Effects of cimetidine, atropine and prostaglandin E2 on rat mucosal erosions produced by intragastric distension

Abstract The effects of three typical antisecretory agents: cimetidine, atrophine and prostaglandin E2 were compared on an acute rat gastric ulcer model which consisted of perfusing the stomach continuously, at a high intraluminal pressure (120 mm H2O), with a simulated gastric juice (0.1 M HCl plus 600 mg pepsin/1). As the acid and pepsin are given exogenously the inhibitory action of the antisecretory drugs is obviated in this model. Cimetidine and atropine failed to reduce gastric erosions, whereas prostaglandin E2 markedly reduced the severity of the mucosal lesions with respect to control values. Although histamine release is recognised as a common event during anaesthesia and surgery, few clinicians judge the resultant cardiorespiratory disturbances serious enough to warrant prophylaxis with antihistamines. We have assessed the incidence and importance of histamine release in a randomised 2 x 2 factorial study. 240 patients representing a routine throughput of major general surgery were studied during a standardised induction of anaesthesia and preoperative loading of the circulation with either Ringer solution or Haemaccel-35, with or without antihistamine prophylaxis with dimetindene (H1) plus cimetidine (H2). Nach Gabe von Cimetidin kann die Wirkungszeit nichtdepolarisierender Muskelrelaxanzien verlangert sein. Wir untersuchten diesen Zusammenhang fur Rocuronium nach einer Einzeldosis von 400 mg Cimetidin intravenos im Vergleich mit einer Kontrollgruppe. Die Antwort des M. adductor pollicis nach Stimulation des N. ulnaris wurde elektromyographisch registriert. Die Anschlags- und Erholungszeiten sind in beiden Gruppen vergleichbar. Der fehlende Einflus von Cimetidin auf die neuromuskulare Blockade von Rocuronium konnte darauf hindeuten, das die Elimination von Rocuronium nicht nur in der Leber stattfindet. Histamine binding to H2 receptors stimulates phospholipid methylation in mast cells

Rat peritoneal mast cells incubated in vitro in the presence of L-[methyl-3H] methionine and exposed to histamine undergo a rapid but transient increase in phospholipid methylation. By using specific H1- and H2-receptor antagonists, and histamine analogues differing in their H2-receptor agonist potency, it has been demonstrated that this metabolic event is dependent on histamine binding to H2-receptors.

In vivo cimetidine immunomodulatory effects on the cutaneous reaction to oxazolone in the chicken.

The effect of the histamine H2-receptor antagonist, cimetidine, on the cutaneous Arthus-like hypersensitivity to oxazolone elicited injecting subcutaneously oxazolone conjugated to egg-albumin (EA-OX) has been examined in the chicken. Cimetidine had opposite effects on the cutaneous reaction to oxazolone in relation to a different immunization schedule. Cimetidine enhanced the cutaneous reaction to oxazolone obtained immunizing chickens with oxazolone dissolved in ethanol (Eth-OX); instead cimetidine inhibited the cutaneous reaction obtained in chickens immunized with oxazolone dissolved in complete Freund adjuvant (CFA-OX). Effects of histamine on spontaneous electrical activity of neurons in rat suprachiasmatic nucleus

Abstract The hypothalamic suprachiasmatic nucleus (SCN) is thought to be a light-entrained pacemaker in mammals, inducing a wide range of endogenous circadian events. In rat brain, histaminergic (HAergic) fibres are particulary rich in the hypothalamus. This prompted an investigation of the influence of bath-applied HAergic compounds on the spontaneous electrical activity of SCN neurons, recorded extracellularly in the hypothalamic slice preparation. Cells activated by bath application of HA ( n = 28) outnumbered those inhibited by HA ( n = 6). 48% of cells tested ( n = 28) were unresponsive to HA application. Responses to histamine and selective H2-receptor agonists in lung parenchymal strips from normal and sensitized guinea-pigs.

Histamine produces concentration-dependent contractions of lung parenchyma strips obtained from normal and sensitized guinea-pigs. The responsiveness of the sensitized lung strips to histamine was significantly increased compared to normal tissues. Clemizole (0.1 microM) was equally effective as an H1-antagonist in normal (dose-ratio 9.12) and sensitized (dose-ratio 9.77) tissues. The concentration-response curves to histamine were displaced to the left by cimetidine (0.1 microM to 0.1 mM) with similar dose-ratios in normal and sensitized tissues. Involvement of prostaglandins and 5-hydroxytryptamine in the contractile effect of platelet-activating factor in rat isolated gastric corpus

Abstract The present study characterizes the nature of the response to the platelet-activating factor (PAF) in isolated gastric corpus with and without mucosa. PAF (10-8 M) induced contraction of rat isolated gastric corpus strips followed by desensitization of this tissue. Incubation of strips with the specific PAF-receptor antagonist WEB 2086 (5 × 10-8 − 5 × 10-5 M), the prostaglandin blocker indomethacin (10-6 M) and the 5-hydroxytryptamine antagonist methysergide (10-5 M) reduced significantly the contraction induced by PAF.
