Biodegradable pH-Sensitive Poly(ethylene glycol) Nanocarriers for Allergen Encapsulation and Controlled Release
2015
In the last decades, the number of allergic patients has increased dramatically. Allergen-specific immunotherapy (SIT) is the only available cause-oriented therapy so far. SIT reduces the allergic symptoms, but also exhibits some disadvantages; that is, it is a long-lasting procedure and severe side effects like anaphylactic shock can occur. In this work, we introduce a method to encapsulate allergens into nanoparticles to avoid severe side effects during SIT. Degradable nanocarriers combine the advantage of providing a physical barrier between the encapsulated cargo and the biological environment as well as responding to certain local stimuli (like pH) to release their cargo. This work int…
Poly(Ethylene Glycol) Dimethacrylates with Cleavable Ketal Sites: Precursors for Cleavable PEG-Hydrogels.
2017
The authors introduce poly(ethylene glycol) (PEG) based macromonomers containing acid-labile ketal moieties as well as terminal methacrylate units that are amenable to radical polymerization. The synthesis of PEGs of different molecular weights (ranging from 2000 to 13 000 g mol-1 with polydispersities <1.15) with a central ketal unit (PEG-ketal-diol) and their conversion to PEG-ketal-dimethacrylates (PEG-ketal-DMA) is introduced. Degradation rates of both PEG-ketal-diols and PEG-ketal-DMA are investigated by in situ 1 H NMR kinetic studies in deuterated phosphate buffer. Hydrogels containing 0, 5, or 10 wt% of PEG-ketal-DMA and 100, 95, or 90 wt% of PEG-DMA, respectively, are synthesized a…
Synthesis of Novel Folic Acid-Functionalized Biocompatible Block Copolymers by Atom Transfer Radical Polymerization for Gene Delivery and Encapsulati…
2005
Two synthetic routes to folic acid (FA)-functionalized diblock copolymers based on 2-(methacryloyloxy)- ethyl phosphorylcholine [MPC] and either 2-(dimethylamino)ethyl methacrylate [DMA] or 2-(diisopropylamino) ethyl methacrylate [DPA] were explored. The most successful route involved atom transfer radical polymerization (ATRP) of MPC followed by the tertiary amine methacrylate using a 9-fluorenylmethyl chloroformate (Fmoc)-protected ATRP initiator. Deprotection of the Fmoc groups produced terminal primary amine groups, which were conjugated with FA to produce two series of novel FA-functionalized biocompatible block copolymers. Nonfunctionalized MPC-DMA diblock copolymers have been previou…
Cellular Uptake of DNA Block Copolymer Micelles with Different Shapes
2008
The cellular uptake of DNA block copolymer micelles composed of DNA-b-PPO in Caco-2 cells was studied. In particular it was investigated if the shape of micelle aggregates influences the internalization. Rod-like polymeric particles were taken up 12 times more efficiently than their spherical counter parts although they were composed of the same constituents. Furthermore, it was observed that internalization of all the micelle systems was more efficient than the pristine DNA controls. A cytotoxicity assay proved the non-toxic nature of DNA-b-PPO micelle aggregates.
Copper carbonate as a solid-bed reactor for spectrophotometric determination of doxycycline and oxytetracycline in an unsegmented continuous flow ass…
1993
The FIA-spectrophotometric determination of doxycycline was carried out by reaction of the drug with cupric ions entrapped in a polymeric material in a packed-bed reactor: the complex formed was then injected into a manifold with an alkaline solution as carrier. The developed colour was monitored at 395.0 nm. The method was applied to the determination of doxycycline in different pharmaceutical formulations. The calibration graph for doxycycline hyclate was linear over the range 10.0-80.0 mg ml-1 (n = 8) with a relative standard deviation of 1.4% (at 25 mg ml-1) and a sample throughput of 128 h-1. The proposed procedure was also applied to the determination of oxytetracycline in pharmaceuti…
Squaric Ester-Based, pH-Degradable Nanogels: Modular Nanocarriers for Safe, Systemic Administration of Toll-like Receptor 7/8 Agonistic Immune Modula…
2021
Small-molecular Toll-like receptor 7/8 (TLR7/8) agonists hold promise as immune modulators for a variety of immune therapeutic purposes including cancer therapy or vaccination. However, due to their rapid systemic distribution causing difficult-to-control inflammatory off-target effects, their application is still problematic, in particular systemically. To address this problem, we designed and robustly fabricated pH-responsive nanogels serving as versatile immunodrug nanocarriers for safe delivery of TLR7/8-stimulating imidazoquinolines after intravenous administration. To this aim, a primary amine-reactive methacrylamide monomer bearing a pendant squaric ester amide is introduced, which i…
Accelerated weathering of polypropylene/wood flour composites
2008
Abstract Wood–plastic composites (WPCs) have received increasing attention during the last decades, because of many advantages related to their use. Some of their main applications are represented by outdoor furnishing and decking; therefore, it is important to assess their behaviour under UV exposure. In this work, polypropylene/wood flour composites were prepared and their resistance to photooxidation investigated. The composites were prepared by extrusion and compression moulding, and were subjected to mechanical tests, FTIR analysis and molecular weight measurements. The results showed that the composites retained a higher fraction of the original mechanical properties after accelerated…
Reduction of the interfacial tension between ‘immiscible’ polymers: to which phase one should add a compatibilizer
2001
Abstract The reduction of interfacial tension σ between polyethylene oxide (PEO) and polypropylene oxide (PPO) by either a triblock copolymer EO–PO–EO or a diblock copolymer styrene–EO was studied by means of the pendant drop method from 70 to 120°C. Time independent data correspond to stationary states and do not represent equilibrium information. The addition of the compatibilizer to the PPO phase is approximately twice as efficient as the addition to the PEO phase for both block copolymers. Knowing the concentration dependence of σ for one of the coexisting phases it is possible to forecast the effects resulting from the addition of the other phase by means of model considerations based …
Thermomechanical degradation of filled polypropylene
2003
The main problem in post-consumer plastics recycling is due to the thermomechanical stress acting on the melt during the reprocessing operations. The macromolecules break because of the temperature and of the mechanical stress. The extent of degradation is then correlated to the level of mechanical stress which, in turn, is proportional to the viscosity of the melt. The presence of fillers increases the viscosity of the polymers and then it is expected that the level of thermomechanical degradation of these systems is larger than that of the unfilled material. In this work the thermomechanical degradation kinetic of a polypropylene sample is investigated as a function of the calcium carbona…
Compared effects of synthetic and natural bile acid surfactants on xenobiotic absorption I. Studies with polysorbate and taurocholate in rat colon
1991
Some expected differences between synthetic and natural bile acid surfactants relative to their influences on xenobiotic absorption are briefly outlined on the basis of literature data. Then, experimental work is presented which shows that these differences exist and that they can be even more relevant than suspected. Absorption tests were developed in rat colon in situ with polysorbate (synthetic) and sodium taurocholate (natural) surfactants, using a homologous series of phenylalkylcarboxylic acids as test compounds. At the critical micelle concentration (CMC), the two previously reported actions of synthetic surfactants on xenobiotic absorption (i.e. the increase in absorbing membrane po…
