Characteristics of haloethylene-induced acetonemia in rats.

A series of halogenated ethylenes (vinyl chloride, vinylidene fluoride,cis- andtrans-1,2-dichloroethylene, perchloroethylene) induces increased acetone exhalation in rats. Exposures of differently pre-treated rats to vinylidene fluoride suggest that a metabolite of the haloethylene must be envolved in eliciting this formation of acetone. This conclusion is based on (a) dependence of acetone exhalation on the concentration of vinylidene fluoride, (b) effect of inducing agents, (c) effect of pyrazol, a metabolic inhibitor, (d) effect of cysteine, (e) effect of hypoxia and (f) the time course of acetone exhalation.

Summary Lactate dehydrogenase from potato tubers was incubated together with various thiol reagents and the effect on the electrophoretic behavior of its isoenzymes studied. while disulfides, alkylating agents, and H202 did not alter the number and electrophoretic behavior of the isoenzymes of L + LDH, reduced glutathione and 2-mercaptoethanol led to an increase in the number of isoenzymes. The reaction was dependent on concentration and pH. On the other hand, cysteine and cysteamine had no such effect. Results are discussed in view of the possible secondary nature of the plant LDH isoenzymes.

AbstractRedox control of proteins that form disulfide bonds upon oxidative challenge is an emerging topic in the physiological and pathophysiological regulation of protein function. We have investigated the role of the neuronal calcium sensor protein visinin-like protein 1 (VILIP-1) as a novel redox sensor in a cellular system. We have found oxidative stress to trigger dimerization of VILIP-1 within a cellular environment and identified thioredoxin reductase as responsible for facilitating the remonomerization of the dimeric protein. N-Heterocyclic carbene gold(I) complexes bearing a fluorescent coumarin ligand were synthesized and characterized by various techniques. The compounds were examined for their antiproliferative effects in normal and tumor cells in vitro; they demonstrated moderate activity and a certain degree of selectivity. The compounds were also shown to efficiently inhibit the selenoenzyme thioredoxin reductase (TrxR), whereas they were poorly effective towards the glutathione reductase (GR) and glutathione peroxidase enzymes. Effect of potential antidotes on the acute toxicity of acrylonitrile

Rats were intoxicated with lethal doses of acrylonitrile by different routes of application, and the effect of potential antidotes was studied. The cyanide antidotes 4-dimethylaminophenol plus thiosulfate showed some protective effect only after oral but not after i.p. or inhalatory acrylonitrile application. Of the sulfhydryl compounds cysteine, N-acetylcysteine, cysteamine and diethyldithiocarbamate the two antidotes cysteine and, to some lesser extent, N-acetylcysteine proved especially effective. Antioxidant properties of human serum albumin (HSA) may explain part of its beneficial role in various diseases related to free radical attack. In the present study, the antioxidant role of Cys and Met was studied by copper-mediated oxidation of human low density lipoproteins and by free radical-induced blood hemolysis which essentially assessed metal-chelating and free radical scavenging activities, respectively. Mild conditions were set up to specifically modify Cys and Met residues by N-ethylmaleimide (NEM) and chloramine T treatments, respectively. Elicitation of defense reactions in tobacco by cryptogein, triggered a production of active oxygen species (AOS) via the NADPH oxidase, NtrbohD, and an accumulation of beta1din, a defense induced beta-type subunit of 20S proteasome. The proteasome inhibitor, MG132, stimulated this AOS production. Tobacco cells transformed with sense constructs of beta1din showed an inhibition of the AOS production following elicitin treatment, whereas the antisense transformed cells showed a strongly enhanced AOS production. The barley cysteine proteinase B (EPB) is the main protease responsible for the degradation of endosperm storage proteins providing nitrogenous nutrients to support the growth of young seedlings. The expression of this enzyme is induced in the germinating seeds by the phytohormone, gibberellin, and suppressed by another phytohormone, abscisic acid. In situ hybridization experiments indicate that EPB is expressed in the scutellar epithelium within 24 h of seed germination, but the aleurone tissue surrounding the starchy endosperm eventually becomes the main tissue expressing this enzyme.
