RNA interference (RNAi) pathways have evolved as important modulators of gene expression that operate in the cytoplasm by degrading RNA target molecules through the activity of short (21-30 nucleotide) RNAs1-6. RNAi components have been reported to have a role in the nucleus, as they are involved in epigenetic regulation and heterochromatin formation(7-10). Specific Defects in Different Transcription Complexes Compensate for the Requirement of the Negative Cofactor 2 Repressor in Saccharomyces cerevisiae

Abstract Negative cofactor 2 (NC2) has been described as an essential and evolutionarily conserved transcriptional repressor, although in vitro and in vivo experiments suggest that it can function as both a positive and a negative effector of transcription. NC2 operates by interacting with the core promoter and components of the basal transcription machinery, like the TATA-binding protein (TBP). In this work, we have isolated mutants that suppress the growth defect caused by the depletion of NC2. A DNA region ofTorulaspora delbrueckii containing theHIS3 gene: sequence, gene order and evolution

We cloned a genomic DNA fragment of the yeast Torulaspora delbrueckii by complementation of a Saccharomyces cerevisiae his3 mutant strain. DNA sequence analysis revealed that the fragment contained two complete ORFs, which share a high similarity with S. cerevisiae His3p and Mrp51p, respectively. The cloned TdHIS3 gene fully complemented the his3 mutation of S. cerevisiae, confirming that it encodes for the imidazoleglycerol-phosphate dehydrate of T. delbrueckii. Two additional ORFs, with a high homology to S. cerevisiae PET56 and DED1 genes, were mapped upstream and downstream from TdHIS3 and TdMRP51, respectively. Down-Regulation of Ku Autoantigen, DNA-Dependent Protein Kinase, and Poly(ADP-ribose) Polymerase during Cellular Senescence

During aging and cellular senescence mutations accumulate in genomic and mitochondrial DNA. Ku autoantigens, DNA-dependent protein kinase, and poly (ADP-ribose) polymerase have an essential role in DNA damage recognition. Our purpose was to find out whether cellular senescence of fibroblasts affects the protein components that recognize DNA damage and induce the repair process. We compared presenescent and replicatively senescent human WI-38 fibroblasts with each other and with SV-40 immortalized and serum-deficient quiescent WI-38 cells. Molecular Typing Reveals Frequent Clustering among Human Isolates of Listeria monocytogenes in Italy

In Italy, the annual incidence of reported cases of listeriosis amounts in recent years (2004 to 2006) to 0.8 cases per million inhabitants. Our study is a subtyping analysis by serotyping, ribotyping, and pulsed-field gel electrophoresis analysis of 44 human isolates from apparently sporadic cases of infection in the Lombardy region and in the Province of Florence, Italy, in the years 1996 to 2007. Based on the results of the different subtyping methods, 10 occasions were detected when strains of L. monocytogenes with the same subtype were isolated from more than one listeriosis case. Settore ICAR/14 - Composizione Architettonica E UrbanaThe essay questions the future's possibilities to operate on the buildings of the second half of the twentieth century according to the Italian Code of Cultural Heritage (2004-2011). When arguing how to encourage the re-use of these artifacts which are often marked by short-life technology cycles obsolescence and abandonment the ambiguities and inconsistencies of current legislative procedures deeply stress the cultural role of research experiences that use project-oriented methodologies. The comparison between some Italian and other European cases highlights the role of a design approach in the construction of a transformative protection perspective for the built matter of the second half of the twentieth century. In a framework aiming to encourage a reconciliation between the recent built environment and the new paradigms of sustainability and circular economy architecture must recognize re-cycle practices as a main theoretical and procedural node. To modernize the late Modern built environment with contemporary construction components is not simply a matter of technical knowledge and expertise: but a theme with theoretical density aesthetic and ethical values. The architectural project in this field today must ask for greater congruence between the bureaucratic and administrative levels and the real consistency of this corpus. It is a matter of identifying strategies to equip it with capabilities meanings and contents remembering that demolition wastes have unsustainable environmental costs and uncertain results concerning the necessary new attribution of political and semantic meaning that these places await.

Il contributo ha l’obiettivo di studiare alcuni casi di ricostruzione dell’abside eseguiti in edifici chiesastici palermitani nella seconda metà del XVI secolo. Si tratta delle chiese di San Sebastiano, San Francesco d’Assisi e San Martino delle Scale (all’interno dell’omonima abbazia), in cui, nella seconda metà del Cinquecento, per motivi liturgici, l’abside originaria viene “trasformata”, in un’abside “piatta”, al fine di potervi inserire il Coro ligneo. These are the churches of St. Sebastiano St. Francesco d’Assisi and St. Martino delle Scale (in the homonymous monastery) in wichfor liturgical reasons the original apse was "transformed" in a "flat" apse to insert in it the wooden choir. The new positioning of the choir after the Tridentine Reform was a central issue in the Europe of the modern age so even in Palermo there were some debates and controversies about this problem. The change of the shape of the apse determined in fact some considerable constructive problems related to its coverage.

Con la sentenza n. 16253/2020 la Suprema Corte ha affermato che l’assenza di nesso causale tra la cessazione di un appalto e il licenziamento del lavoratore integra un’ipotesi di manifesta insussistenza del fatto posto alla base del licenziamento - con conseguente applicabilità della tutela reintegratoria attenuta di cui all’art. 18, comma 5, della legge n. 300/1970 - non riconducibile, pertanto, agli altri casi in cui il giudice dichiara risolto il rapporto concedendo la sola tutela indennitaria forte (da dodici a ventiquattro mensilità).

Nella fiaba illustrata di Dino Buzzati La famosa invasione degli orsi in Sicilia (1945) e nella sua trasposizione filmica ad opera di Lorenzo Mattotti (2019) l’ambientazione siciliana è determinante. In entrambi i casi la Sicilia è un’isola con un grande vulcano – trasfigurata in un ambiente fantastico o riscritta secondo alcuni stereotipi – che i due autori utilizzano con accezioni diverse nella costituzione dell’ecosistema delle loro narrazioni. In both cases Sicily is an island with a large volcano – transfigured into a fantastic setting or rewritten according to certain stereotypes – that the two authors use with different meanings within the ecosystem of their narratives. Only through a comparison between the two works will it be possible to distinguish the representation of the two Sicilies which are narrated within an interval of about seventy years. If one does not consider the story of the Bears merely a children’s fairy tale using simplistic stylistic devices but on the contrary sees it as an opportunity for the author to deal with serious and complex issues such as an ante litteram Ecocritical perspective or questions of inter-group conflicts derived from Subaltern or Migration Studies the images of the two Sicilies can provide altogether new interpretations.
