variable even in patients carrying the same LDL receptor (LDL-R) gene mutation. This variability might be due to environmental factors as well as to modifying genes affecting lipoprotein metabolism. We investigated Apo E (2, 3, 4), MTP (-493G/T), Apo B (-516C/T), Apo A-V (-1131T/C), HL (-514C/T and -250G/A), FABP-2 (A54T), LPL (D9N, N291S, S447X) and ABCA1 (R219K) polymorphisms in 221 unrelated FH index cases and 349 FH relatives with defined LDL-R gene mutations. Abstract Background and Aims: Type III hyperlipoproteinemia, or dysbetalipoproteinemia, is commonly associated with apolipoprotein E2 homozygosity (Cy Background and Aims: 12, Cy Background and Aims: 58). Apo E2-Christchurch (Arg136→Ser), a rare mutation of the Apo E gene, located in the receptor-binding domain of the protein, has been found to be associated in the vast majority of cases of dysbetalipoproteinemia. Quasisymmetric spheres over Jordan domains

Let $\Omega$ be a planar Jordan domain. We consider double-dome-like surfaces $\Sigma$ defined by graphs of functions of $dist( \cdot ,\partial \Omega)$ over $\Omega$. The goal is to find the right conditions on the geometry of the base $\Omega$ and the growth of the height so that $\Sigma$ is a quasisphere, or quasisymmetric to $\mathbb{S}^2$. An internal uniform chord-arc condition on the constant distance sets to $\partial \Omega$, coupled with a mild growth condition on the height, gives a close-to-sharp answer. Our method also produces new examples of quasispheres in $\mathbb{R}^n$, for any $n\ge 3$.

Non-linear oscillators under parametric and external poisson pulses

The extended Ito calculus for non-normal excitations is applied in order to study the response behaviour of some non-linear oscillators subjected to Poisson pulses. The results obtained show that the non-normality of the input can strongly affect the response, so that, in general, it can not be neglected.

Contrasting probabilistic and anti-optimization approaches in an applied mechanics problem

Probabilistic and non-probabilistic, anti-optimization analyses of uncertainty are contrasted in this study. Specifically, the comparison of these two competing approaches is conducted for an uniform column, with initial geometric imperfection, subjected to an impact axial load. The reliability of the column is derived for the cases when the initial imperfections posses either (a) uniform probability density, (b) truncated exponential density or (c) generic truncated probability density.
