Production of β-glucosidases by European Armillaria species

International audience; Abstract Production of β-glucosidase was investigated in nine isolates of Armillaria representing four species found in Europe: Armillaria mellea and Armillaria ostoyae, considered to be pathogenic and moderately pathogenic, respectively, and Armillaria. gallica and Armillaria cepistipes, both considered to be non-pathogenic. β-glucosidase was predominantly produced in the rhizomorphs, while the vegetative mycelium produced only a low amount of enzyme. Mammalian fetuin-A and fetuin-B are abundant serum proteins with pleiotropic functions. Fetuin-B is a highly selective and potent inhibitor of metallopeptidases (MPs) of the astacin family, which includes ovastacin in mammals. By inhibiting ovastacin, fetuin-B is essential for female fertility. The crystal structure of fetuin-B was determined unbound and in complex with archetypal astacin, and it was found that the inhibitor has tandem cystatin-type modules (CY1 and CY2). They are connected by an exposed linker with a rigid, disulfide-linked `CPDCP-trunk', and are followed by a C-terminal region (CTR) with little regular secondary structure. Immunohistopathology in Diagnostic Neuropathology

Within the field of surgical pathology, immunohistochemistry is now frequently applied to the morphological diagnosis of lymphomas (11), certain carcinomas and other selected types of tumor (12). In neuropathology, the study of neuro-oncological and non-neoplastic diseases may also receive diagnostic support from performing immunohistological techniques, which encompass immunofluorescent and immunoperoxidase methods. The following report represents a survey of our experience in this recently developed field of diagnostic neuropathology.

Cellular mechanisms of IL-17-induced blood-brain barrier disruption.

Recently T-helper 17 (Th17) cells were demonstrated to disrupt the blood-brain barrier (BBB) by the action of IL-17A. The aim of the present study was to examine the mechanisms that underlie IL-17A-induced BBB breakdown. Barrier integrity was analyzed in the murine brain endothelial cell line bEnd.3 by measuring the electrical resistance values using electrical call impedance sensing technology. First synthesis of single-“strapped cyclam-porphyrins”

The first synthesis of strapped cyclam porphyrins is described ; these will be studied as dioxygen carriers as well as cytochrome C oxidase models. The original derivatives are characterized by mass spectrometry and 1H NMR spectroscopy. The cofacial conformation of the two different macrocycles is confirmed by 1H NMR data.

International audience; Three white-rot basidiomycetes, Bjerkandera adusta, Ischnoderma benzoinum and Dichomitus squalens, were cultivated on a liquid medium supplemented with l-phenylalanine, a precursor for benzaldehyde (bitter almond aroma) and benzyl alcohol. Remarkable amounts of benzaldehyde (587 mg l−1) were found in cultures of B. adusta. Immobilization of this fungus on polyurethane foam cubes allowed an 8.3-fold increase of the production of benzaldehyde and a 15-fold increase of the productivity as compared with non-immobilized cells. Type 2 diabetes mellitus (T2DM) is an important metabolic disorder for which there is an urgent need for new antidiabetic drugs. α-Glucosidase inhibition is an established protocol for T2DM therapy. pH Feedback Lifecycles Programmed by Enzymatic Logic Gates Using Common Foods as Fuels

Abstract Artificial temporal signaling systems, which mimic living out‐of‐equilibrium conditions, have made large progress. However, systems programmed by enzymatic reaction networks in multicomponent and unknown environments, and using biocompatible components remain a challenge. Herein, we demonstrate an approach to program temporal pH signals by enzymatic logic gates. They are realized by an enzymatic disaccharide‐to‐monosaccharide‐to‐sugar acid reaction cascade catalyzed by two metabolic chains: invertase‐glucose oxidase and β‐galactosidase‐glucose oxidase, respectively. Physico-Chemical Characterization of an Exocellular Sugars Tolerant Β-Glucosidase from Grape Metschnikowia pulcherrima Isolates

A broad variety of microorganisms with useful characteristics in the field of biotechnology live on the surface of grapes; one of these microorganisms is Metschnikowia pulcherrima. This yeast secretes a β-glucosidase that can be used in fermentative processes to liberate aromatic compounds. In this work, the synthesis of an exocellular β-glucosidase has been demonstrated and the optimal conditions to maximize the enzyme’s effectiveness were determined.
