Revisited BIA-MS combination: Entire "on-a-chip" processing leading to the proteins identification at low femtomole to sub-femtomole levels

International audience; We present the results of a study in which biomolecular interaction analysis (BIA, Biacore 2000) was combined with mass spectrometry (MS) using entire "on-a-chip" procedure. Most BIA-MS studies included an elution step of the analyte prior MS analysis. Here, we report a low-cost approach combining Biacore analysis with homemade chips and MS in situ identification onto the chips without elution step. Sperm analysis following World Health Organization guidelines is unable to explain the molecular causes of male infertility when basic sperm parameters are within a normal range and women do not present gynecologic pathology. Consequently, there is a need for accurate diagnostic tools in this area, and microarray technology emerges as promising. We present, for the first time, preliminary results of a comparison of sperm mRNA expression profiles between fertile and infertile men with normal semen parameters, discovering profound discrepancies between groups, with potential diagnostic and therapeutic possibilities.

Luminometric Sub-nanoliter Droplet-to-droplet Arrays (LUMDA-chip) for Drug Screening on Phase I Metabolism Enzymes

Here we show the fabrication of the Luminometric Sub-nanoliter Droplet-to-droplet Array (LUMDA chip) by inkjet printing. The chip is easy to be implemented and allows for amultiplexed multi-step biochemical assay in sub-nanoliter liquid spots. This concept is here applied to the integral membrane enzyme CYP3A4, i.e. themost relevant enzymatic target for phase I drug metabolism, and to some structurally-related inhibitors.

Model and Control Strategy Simulation of a Racing Series Hybrid Car

Action-reaction principle of Energetic Macroscopic Representation (EMR) and its Inversion Based Control (IBC) is used to organize subsystems interconnection of Noao, a plug-in series hybrid race car equipped with lithium-ion battery pack and an engine/generator set as its range extender, according to the physical causality. Results from drive test of the real car are used to validate the model. The objective of this paper is to provide a dynamic model of this car in order to improve its control scheme and analyse the impact of its optimisation.

Additions to knowledge of the biodiversity of benthic hydroids (Cnidaria: Hydrozoa) in the Balleny Islands (Antarctica)

The Balleny Islands, an isolated archipelago located 240 km off the coast of Victoria Land (East Antarctica), are separated from the Antarctic continental shelf by waters more than 2000 m deep. The benthic hydroid fauna of these islands is little known, with only 25 species reported so far. The present study contributes to a better knowledge of the benthic hydrozoans inhabiting this archipelago by studying a small collection held at the National Institute of Water and Atmospheric Research Invertebrate Collection at Wellington, New Zealand. The chapter first examines the major characteristics of the climate, dominated by the seasonal alternation of the Amihan and Habagat monsoons, leading to sharp differences between regions for their patterns of rainy seasons. Preparation of Biomolecule Microstructures and Microarrays by Thiol-ene Photoimmobilization

A mild, fast and flexible method for photoimmobilization of biomolecules based on the light-initiated thiol-ene reaction has been developed. After investigation and optimization of various surface materials, surface chemistries and reaction parameters, microstructures and microarrays of biotin, oligonucleotides, peptides, and MUC1 tandem repeat glycopeptides were prepared with this photoimmobilization method. Furthermore, MUC1 tandem repeat glycopeptide microarrays were successfully used to probe antibodies in mouse serum obtained from vaccinated mice.
