A NEW PCR-BASED TYPING OF THE RODGERS AND CHIDO ANTIGENIC DETERMINANTS OF THE FOURTH COMPONENT OF HUMAN COMPLEMEMT
1994
The Rodgers (Rg) and Chido (Ch) blood groups are antigenic determinants of the fourth component of human complement C4. They are associated with the two isotypes of C4, C4A and C4B, respectively. They serve as markers to distinguish C4A from C4B as well as for the definition of subtypes of common and rare allotypes. As an alternative to the serological typing method using human alloantisera, a PCR typing procedure with sequence-specific primers (PCR-SSP) was designed. The method was tested on selected DNA samples from individuals with well-defined C4 allotypes. No false-positive or false-negative typing results were obtained and all the determinant combinations could be distinguished. The P…
Characterization of the NAD-Dependent Human Histone Deacetylases Gene Sirtuin 1 and Its Implications on Aging and the Development of Malignant Diseas…
2006
Abstract A dysregulation of the tightly controlled equilibrium of acetylation and deacetylation plays a causative role in the generation as well as in the suppression of cancer. Histone acetylation modifiers are therefore gaining increasing attention as potential targets in the treatment of cancer. Sirtuin 1 (SIRT1) is a nicotinamide adenine dinucleotide (NAD+)-dependent deacetylase, which belongs to the silent information regulator 2 (Sir2) family of sirtuin histone deacetylases (HDACs). The yeast Sir2 protein and its mammalian derivatives play a central role in epigenetic gene silencing, DNA repair and recombination, cell-cycle, microtubule organization, and in the regulation of aging. We…
GENETIC POLYMORPHISM OF THE FOURTH COMPONENT OF HUMAN COMPLEMENT: POPULATION STUDY AND PROPOSAL FOR A REVISED NOMENCLATURE BASED ON GENOMIC PCR TYPIN…
1996
SUMMARY The fourth component of human complement (C4) is coded for by two homologous genes, C4A and C4B, located in the class III region of the major histocompatibility complex (MHC). Genetic typing of C4A and B alleles is routinely carried out by high-voltage agarose gel electrophoresis. The electrophoretic C4 polymorphism can be further subdivided by the Rodgers (Rg) and Chido (Ch) blood groups, which are antigenic determinants of the C4A and B alpha-chains, respectively. We have used a recently described direct PCR typing method using sequence-specific primers (PCR-SSP) in combination with electrophoretic C4 typing as well as genomic RFLP analysis to determine the frequency of C4 allotyp…
Direct evidence for positive selection of skin, hair, and eye pigmentation in Europeans during the last 5,000 y
2014
Significance Eye, hair, and skin pigmentation are highly variable in humans, particularly in western Eurasian populations. This diversity may be explained by population history, the relaxation of selection pressures, or positive selection. To investigate whether positive natural selection is responsible for depigmentation within Europe, we estimated the strength of selection acting on three genes known to have significant effects on human pigmentation. In a direct approach, these estimates were made using ancient DNA from prehistoric Europeans and computer simulations. This allowed us to determine selection coefficients for a precisely bounded period in the deep past. Our results indicate t…
Hypoxia and Human Genome Stability: Downregulation of BRCA2 Expression in Breast Cancer Cell Lines
2013
Previously, it has been reported that hypoxia causes increased mutagenesis and alteration in DNA repair mechanisms. In 2005, an interesting study showed that hypoxia-induced decreases in BRCA1 expression and the consequent suppression of homologous recombination may lead to genetic instability. However, nothing is yet known about the involvement of BRCA2 in hypoxic conditions in breast cancer. Initially, a cell proliferation assay allowed us to hypothesize that hypoxia could negatively regulate the breast cancer cell growth in short term in vitro studies. Subsequently, we analyzed gene expression in breast cancer cell lines exposed to hypoxic condition by microarray analysis. Interestingly,…
Posttraumatic stress disorder among earthquake survivors of the Wenchuan area (Sichuan, China)
2014
Background : On May 12, 2008, an earthquake with a power of 8.0 M on the Richter scale occurred in the Wenchuan County of Sichuan Province in southwest China, which was unprecedented in magnitude and aftermath. Approximately 70,000 people were killed and nearly 20,000 went missing. The earthquake caused a wide number of mental and physical health outcomes among survivors, and posttraumatic stress disorder (PTSD) was one of the most commonly studied. Methods : We conducted a systematic overview to assess research achievements about PTSD in the past 6 years after the Wenchuan earthquake, including symptoms and risk factors about PTSD among Wenchuan earthquake survivors, as well as research de…
H$^{-}$ extraction systems for CERN’s Linac4 H$^{-}$ ion source
2018
Linac4 is a 160 MeV linear H accelerator at CERN. It is an essential part of the beam luminosity upgrade of the Large Hadron Collider (LHC) and will be the primary injector into the chain of circular accelerators. It aims at increasing the beam brightness by a factor of 2, when compared to the currently used 50 MeV linear proton accelerator, Linac2. Linac4’s ion source is a cesiated RF-plasma H ion source. Several beam extraction systems were designed for H beams of 45 keV energy, 50 mA intensity and an electron to H ratio smaller than 5. The goal was to extract a beam with an rms-emittance of mm mrad. One of the main challenges in designing an H extraction system is dumping of the co-extra…
Rational normal curves and Hadamard products
2021
AbstractGiven $$r>n$$ r > n general hyperplanes in $$\mathbb P^n,$$ P n , a star configuration of points is the set of all the n-wise intersection of the hyperplanes. We introduce contact star configurations, which are star configurations where all the hyperplanes are osculating to the same rational normal curve. In this paper, we find a relation between this construction and Hadamard products of linear varieties. Moreover, we study the union of contact star configurations on a same conic in $$\mathbb P^2$$ P 2 , we prove that the union of two contact star configurations has a special h-vector and, in some cases, this is a complete intersection.
Autistic Adult Health and Professional Perceptions of It: Evidence From the ASDEU Project
2021
The Autism Spectrum Disorders in the European Union (ASDEU) survey investigated the knowledge and health service experiences of users and providers to generate new hypotheses and scientific investigations that would contribute to improvement in health care for autistic adults. An online survey designed for autistic adults, carers of autistic adults, and professionals in adult services was translated into 11 languages and distributed electronically by organizations and in-country adult service facilities in 2017; 522 autistic adults, 442 carers, and 113 professionals provided answers to the health questions. Professionals, the majority in non-medical services, appeared to be poorly informed …
Negative and positive feedback from a supernova remnant with SHREC
2022
Supernova remnants (SNRs) contribute to regulate the star formation efficiency and evolution of galaxies. As they expand into the interstellar medium (ISM), they transfer vast amounts of energy and momentum that displace, compress and heat the surrounding material. Despite the extensive work in galaxy evolution models, it remains to be observationally validated to what extent the molecular ISM is affected by the interaction with SNRs. We use the first results of the ESO-ARO Public Spectroscopic Survey SHREC, to investigate the shock interaction between the SNR IC443 and the nearby molecular clump G. We use high sensitivity SiO(2-1) and H$^{13}$CO$^+$(1-0) maps obtained by SHREC together wit…
