In the wake of the Mesozoic, the Early Triassic (~251.95 Ma) corresponds to the aftermath of the most severe mass extinction of the Phanerozoic: the end-Permian crisis, when life was nearly obliterated (e.g., 90% of marine species disappeared). Consequences of this mass extinction are thought to have prevailed for several millions of years, implying a delayed recovery lasting the whole Early Triassic, if not more. Several paradigms have been established and associated to a delayed biotic recovery scenario expected to have resulted from harsh and deleterious paleoenvironments. The substances analyzed mainly at the “Chemical Investigation Laboratory” of the Regional Cabinet of Palermo, in the period from 2013 to 2018, are heroin and cocaine. For analytical techniques, mass spectrometry techniques associated with gas chromatography were used. The existence of a close relation between personality and drug consumption is recognized, but the corresponding causal connection is not well known. Neither is it well known whether personality exercises an influence predominantly at the beginning and development of addiction, nor whether drug consumption produces changes in personality. This paper presents a dynamic mathematical model of personality and addiction based on the unique personality trait theory (UPTT) and the general modelling methodology. A series of 6alpha- and 6beta-substituted benztropines were synthesized. A marked enantioselectivity was observed for the 6beta-methoxylated benztropines, the (1R)-isomers being more potent than the corresponding (1S) compounds. The racemic 6alpha-methoxy-3-(4',4' '-difluorodiphenylmethoxy)tropane (5 g) was the most potent compound. It has been found that modifications at the 6-position of benztropine might reduce the DAT binding affinity, maintaining otherwise a significant dopamine uptake inhibitory activity. A reinvestigation of the absolute configuration of 6beta-methoxytropinone proved the 6R configuration for the (+)-enantiomer.

Abstract Converging evidence indicates that repeated exposure to motor stimulants such as cocaine and amphetamine produces marked alterations in network responsiveness of striatal neurons to subsequent challenge with the same stimulant drug. Such alterations, which correlate with persistent patterns of repetitive behavior, associate with distinct compartmental changes in the neostriatum. Striatal matrix system neurons undergo “silencing” following repeated drug challenges, allowing striosome system neurons to exhibit preferential activation.
