The endophytic microbiota of Citrus limon is transmitted from seed to shoot highlighting differences of bacterial and fungal community structures

2021

AbstractCitrus limon (L.) Burm. F. is an important evergreen fruit crop whose rhizosphere and phyllosphere microbiota  have been characterized, while seed microbiota is still unknown. Bacterial and fungal endophytes were isolated from C. limon surface-sterilized seeds. The isolated fungi—belonging to Aspergillus, Quambalaria and Bjerkandera genera—and bacteria—belonging to Staphylococcus genus—were characterized for indoleacetic acid production and phosphate solubilization. Next Generation Sequencing based approaches were then used to characterize the endophytic bacterial and fungal microbiota structures of surface-sterilized C. limon seeds and of shoots obtained under aseptic conditions fr…

The seed endosphere of Anadenanthera colubrina is inhabited by a complex microbiota, including Methylobacterium spp. and Staphylococcus spp. with pot…

2018

Background and aims: Plant seeds are emerging micro–habitats, whose importance as reservoir and vector of beneficial microbes just begins to be recognized. Here we aimed to characterize the bacterial microbiota of the Anadenanthera colubrina seed endosphere, with special focus to beneficial traits and to the colonization pattern. Methods: Cultivation–dependent (isolation from surface–sterilized seeds) and cultivation–independent (pyrosequencing of 16S rRNA gene from metagenomic seed DNA) analyses, functional tests and microscopical investigations (fluorescence in situ hybridization coupled with confocal laser scanning microscopy (FISH-CLSM) were performed. Results: We isolated several Methy…

Endophytes from medicinal plants’ seeds: exploring new reservoirs of bioactive molecules

Plant-associated microorganisms have recently gained more attention for their influence on plant health and biotechnological relevance. Bioactive molecules have been already isolated from plant-associated bacteria. Seed-borne bacterial endophytes have not been much explored though. Such endophytes are particularly important, since they can influence germination and be transmitted from generation to generation. Interestingly, seed endophytes from medicinal plants could influence the production of molecules with therapeutic properties. Bacterial endophytic strains were extracted from surface-sterilized seeds of the medicinal plant Echinacea purpurea and their 16S rRNA genes were sequenced. Th…

A method for measuring mitochondrial mass and activity

2007

Mitochondria, responsible for the energy-generating process essential for the cell metabolism, differ for the number, localization and activity in animal cells and tissues in relation to the energetic needs. Using fluorescent probes specific for mitochondria, Mitotracker Green (MTG) and Orange (MTO), and Confocal Laser-Scanning Microscope (CLSM), we elaborated a method to measure in vivo the mitochondrial mass and activity, in sea urchin Paracentrotus lividus eggs and embryos. The analysis of captured images, revealed a variation of mitochondrial distribution and an increase of activity after fertilization.

Confocal microscopy study of the distribution, content and activity of mitochondria during Paracentrotus lividus development

2007

Summary In the present paper we applied confocal microscopy andfluorescence technologies for studying the distribution andthe oxidative activity of sea urchin ( Paracentrotus lividus )mitochondria during development, by in vivo incubating eggsand embryos with cell-permeant MitoTracker probes. Wecalculated, by a mathematical model, the intensity values, the variations of intensity, and the variation index of incorporatedfluorochromes. Data demonstrate that mitochondrial massdoes not change during development, whereas mitochondrialrespirationincreases.Inaddition,startingfrom16blastomeresstage, some regions of the embryo contain organelles moreactive in oxygen consumption.

The seed endophytic microbiota of Citrus limon L. Burm. F

2017

Background Plant seeds possess a complex microbiota which may play a crucial rolein a plethora of mechanisms, such as preservation, germination, seedling development, plant growth and health. In particular, seed endophytic microbiota is gaining more and more consideration due to the fact that it may be vertically transmitted to ensure individual competitive advantages. The Citrusspecies, including C.limon, areone of the most economically important evergreen fruit crops in the world. Objectives i)Identification and characterization ofmicrobial isolates from Citrus limon L. Burm. F. seed endosphere. ii) Structure of Citrus limon L. Burm. F. seed endosphytic microbiota.

Confocal laser scanning microscopy for the study of the morphological changes of the postextraction sites

2012

A better understanding of the remodeling process of postextraction sockets is essential in dental treatment planning. The aim of this study was to evaluate whether confocal laser scanning microscopy (CLSM) can be applied to imaging contour changes of postextraction sites, as well as to its quantification with image analysis of obtained three-dimensional images. This work describes a new application of the CLSM technique. The system used was the OLS3100-USS, LEXT model (Olympus((R))). CLSM was used for the surface analysis of the extraction site. The measurements taken with CLSM were: (1) mesio-distal distance, (2) alveolar ridge thickness, and (3) vestibular and lingual alveolar ridge heigh…
