Immune evasion proteins gpUS2 and gpUS11 of human cytomegalovirus incompletely protect infected cells from CD8 T cell recognition

AbstractHuman cytomegalovirus (HCMV) encodes four glycoproteins, termed gpUS2, gpUS3, gpUS6 and gpUS11 that interfere with MHC class I biosynthesis and antigen presentation. Despite gpUS2–11 expression, however, HCMV infection is efficiently controlled by cytolytic CD8 T lymphocytes (CTL). To address the role of gpUS2 and gpUS11 in antigen presentation during viral infection, HCMV mutants were generated that expressed either gpUS2 or gpUS11 alone without coexpression of the three other proteins. Fibroblasts infected with these viruses showed reduced HLA-A2 and HLA-B7 surface expression. Human traffickers frequently take advantage of operative protocols that make them unpunishable pursuant the Italian Criminal Law because according to Art. 6 of the Italian Penal Code and International Convention on the High Sea jurisdictional powers cannot be exercised when the conduct is committed on the High Sea. So illustrating the territorial limitations of the efficacy of Criminal Law the research focuses on the hermeneutical solution given by the Court of Cassation that apparently with the intent of pursuing a more effectivity of the Criminal response recurred to the controversial category of the indirect offender in order to sanction smuggling conducts that enjoy the rescue intervention of the Italian authorities to reach their criminal aims. In turn in order to better identify the Italian jurisdiction in the crime of Art. 12 T.U. imm. I propose to correctly apply the concorso di persone (art. 110 c.p.) and the reato eventualmente permanente. MIXED lymphocyte culture (MLC)1 has been used extensively as an in vitro model to analyse the reactivity of T cells to antigens coded by the major histocompatibility complex (MHC). When murine T responder cells are exposed in vitro to allogeneic lymphoid cells (stimulator cells) they proliferate and cytotoxic T lymphocytes (CTL) are generated2,3. Antigens coded by the central I region of the MHC are chiefly responsible for triggering proliferation4,5, whereas the target antigen of the CTL generated is either a H–2K or H–2D region or a I–A subregion gene product5–8. We have previously shown that human IL-10-treated dendritic cells (DC) induce an antigen-specific anergy in CD4+ T lymphocytes. These anergic T cells are characterized by an inhibited proliferation, a reduced production of IL-2, and additionally display antigen-specific suppressor activity. In this study we investigated the mechanisms underlying the anergic state and regulatory function of these T cells. We did not observe enhanced rates of programmed cell death of anergic CD4+ suppressor T cells compared to T cells stimulated with mature DC. A subpopulation of peripheral human CD4(+)CD25(+) T cells that expresses CD45RO, histocompatibility leukocyte antigen DR, and intracellular cytotoxic T lymphocyte-associated antigen (CTLA) 4 does not expand after stimulation and markedly suppresses the expansion of conventional T cells in a contact-dependent manner. After activation, CD4(+)CD25(+) T cells express CTLA-4 on the surface detectable for several weeks. The CD8-positive CTL line MZ1257-CTL-5 (CTL-5) has been shown to lyse autologous cultured RCC cells in an HLA-A2 restricted fashion.
