Sesquiterpenes from Centaurea aspera

Abstract The aerial parts of two subspecies of Centaurea aspera L. (Asteraceae) yielded the germacranolides 1a – h , 2 , 3 , 4 and 5 , the elemane derivatives 6d and 6f , the lignan matairesinol, the degraded terpene loliolide, and the onopordopicrin–valine dimeric adduct 7 . From these, compounds 1e , 3 and 6d are natural products. The chemical composition of the two subspecies is very similar, a circumstance which does not support a taxonomic subdivision of the species.

Acid rearrangment of epoxy-germacranolides and absolute configuration of 1beta, 10alpha-epoxy-salonitenolide

The acid-catalyzed cyclization of mono epoxides of cnicin acetonide (3) was investigated. Several 6,12-eudesmanolides were obtained, and their stereochemistry established by extensive spectroscopic analyses. Chemical correlations also led to the assignment of the absolute configuration of 1beta,10alpha-epoxy-salonitenolide (13), a previously isolated natural product. The cytotoxic activities of some compounds were determined against A549 and MCF-7 tumor cell lines. The esterified germacranolides 2-6 were selectively cytotoxic against the MCF-7 breast cancer cell line.

The genus Centaurea Linnaeus (1753: 909) (Asteraceae) comprises a large number of taxa mainly occurring in the Mediterranean region and southwest Asia (Susanna & Garcia-Jacas 2007). Centaurea seridis Linnaeus (1753: 915) is distributed from south Europe (Spain incl. Balearic Islands, Italy incl. Sicily, France, Albania and Greece) to northwest Africa (Algeria, Morocco and Tunisia). From a nomenclatural standpoint, Centaurea seridis has not been typified (Jarvis 2007: 402). This paper proposes the designation of a lectotype for this Linnaean name, based on the analysis of the protologue and original material.

The genus Centaurea is recognized in folk medicine for anti-inflammatory, anti-itch, antitussive, purgative, astringent, and tonic activities. To study the chemical determinant for antimicrobial activity essential oils (EOs), five Centaurea species were analyzed including: C. scoparia, C. calcitrapa, C. glomerata, C. lipii and C. alexandrina. Conventional hydro-distillation (HD) and microwave-assisted extraction (MAE), as new green technologies, were compared for the extraction of essential oils. A floristic investigation was carried out in “Pizzo Cane, Pizzo Trigna and Grotta Mazzamuto” Nature Reserve, one of the largest in Sicily. In spite of the considerable botanical interest of this area, few data were available from classic and recent literature. Based on field investigations, coupled with bibliographic and herbarium researches, we have listed 951 infrageneric taxa belonging to 103 families and 446 genera. A lot of these entities are reported in this area for the first time, for others ancient records are confirmed. In the present study, the chemical composition of the essential oil from aerial parts of the endemic plant species, Centaurea vandasii Velen (sect. Phalolepis, Asteraceae), collected in its locus classicus in Rhodope Mts, Bulgaria, was evaluated by GC-MS. The main components of the oil were hexadecanoic acid (18.3%), tetradecanoic acid (13.8%), caryophyllene oxide (12.1%) and germacrene D (8.4%).
