Purification and characterization of an intracellular family 3 β-glucosidase from Lactobacillus sanfranciscensis CB1

Lactobacillus sanfranciscensis CB1 uses cellobiose and other β-glucosides (methyl-β-glucoside, arbutin, amygdalin and salicin) as carbon sources. A hexameric ca. 288 kDa β-glucosidase was purified to homogeneity from Lact. sanfranciscensis CB1 by four chromatographic steps. The enzyme was optimally active at pH 7.5 and 40°C. It had a pI of ca. 4.38 and a D55°C value of ca. 27 sec. Almost total inhibition was found with sulfhydryl-modifying agents and divalent cations such as Cu2+, Co2+, Hg2+, Ni2+ and Fe2+ at a concentration of 2 mM. The morphological aspects of Styela plicata fertilization after treatment with tributyltin(IV) chloride are described by means of scanning and transmission electron microscopy investigations. Alterations have been shown both on female and male gametes; spermatozoa, all the egg envelopes and the mitochondria of the egg cortical cytoplasm are modified in relation to incubation time. As a consequence, the damage to gametes blocks sperm–egg interaction and fertilization does not occur. Copyright © 2003 John Wiley & Sons, Ltd.

A Cystoseira brachycarpa community from a vent area off Panarea Island (Italy) was investigated in two sites at different pH values. At low pH, species richness and coverage were low and the community displayed a reduced reproductive capacity. Conversely, at normal pH, dense canopies of fertile C. brachycarpa were found.

Onset of DNA synthesis in experimentally activated ascidian eggs

DNA synthesis was studied autoradiographically in unfertilized ascidian eggs (Ascidia malaca and Ciona intestinalis) that had been artificially activated by a K+-free external medium or a Ca-ionophore. Naked eggs of A. malaca were incubated in K+-free seawater that contained [3H]-thymidine for 30 min, and naked eggs of C. intestinalis were incubated in seawater supplemented with a Ca-ionophore and [3H]-thymidine for 15 min. One suggested anti-predator function of alarm calls is to deliver a message to a predator that it has been detected. Moreover, giving the alarm call could provide a signal to the predator that capturing the individual giving the alarm is more difficult than capturing its silent group members, as the caller is probably the most aware of the predator's location. In an aviary experiment using stuffed dummy Willow Tits Poecile montanus, we assessed whether an authentic alarm call given by Willow Tit affected Pygmy Owl Glaucidium passerinum prey preference. The morphological aspect of the follicle cells of Styela plicata eggs is described by means of scanning electron microscope investigations. The follicular layer is made of spaced, cylindrical box-like cells which are arranged hexagonally. They adhere to the egg through a complex network of membrane extensions making an overall thin layer on the vitelline coat. The walls of the follicle cells are plentifully provided with microvilli, filopodia and lamellipodia, which allow a connection among the cells. A polyphasic approach in order to identify dominant lactic acid bacteria during pasta manufacturing

Using a polyphasic approach, we have isolated and identified, lactic acid bacteria (LAB) in samples directly collected from an artisanal pasta-making manufactory located in Puglia (South Italy). Samples were collected during several steps of pasta processing and LAB were isolated on MRS and M17 plates. Furthermore, strains were grouped in a total of eight species by means of randomly amplified polymorphic DNA (RAPD)-polymerase chain reaction (PCR) typing and 16S rDNA sequencing. Effect of season and soil treatments on carbohydrate concentrations in Norway spruce (Picea abies) mycorrhizae

We studied effects of season and soil treatments (watering, acidification, liming and combinations of these treatments) on soluble carbohydrates of mycorrhizal roots of Norway spruce (Picea abies (L.) Karst.). Arabinose, arabitol, fructose, glucose, inositol, lactose, mannitol, pinite, quinate, raffinose, shikimate, stachyose and trehalose were identified by HPLC. Concentrations of inositol, lactose and pinite were constant throughout the year, whereas concentrations of raffinose, stachyose and trehalose were higher in winter than in summer, and concentrations of glucose, fructose and mannitol increased from February to September.
