AMD3100: A Versatile Platform for CXCR4 Targeting 68 Ga-Based Radiopharmaceuticals

International audience; CXCR4 is a G protein-coupled receptor (GPCR), which is overexpressed in numerous diseases, particularly in multiple cancers. Therefore, this receptor represents a valuable target for imaging and therapeutic purposes. Among the different approaches, which were developed for CXCR4 imaging, a CXCR4 antagonist biscyclam system (AMD3100, also called Mozobil), currently used in the clinic for the mobilization of hematopoietic stem cells, was radiolabeled with different radiometals such as 62Zn, 64Cu, 67Ga, or 99mTc. First synthesis of single-“strapped cyclam-porphyrins”

The first synthesis of strapped cyclam porphyrins is described ; these will be studied as dioxygen carriers as well as cytochrome C oxidase models. The original derivatives are characterized by mass spectrometry and 1H NMR spectroscopy. The cofacial conformation of the two different macrocycles is confirmed by 1H NMR data.

Plerixafor with and without chemotherapy in poor mobilizers: results from the German compassionate use program.

The CXCR4-inhibitor plerixafor mobilizes hematopoietic stem cells amplifying the effects of granulocyte-CSF (G-CSF). Before approval plerixafor was used in a compassionate use program (CUP) for patients who failed a previous mobilization. In the German CUP 60 patients from 23 centers (median age 56.5 years (2-75)) were given 240 μg/kg plerixafor SC 9-11 h before apheresis. A total of 78.3% (47/60) received G-CSF for 4 days before plerixafor administration; 76.6% of those (36/47) yielded at least 2.0 × 10(6) CD34(+) cells/μL. Cyclam-functionalized silica-modified electrodes for selective determination of Cu(II)

Cyclam derivatives have been grafted via 1, 2 or 4 silylated arms to the surface of mesoporous silica and the resulting materials have been incorporated into carbon paste electrodes and applied to the preconcentration electroanalysis of Cu(II). Sensitive and selective detection of the target analyte was achieved owing to the attractive binding properties of this macrocyclic ligand towards Cu(II) species. Palladium-Catalysed Amination of 1,8- and 1,5-Dichloroanthracenes and 1,8- and 1,5-Dichloroanthraquinones

Diamino derivatives of anthracene and anthraquinone have been synthesised by palladium-catalysed coupling of 1,8-dichloroanthracene and 1,8-dichloroanthraquinone with a wide range of aliphatic and aromatic primary and secondary amines. The use of polyamines gave rise to a large number of new nitrogen- and oxygen-containing macrocycles incorporating anthracene or anthraquinone moieties. The method has also been employed for the preparation of bismacrocycles in which two cyclam or azacrown units are linked together by an anthracene bridge through C(sp2)−N bonds. Highly selective fluorescence detection of hydrogen sulfide by using an anthracene-functionalized cyclam-CuII complex

An anthracene-functionalised cyclam-copper(II) complex for the detection of HS- in aqueous environments has been prepared. This probe displays poor fluorescence but can selectively and sensitively detect HS- anions in water over other anions, biothiols and common oxidants such as H2O2 through remarkably enhanced emission. This turn-on response in the presence of the HS- anion is ascribed to a demetallation reaction that inhibits emission quenching observed in the initial complex as a result of the presence of the paramagnetic Cu2+ centre. En route toward the development of hybrid organic−inorganic extracting materials incorporating lead-selective chelators and their implementation in water purification processes, the lead(II) binding properties of three N-carbamoylmethyl-substituted 1,4,8,11-tetraazacyclotetradecanes (cyclams) have been fully investigated by spectroscopic (IR, UV−vis, MALDI-TOF MS, 1H and 13C NMR), X-ray crystallographic, potentiometric, and kinetic methods. A cyano-bridged bimetallic ferrimagnet: Synthesis, X-ray structure and magnetic study

Mixing of trans-[Mn(cyclam)Cl2]Cl (cyclam = 1,4,8,11-tetraazacyclotetradecane) and potassium hexacyanochromate (K3[Cr(CN)6]) aqueous solutions instantaneously yields a 1D infinite chain complex {[Mn(cyclam)(l-CN)2Cr(CN)4]� H2O}n (1). The crystal structure of 1, crystallizing in the monoclinic system with space group P21/n has been solved from X-ray powder diffraction data following direct space approach and refined by the Rietveld method. The structure analysis of 1 reveals alternating [Cr(CN)6] 3� and [Mn(cyclam)] 3+
