Lovastatin protects human endothelial cells from the genotoxic and cytotoxic effects of the anticancer drugs doxorubicin and etoposide

2006

Background and purpose: 3-Hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhibitors (statins) are frequently used lipid-lowering drugs. Moreover, they exert pleiotropic effects on cellular stress responses and death. Here, we analysed whether lovastatin affects the sensitivity of primary human endothelial cells (HUVEC) to the anticancer drug doxorubicin. Experimental approach: We investigated whether pretreatment of HUVEC with low dose of lovastatin influences the cellular sensitivity to doxorubicin. To this end, cell viability, proliferation and apoptosis as well as DNA damage-triggered stress response were analysed. Key results: Lovastatin reduced the cytotoxic potency of doxorub…

Processing of O6-methylguanine into DNA double-strand breaks requires two rounds of replication whereas apoptosis is also induced in subsequent cell …

2009

The DNA adduct O(6)-methylguanine (O(6)MeG) induced by environmental genotoxins and anticancer drugs is a highly mutagenic, genotoxic and apoptotic lesion. Apoptosis induced by O(6)MeG requires mismatch repair (MMR) and proliferation. Models of O(6)MeG-triggered cell death postulate that O(6)MeG/T mispairs activate MMR giving rise to either direct genotoxic signaling or secondary lesions that trigger apoptotic signaling in the 2(nd) replication cycle. To test these hypotheses, we used a highly synchronized cell system competent and deficient for the repair of O(6)MeG adducts, which were induced by the S(N)1 methylating agent N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). We show that DNA doub…

FRIPON: a worldwide network to track incoming meteoroids

2020

Context. Until recently, camera networks designed for monitoring fireballs worldwide were not fully automated, implying that in case of a meteorite fall, the recovery campaign was rarely immediate. This was an important limiting factor as the most fragile - hence precious - meteorites must be recovered rapidly to avoid their alteration. Aims. The Fireball Recovery and InterPlanetary Observation Network (FRIPON) scientific project was designed to overcome this limitation. This network comprises a fully automated camera and radio network deployed over a significant fraction of western Europe and a small fraction of Canada. As of today, it consists of 150 cameras and 25 European radio receiver…

Darba vides riska faktori aptiekā strādājošajiem farmaceitiem un preventīvie pasākumi

2016

Industrial Environment Mapping Using Distributed Static 3D Sensor Nodes

2018

This paper presents a system architecture for mapping and real-time monitoring of a relatively large industrial robotic environment of size 10 m × 15 m × 5 m. Six sensor nodes with embedded computing power and local processing of the 3D point clouds are placed close to the ceiling. The system architecture and data processing is based on the Robot Operating System (ROS) and the Point Cloud Library (PCL). The 3D sensors used are the Microsoft Kinect for Xbox One and point cloud data is collected at 20 Hz. A new manual calibration procedure is developed using reflective planes. The specified range of the used sensor is 0.8 m to 4.2 m, while depth data up to 9 m is used in this paper. Despite t…

FABC: Retinal Vessel Segmentation Using AdaBoost

2010

This paper presents a method for automated vessel segmentation in retinal images. For each pixel in the field of view of the image, a 41-D feature vector is constructed, encoding information on the local intensity structure, spatial properties, and geometry at multiple scales. An AdaBoost classifier is trained on 789 914 gold standard examples of vessel and nonvessel pixels, then used for classifying previously unseen images. The algorithm was tested on the public digital retinal images for vessel extraction (DRIVE) set, frequently used in the literature and consisting of 40 manually labeled images with gold standard. Results were compared experimentally with those of eight algorithms as we…

Role of ambient light in the detection of contrast elements in digital dental radiography.

2018

Objective This study aimed to evaluate the effect of different ambient light levels on observer detection of small contrast differences in a contrast phantom by using a high-end liquid crystal display (LCD) monitor. Study Design An aluminum step wedge was converted into a contrast phantom by the addition of bore holes. Radiographic images of the contrast elements were presented to 20 observers. Images were displayed in random order under different ambient light levels (0, 50, 200, and 500 lux) twice, and the observers had to determine if contrast elements were visible. Sensitivity and specificity were determined to calculate areas under receiver operating characteristic curves and Friedmann…

Imaging of nitric oxide in a living vertebrate using a diaminofluorescein probe

2007

Abstract Numerous approaches have been described to identify nitric oxide (NO), a free radical involved in various physiological and pathophysiological processes. One of these approaches is based on the use of chemical probes whose transformation by NO generates highly fluorescent derivatives, permitting detection of NO down to nanomolar concentrations. Here, we show that the cell-permeant diaminofluorophore 4-amino-5-methylamino-2′-7′-difluorofluorescein diacetate (DAF-FM-DA) can be used to detect NO production sites in a living vertebrate, the zebrafish Danio rerio. The staining pattern obtained in larvae includes the bulbus arteriosus, forming bones, the notochord, and the caudal fin. Th…
