The mechanism of aquaporin inhibition by gold compounds elucidated by biophysical and computational methods

The inhibition of water and glycerol permeation via human aquaglyceroporin-3 (AQP3) by gold(iii) complexes has been studied by stopped-flow spectroscopy and, for the first time, its mechanism has been described using molecular dynamics (MD), combined with density functional theory (DFT) and electrochemical studies. The obtained MD results showed that the most effective gold-based inhibitor, anchored to Cys40 in AQP3, is able to induce shrinkage of pores preventing glycerol and water permeation. Though most of the studies have focused on the effects of free fatty acids on T-cell activation, fatty acids incorporated into plasma membrane phospholipids may also affect cell signaling via diacylglycerol (DAG), generally produced by phospholipid hydrolysis. In the present study, we have synthesized a DAG-containing oleic acid and studied its implication in the modulation of calcium signaling in human Jurkat T-cells. 1-palmitoyl-2-oleoyl-sn-glycerol (POG) induced a dose-dependent increase in [Ca(2+)](i). Reduction of total lipids (TL) content and significant variations of triglyceride (TG) and phospholipid (PL) fractions were observed as a consequence of exposure of Ciona intestinalis ovaries to tributyltin chloride (TBTCL) solutions. In particular, an evident TG decrease and a PL increase were observed, which probably provoked an increment in membrane fluidity, because of the high concentration of long chain fatty acids and, as a consequence, PL. El presente trabajo tiene como objetivo profundizar en el análisis de los patrones de comportamiento espacio-temporal de los visitantes en destino, una escala de trabajo insuficientemente desarrollada. La investigación aplica un enfoque multimétodo, que combina el uso de GPS con una encuesta, al conocimiento de los patrones espaciales de los cruceristas que visitan la ciudad de Valencia de manera independiente. Los resultados obtenidos evidencian la polarización de los flujos en el centro histórico de la ciudad, dentro de un patrón mayoritario de único nodo, una dinámica que conviene gestionar adecuadamente para prevenir la congestión y poner en valor otros atractivos urbanos.
