Exploring alternative hapten tethering sites for high-affinity anti-picoxystrobin antibody generation

The relevance of the linker tethering site in haptens was investigated for antibody generation and immunoassay development. Three derivatives of the strobilurin fungicide picoxystrobin were synthesized with the same functionalized spacer arm located at three different positions. Protein conjugates of those haptens were employed as immunogens, and novel polyclonal antibodies were produced and characterized. All haptens afforded highly specific antibodies, but different affinities to the free analyte were observed among the obtained antisera. Science based calibration for the extraction of 'analyte-specific' HPLC-DAD chromatograms in environmental analysis

Multivariate science based calibration (SBC) has been applied to the resolution of overlapped peaks in liquid chromatography with diode array detection (LC-DAD). Complex river water samples spiked with 11 pharmaceutical substances resulted in poorly resolved chromatograms containing additional peaks from interfering matrix compounds and a change in the background absorbance due to the mobile phase gradient. Applying the present multivariate approach it was possible to resolve all 11 analytes from overlapping peaks, obtaining linear calibration lines (R2 > 0.96). Strategies for the enantiomeric determination of amphetamine and related compounds by liquid chromatography.

This paper summarizes recent research on the stereospecific analysis of amphetamine, its analogs and metabolites, by liquid chromatography. The different methods proposed have been evaluated and compared in terms of resolution power, time of analysis, sensitivity, or potential for automation. Chiral derivatization, followed by separation of the diastereomers formed in achiral chromatographic systems, is still the method preferred for the analysis of amphetamines at trace levels, as derivatization also improves analyte detectability. SIGNAL PROCESSING ALGORITHM FOR SCHLIEREN EFFECT CORRECTION IN FLOW ANALYSIS

Due to the radial refractive index gradients that sometimes develop in flow cells during the travelling of the samples through them, the absorbance records obtained in such conditions have a strange shape, particularly at low analyte levels. To correct this phenomenon, called Schlieren effect, a simple and reliable signal processing algorithm is described in correlation to the detector characteristics of noise. The algorithm is illustrated in nitrite and nitrate determinations with proflavin from more real samples. It has a solid theoretical support and involves the subtraction from the sample records of a smoothed signal, acquired by working with a blank solution.

A solid colorimetric sensor for the analysis of amphetamine-like street samples.

A solid sensor obtained by embedding 1,2-naphthoquinone-4-sulfonate (NQS) into polydimethylsiloxane/tetraethylortosilicate/silicon dioxide nanoparticles composite has been developed to identify and determine amphetamine (AMP), methamphetamine (MAMP), 3,4-methylenedioxymetamphetamine (MDMA) and 3,4-methylenedioxyamphetamine (MDA). The analytes are derivatized inside the composite for 10 min to create a colored product which can be then quantified by measuring the diffuse reflectance or the color intensity after processing the digitalized image. Preanalytical stability of a drug and its major metabolites is an important consideration in pharmacokinetic studies or whenever the analyte pattern is used to estimate drug habits. Firstly, the stability of free and glucuronidated 11-nor-delta9-tetrahydrocannabinol-9-carboxylic acid (THCCOOH, THCCOOglu) in authentic urine samples was investigated. Electrochemical pH Control at Gold Nanowires

In this work, interdigitated arrays of nanowire electrodes are used with one array acting as the working electrode while the other is used to generate the required protons. Finite element simulations of the pH control electrodes were performed to provide insight on the generation and subsequent diffusion of protons. This informed the inter-tine spacing of the electrodes used.. This electrochemical pH control method was then used to enable the detection of analytes of interest.

Immuno-supported liquid membrane (immuno-SLM) extraction is a new technique that makes use of antibody (Ab)-antigen interactions as the "extraction force" to drive the mass transfer in a selective way. In immuno-SLM, anti-analyte (Ag) Abs are introduced into the acceptor phase of the SLM unit to trap the Ag that passes from the flowing donor through the SLM into the stagnant acceptor.
