Pitfalls to be considered on the metabolomic analysis of biological samples by HR-MAS

2014

HR-MAS (High-Resolution Magic AngleSpinning) is considered a powerfultechnique for metabolomic studies ofbiological samples that provides “intact”tissue spectra (Cheng et al., 1998; Waterset al., 2000; Sitter et al., 2002; Martinez-Bisbal et al., 2004; Payne et al., 2006;Coen et al., 2007; Bathen et al., 2010).The performance of HR-MAS, fol-lowed by quantitative histopathology hasdemonstrated that, despite some changes,HR-MAS can preserve approximately thetissue histopathologic features producingwell-resolved spectra of cellular metabo-lites (Cheng et al., 2000). Nevertheless,therearesomeaspectsarousedintheliter-ature about the possible biochemical andstructural changes that can occur durin…

Regulation of microRNAs and their role in liver development, regeneration and disease

2013

Abstract Since their discovery more than a decade ago microRNAs have been demonstrated to have profound effects on almost every aspect of biology. Numerous studies in recent years have shown that microRNAs have important roles in development and in the etiology and progression of disease. This review is focused on microRNAs and the roles they play in liver development, regeneration and liver disease; particularly chronic liver diseases such as alcoholic liver disease, non-alcoholic fatty liver disease, non-alcoholic steatohepatitis, viral hepatitis and primary liver cancer. The key microRNAs identified in liver development and chronic liver disease will be discussed together with, where pos…

Association between the PNPLA3 (rs738409 C>G) variant and hepatocellular carcinoma: Evidence from a meta-analysis of individual participant data

2014

The incidence of hepatocellular carcinoma (HCC) is increasing in Western countries. Although several clinical factors have been identified, many individuals never develop HCC, suggesting a genetic susceptibility. However, to date, only a few single-nucleotide polymorphisms have been reproducibly shown to be linked to HCC onset. A variant (rs738409 C>G, encoding for p.I148M) in the PNPLA3 gene is associated with liver damage in chronic liver diseases. Interestingly, several studies have reported that the minor rs738409[G] allele is more represented in HCC cases in chronic hepatitis C (CHC) and alcoholic liver disease (ALD). However, a significant association with HCC related to CHC has not b…

Personalized medicine for allergy treatment: Allergen immunotherapy still a unique and unmatched model

2021

International audience; The introduction of personalized medicine (PM) has been a milestone in the history of medical therapy, because it has revolutionized the previous approach of treating the disease with that of treating the patient. It is known today that diseases can occur in different genetic variants, making specific treatments of proven efficacy necessary for a given endotype. Allergic diseases are particularly suitable for PM, because they meet the therapeutic success requirements, including a known molecular mechanism of the disease, a diagnostic tool for such disease, and a treatment blocking the mechanism. The stakes of PM in allergic patients are molecular diagnostics, to dete…

Understanding the complexity of IgE-related phenotypes from childhood to young adulthood: A Mechanisms of the Development of Allergy (MeDALL) seminar.

2012

Mechanisms of the Development of Allergy (MeDALL), a Seventh Framework Program European Union project, aims to generate novel knowledge on the mechanisms of initiation of allergy. Precise phenotypes of IgE-mediated allergic diseases will be defined in MeDALL. As part of MeDALL, a scientific seminar was held on January 24, 2011, to review current knowledge on the IgE-related phenotypes and to explore how a multidisciplinary effort could result in a new integrative translational approach. This article provides a summary of the meeting. It develops challenges in IgE-related phenotypes and new clinical and epidemiologic approaches to the investigation of allergic phenotypes, including cluster a…

Oxidative stress in Alzheimer’s Disease: Implications for Prevention and Therapy

2006

Oxidative stress is a marker of neurodegeneration and has been recently shown to be also involved in the early stages of the pathogenesis of various neurodegenerative disorders. In general, all biomolecules of the cell can be oxidized and thereby damaged. Consequently, the concept of neuroprotection by antioxidants has been developed. In many cases the direct scavanging of free radicals have been used as a strategy to prevent oxidative stress damage and a variety of physiological and synthetic antioxidant molecules have been identified and synthesized including the female sex homone estrogen. In Alzheimer’s Disease amyloid-β protein on its way to brain deposition can also induce oxidative c…

Novel Insights and Therapeutical Applications in the Field of Inhibitors of COX-2

2004

The discovery of the two isoenzymes COX-1 and COX-2 and the knowledge of their function, localisation and regulation has initiated the development of COX-2 selective inhibitors (coxibs). Inducible COX-2 at the peripheral site of inflammation has been detected in the early 1990s, the involvement of recently detected spinal COX-2 has led to new insights into mechanisms of pain and may explain analgesic and antipyretic properties of COX-2 selective inhibitors. The coxibs rofecoxib and celecoxib have been introduced into therapy and seem to offer some advantages over the classical non-selective NSAIDs. The search for new COX-2 inhibitors is going on, the development of etoricoxib and lumiracox…

Extraction and Sample Preparation

2015

As everyone knows, the role played by extraction and sample preparation in the analytical sciences cannot be overemphasized. Despite tremendous advances in chromatography, detection, and other aspects of analysis, extraction and sample preparation remain a preanalysis ritual of critical importance. It has been estimated that around 50% to 70% possibly even more of the time and effort that goes into an analytical process comprises extraction and sample preparation. Sample preparation procedure can vary in the degree of selectivity, speed, and convenience, depending on the approach and conditions used, as well as on the geometric configurations of the extraction phase and conditions. Proper d…

Ancestral Reconstruction and Investigations of Genomic Recombination on some Pentapetalae Chloroplasts

2019

Abstract In this article, we propose a semi-automated method to rebuild genome ancestors of chloroplasts by taking into account gene duplication. Two methods have been used in order to achieve this work: a naked eye investigation using homemade scripts, whose results are considered as a basis of knowledge, and a dynamic programming based approach similar to Needleman-Wunsch. The latter fundamentally uses the Gestalt pattern matching method of sequence matcher to evaluate the occurrences probability of each gene in the last common ancestor of two given genomes. The two approaches have been applied on chloroplastic genomes from Apiales, Asterales, and Fabids orders, the latter belonging to Pe…
