Endogenous fluctuations of DNA topology in the chloroplast of Chlamydomonas reinhardtii.

1998

DNA supercoiling in the chloroplast of the unicellular green alga Chlamydomonas reinhardtii was found to change with a diurnal rhythm in cells growing in alternating 12-h dark-12-h light periods. Highest and lowest DNA superhelicities occurred at the beginning and towards the end of the 12-h light periods, respectively. The fluctuations in DNA supercoiling occurred concurrently and in the same direction in two separate parts of the chloroplast genome, one containing the genes psaB, rbcL, and atpA and the other containing the atpB gene. Fluctuations were not confined to transcribed DNA regions, indicating simultaneous changes in DNA conformation all over the chloroplast genome. Because the d…

The redox state regulates RNA degradation in the chloroplast of Chlamydomonas reinhardtii.

1999

Abstract A Chlamydomonas reinhardtii chloroplast transformant, designated MU7, carrying a chimeric (rbcL promoter: β-glucuronidase [GUS]:psaB 3′ end) gene whose transcripts have been found previously to be unstable in light (half-life of 20 min in light as opposed to a half-life of 5 h in the dark), was used to study the role of electron transport and of the redox state in the degradation of chloroplast transcripts in the light. Blocking photosynthetic electron transport with 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) prevented the light-dependent breakdown of the pool of GUS transcripts in MU7 cells. Diamide, an oxidizing agent, caused a measurable delay in the degradation of GUS trans…

Processing of RNAs of the Family Avsunviroidae in Chlamydomonas reinhardtii Chloroplasts

2007

ABSTRACT The family Avsunviroidae comprises four viroid species with the ability to form hammerhead ribozymes that mediate self-cleavage of the multimeric plus and minus strands resulting from replication in the chloroplast through a symmetric rolling-circle mechanism. Research on these RNAs is restricted by their host range, which is limited to the plants wherein they were initially identified and some closely related species. Here we report cleavage and ligation in transplastomic Chlamydomonas reinhardtii expressing plus- and minus-strand dimeric transcripts of representative members of the family Avsunviroidae . Despite the absence of viroid RNA-RNA transcription, the C. reinhardtii -bas…

Cabut/dTIEG associates with the transcription factor Yorkie for growth control

2015

The Drosophila transcription factor Cabut/dTIEG (Cbt) is a growth regulator, whose expression is modulated by different stimuli. Here, we determine Cbt association with chromatin and identify Yorkie (Yki), the transcriptional co-activator of the Hippo (Hpo) pathway as its partner. Cbt and Yki co-localize on common gene promoters, and the expression of target genes varies according to changes in Cbt levels. Down-regulation of Cbt suppresses the overgrowth phenotypes caused by mutations in expanded (ex) and yki overexpression, whereas its up-regulation promotes cell proliferation. Our results imply that Cbt is a novel partner of Yki that is required as a transcriptional co-activator in growth…

Overlapped moving windows followed by principal component analysis to extract information from chromatograms and application to classification analys…

2015

Variable generation from chromatograms is conveniently accomplished using unsupervised rather than manual techniques. With unsupervised techniques, there is no need for selecting a few peaks for manual integration and valuable information is quickly and efficiently collected. The generation of variables can be performed by using either peak searching or moving window (MW) strategies. With a MW approach, the peaks are ignored and many variables, only part of them carrying information, are generated. Thus, variable generation by MWs should be followed by data compression to generate the variables to be further used for classification or quantitation purposes. In this work, unsupervised proces…

Antennas for the detection of radio emission pulses from cosmic-ray induced air showers at the Pierre Auger Observatory.

2012

The Pierre Auger Observatory is exploring the potential of the radio detection technique to study extensive air showers induced by ultra-high energy cosmic rays. The Auger Engineering Radio Array (AERA) addresses both technological and scientific aspects of the radio technique. A first phase of AERA has been operating since September 2010 with detector stations observing radio signals at frequencies between 30 and 80 MHz. In this paper we present comparative studies to identify and optimize the antenna design for the final configuration of AERA consisting of 160 individual radio detector stations. The transient nature of the air shower signal requires a detailed description of the antenna s…

Searches for Large-Scale Anisotropy in the Arrival Directions of Cosmic Rays Detected above Energy of $10^{19}$ eV at the Pierre Auger Observatory an…

2014

Spherical harmonic moments are well-suited for capturing anisotropy at any scale in the flux of cosmic rays. An unambiguous measurement of the full set of spherical harmonic coefficients requires full-sky coverage. This can be achieved by combining data from observatories located in both the northern and southern hemispheres. To this end, a joint analysis using data recorded at the Telescope Array and the Pierre Auger Observatory above 1019 eV is presented in this work. The resulting multipolar expansion of the flux of cosmic rays allows us to perform a series of anisotropy searches, and in particular to report on the angular power spectrum of cosmic rays above 1019 eV. No significant devia…

Performance of jet substructure techniques for large-$R$ jets in proton-proton collisions at $\sqrt{s}$ = 7 TeV using the ATLAS detector

2013

This paper presents the application of a variety of techniques to study jet substructure. The performance of various modified jet algorithms, or jet grooming techniques, for several jet types and event topologies is investigated for jets with transverse momentum larger than 300 GeV. Properties of jets subjected to the mass-drop filtering, trimming, and pruning algorithms are found to have a reduced sensitivity to multiple proton-proton interactions, are more stable at high luminosity and improve the physics potential of searches for heavy boosted objects. Studies of the expected discrimination power of jet mass and jet substructure observables in searches for new physics are also presented.…

Optimized Multipactor-Resistant Wedge-Shaped Waveguide Bandpass Filters

2013

[EN] Wedge-shaped waveguides present a certain advantage with respect to rectangular waveguides regarding their resistance to multipactor discharges. In this paper, the optimal configuration for the wedge geometry is investigated based on theoretical results, on a precise multipactor prediction tool, and on previous experience. In addition, design rules are presented, which allow us to achieve for wedge-shaped filters electrical performances comparable to the ones of rectangular waveguide filters, while at the same time improving the multipactor-free power range. As a proof of concept, two three-pole bandpass filters with equivalent electrical characteristic of 150-MHz bandwidth, centered a…

Circular dichroism of magnetically induced transitions for D 2 lines of alkali atoms

2018

In this letter we study magnetic circular dichroism in alkali atoms exhibiting asymmetric behaviour of magnetically induced transitions. The magnetic field induces transitions between hyperfine levels of alkali atoms and in the range of magnetic field, the intensities of these transitions experience significant enhancement. We have inferred a general rule applicable for the D 2 lines of all alkali atoms, that is the transition intensity enhancement is around four times larger for the case of than for excitation for , whereas it is several hundreds of thousand times larger in the case of than that for polarization for . This asymmetric behaviour results in circular dichroism. For experimenta…
