Multicomponent solid dispersion as a formulation strategy to improve drug permeation: A case study on the anti-colorectal cancer irinotecan
2019
Abstract Multicomponent solid dispersions (MSD)s are frequently proposed as efficient drug delivery systems to improve drug solubility and bioavailability. In this study, the effects of specific excipients, such as mannitol, inulin, poly(methyl methacrylate-co-methacrylic)acid (PMMA) and cellulose acetate phthalate (CAP) have been tested to potentially improve irinotecan (IRN) permeation in the intestinal tract with the intention to protect the drug from the gastric environment. MSDs were formulated as microparticles by Spray-Drying technique. Raw materials and microparticles have been characterized by FTIR analysis to determine hydrogen bonding. SEM images were recorded to investigate morp…
Ionic Liquid Based Headspace Solid-Phase Microextraction-Gas Chromatography for the Determination of Volatile Polar Organic Compounds
2010
Solid-phase microextraction (SPME) with an ionic liquid (IL) coating was developed for headspace extraction of a group of low molecular weight alcohols (ethanol, n-propanol, butanol, and isopropyl alcohol), acetone, ethyl acetate, and acetonitrile. A first SPME fiber was simply coated with a dedicated IL whose synthesis is described. A second SPME fiber was prepared by gluing silica (Si) particles on which the synthesized IL was chemically bonded. The analytes SPME extraction was optimized for time, temperature, and NaCl salting out content. The headspace extracted analytes were determined by simple temperature desorption into the hot injection port of a gas chromatograph. The coated-IL fib…
Thin-layer chromatography of chlorinated anisoles and veratroles
1989
The thin-layer chromatography of chlorinated anisoles (methoxybenzenes) and veratroles (1,2-dimethoxybenzenes) has been examined on silica gel G60 and RP-18 thin-layer plates. More than fifty solvent systems were screened and some of them recommended for particular separations. Acetone was shown to be suitable for group separation of both chlorinated anisoles and veratroles on a silica gel G60 stationary phase having a very narrow range of RF values. On the other hand, benzene, dichloromethane and the various mixtures of light petroleum (b.p. 40–60°C) and a more polar eluent (such as diethyl ether, acetone or ethyl acetate) were recommended for separation of certain individual isomers. The …
Liquid–liquid equilibria of the systems isobutyl acetate+isobutyl alcohol+water and isobutyl acetate+isobutyl alcohol+glycerol at different temperatu…
2008
Abstract In this work, experimental liquid–liquid equilibria (LLE) data of the isobutyl acetate + isobutyl alcohol + water and isobutyl acetate + isobutyl alcohol + glycerol systems are presented. The LLE of both systems have been measured at 283.15 and 323.15 K. The NRTL and UNIQUAC models were applied to both ternary systems. The interaction parameters obtained from both models successfully correlated the equilibrium compositions. The experimental tie lines were compared to the values predicted by the UNIFAC method. Moreover, the solvent capabilities of water and glycerol were compared.
Flavans and other chemical constituents of Crinum biflorum (Amaryllidaceae)
2019
Abstract The ethyl acetate extract from the whole plant of Crinum biflorum Rottb. Showed a moderate activity against Enterococcus faecalis. Its phytochemical investigation led to the isolation of a new flavan-3-ol derivative namely (2R,3R)-3-hydroxy-7-methoxy-3′,4′-methylenedioxyflavan, together with (2S)-7-hydroxy-3′,4′-methylenedioxyflavan, (2R,3R)-7-methoxy-flavan-3-ol, (2S)-7-hydroxy-3′,4′-dimethoxyflavan, 3′,7-dihydroxy-4′-methoxyflavan, 4′,7-dimethoxy-3′-hydroxyflavan, farrerol, β-sitosterol, β-sitosterol-3-O-β-D-glucopyranoside, oleanolic acid, kaempferol, pancratistatin, lupeol, aurantiamide acetate, Narciprimine and 2,3-dihydroxypropyl palmitate. Their structures were elucidated ma…
Improved detection of alkaloids in crude extracts applying capillary electrophoresis with field amplified sample injection
1997
Abstract A simple and effective method for the sensitive detection of alkaloids in crude plant extracts applying capillary electrophoresis with field amplified sample injection (FASI) is described. This method was compared with normal pressure injection for the determination of alkaloids in methanolic extracts from roots of Berberis vulgaris L. (Berberidaceae) and Hydrastis canadensis L. (Ranunculaceae) using a 1:1 mixture of 200 mM ammonium acetate at pH 3.1 and methanol. By introducing a short plug of 70% methanol (v/v) before electrokinetic injection with 16 kV for 8 s the concentration sensitivity was 1000-times higher compared to hydrodynamic injection for 1 s. No difference between bo…
Influence of the food matrix structure on the retention of aroma compounds.
2000
The retention of the aroma compounds in a multicomponent medium like the food matrix is influenced by their affinity with the protein when lipid is present at a low level (0.5%). The effect of the structure of the media is also studied by using two media with the same composition; one was emusified, and the other was not. Among the studied aroma compounds, 2-nonanone and isoamyl acetate present opposite behaviors: the volatility of isoamyl acetate is not affected by the change of the medium structure whereas that of 2-nonanone increases. The decrease of retention of 2-nonanone in an emulsified system would be due to a modification of the fixation site for this compound on the protein or to …
Two heterologously expressed Planobispora rosea proteins cooperatively induce Streptomyces lividans thiostrepton uptake and storage from the extracel…
2010
Abstract Background A bacterial artificial chromosomal library of Planobispora rosea, a genetically intractable actinomycete strain, was constructed using Escherichia coli-Streptomyces artificial chromosome (ESAC) and screened for the presence of genes known to be involved in the biosynthesis of antibiotics. Results One clone with a 40 kb insert showed antimicrobial activity against Gram positive bacteria. Insert sequence analysis and subcloning experiments revealed that the bioactivity was due to a 3.5 kb DNA fragment containing two open reading frames. These orfs encode two proteins with high similarity to a putative membrane protein of Streptomyces coelicolor and to the nogalamycin resis…
Induced secondary metabolites from the endophytic fungus Aspergillus versicolor through bacterial co-culture and OSMAC approaches
2018
Abstract Two new cryptic 3,4-dihydronaphthalen-(2H)-1-one (1-tetralone) derivatives, aspvanicin A (1) and its epimer aspvanicin B (2), as well as several known cryptic metabolites (3–8), were obtained from the ethyl acetate extract of the co-culture of the endophytic fungus Aspergillus versicolor KU258497 with the bacterium Bacillus subtilis 168 trpC2 on solid rice medium. When A. versicolor was cultured axenically in liquid Wickerham medium supplemented with 3.5% DMSO, an additional three known secondary metabolites (9–11) were isolated that were lacking when the fungus was fermented on rice medium. The structures of the new compounds were elucidated using one- and two-dimensional NMR spec…
Absolute Configurations of Three Naturally Occurring Acetylenic Spiroacetal Enol Ethers
1993
The absolute configurations of three 6,7-oxygenated acetylenic spiroacetal enol ethers were determined by using the kinetic method of Horeau. The results were confirmed by the circular dichroism of the compounds in correlation with the corresponding 6,7-epoxide.
