Human artificial membranes in (bio)analytical science: Potential for in vitro prediction of intestinal absorption-A review

2021

Abstract Artificial membranes for investigation of the human absorption (oral, dermal or respiratory) of target organic compounds are aimed at mimicking the interactions occurring at and within the cell lipid membrane. Biomolecules such as proteins are also integral components of the lipid membranes and play a pivotal role towards understanding the complex mechanisms of human absorption. In this review, we will differentiate biomimetic platforms based on static (batchwise) and dynamic modes. In the former, a synthetic membrane placed between two phases (donor and acceptor) mimics a given biological system to study permeability. Parallel artificial membrane permeation assays are the most com…

PAMPA—a drug absorption in vitro model

2004

Parallel artificial membrane permeability assay (PAMPA) was used to measure the effective permeability, P(e), as a function of pH from 4 to 10, of 17 fluoroquinolones, including three congeneric series with systematically varied alkyl chain length at the 4'N-position of the piperazine residue. The permeability values spanned over three orders of magnitude. The intrinsic permeability, P(o), and the membrane permeability, P(m), were determined from the pH dependence of the effective permeability. The pK(a) values were determined potentiometrically. The PAMPA method employed stirring, adjusted such that the unstirred water layer (UWL) thickness matched the 30-100 microm range estimated to be i…

On the stability of the 24-meric hemocyanin from Eurypelma californicum.

1998

The stability of the 24-meric hemocyanin from Eurypelma californicum towards various denaturants (GdnHCl, urea, urea derivatives and salts of the Hofmeister series) indicates that the quaternary structure is stabilized by hydrophilic and polar forces. Thus, the interaction between the seven different subunit types of this cheliceratan hemocyanin is comparable with that of the closely related crustacean hemocyanins. In contrast, no significant influence of divalent ions such as Ca2+ and Mg2+ on the stability is observed at pH 8.0 and pH 8.5 but not at pH 7.0. Studies, both in the presence of urea and GdnHCl indicate that the denaturation process consists of a dissociation of the oligomeric s…

Different approaches in sulfonated poly (ether ether ketone) conductivity measurements

2019

Ion conductivity of sulfonated poly (ether ether ketone) (SPEEK) membranes with various degree of sulfonation (DS) was investigated using impedance analysis with different measuring cell configuration and ion conductivity was calculated from resistances of polymer membranes. SPEEK was synthesized from poly (ether ether ketone) (PEEK) via sulfonation reaction in concentrated sulfuric acid (95-98%). Scanning electron microscopy (SEM) analysis of membrane surface was performed to determine possible mechanical damage to the membrane during resistance measurements.

Biological evaluation of PLLA membranes, with different pore diameters, to stimulate cell adhesion and growth in vitro

2015

Polymeric membranes prepared via DIPS (Diffusion Induced Phase Separation) are widely studied and utilized as scaffolds for the regeneration of tissue. In this work, poly (L)-lactide membrane are prepared through a DIPS protocol starting from a ternary solution made of polymer, dioxane (solvent) and water (non-solvent). A three-dimensional, porous and mechanically stable membrane is desirable for ingrowth of human bronchial epithelial cells. CeO2−x nanorods with intrinsic urease-like activity

2018

The large-scale production and ecotoxicity of urea make its removal from wastewater a health and environmental challenge. Whereas the industrial removal of urea relies on hydrolysis at elevated temperatures and high pressure, nature solves the urea disposal problem with the enzyme urease under ambient conditions. We show that CeO2−x nanorods (NRs) act as the first and efficient green urease mimic that catalyzes the hydrolysis of urea under ambient conditions with an activity (kcat = 9.58 × 101 s−1) about one order of magnitude lower than that of the native jack bean urease. The surface properties of CeO2−x NRs were probed by varying the Ce4+/Ce3+ ratio through La doping. Although La substit…

Mechanical Properties of Composite SPEEK Polymer Membranes Modified with Ionic Liquids

2015

In this work, the mechanical properties of sulphonated polyetheretherketone (SPEEK) membranes impregnated with 3 different ionic liquids (1-butyl-2,3-dimethyl- imidazolium dimethylphosphate ([BMMIM][Me2PO4])), 1,2,3-trimethylimidazolium dimethylphosphate ([MMMIM][Me2PO4])), 1,3-dimethylimidazolium dimethylphosphate ([MMIM][Me2PO4])) have been investigated. Prepared SPEEK/ionic liquid composite membranes are characterized by mechanical testing both in room and elevated temperatures. It was found that the stiffness and tensile strength of composites decreased by increasing the content of ionic liquid and the length of alkyl radical in ionic liquid as well as by increasing the temperature.

Spin crossover in mononuclear and binuclear iron(III) complexes with pentadentate Schiff-base ligands

2000

Abstract A series of mononuclear hexacoordinate iron(III) complexes, [Fe( 5 L )(py)]BPh 4 , and binuclear hexacoordinate iron(III) complexes, [( 5 L )Fe(μ 2 -bpy)Fe( 5 L )](BPh 4 ) 2 , has been prepared and their magnetic properties were investigated; the pentadentate ligands were derivatives of 5 L =saldptn=N,N′-bis(2-hydroxybenzyliden)-1,7-diamino-4-azaheptane. Temperature variation of the effective magnetic moment for them shows that a spin transition from the low-spin to the high-spin state occurs. The magnetic data were fitted to an Ising-like model appropriate for the mono- and binuclear systems.
