ABSTRACT Four subpopulations of a Plutella xylostella (L.) strain from Malaysia (F 4 to F 8 ) were selected with Bacillus thuringiensis subsp. kurstaki HD-1, Bacillus thuringiensis subsp. aizawai , Cry1Ab, and Cry1Ac, respectively, while a fifth subpopulation was left as unselected (UNSEL-MEL). Fluorescence In Situ Hybridization (FISH) is a powerful technique for localizing specific DNA targets directly in the fixed tissue or cells. Bacterial artificial chromosome (BAC) as well as commercial probes, which could be supplied ready for use or concentrated and must be diluted following the manufacturers instructions, can be used. The technique requires 2 days, as an overnight incubation of the FISH probes is needed for optimal hybridization. A link between ectoparasite infection and susceptibility to bacterial disease in rainbow trout

Rainbow trout, Oncorhynchus mykiss, were infected concomitantly with Argulus coregoni and Flavobacterium columnare and their survival was compared with that of fish infected with either the parasite or the bacterium alone. The mortality of fish challenged with A. coregoni was negligible while infection with F. columnare alone led to significantly lower survival. However, compared with single infections, the mortality was significantly higher and the onset of disease condition was earlier among fish, which were concomitantly infected by A. coregoni and F. columnare. Enhancing methane production from lignocellulosic biomass by combined steam-explosion pretreatment and bioaugmentation with cellulolytic bacterium

Background Biogas production from lignocellulosic biomass is generally considered to be challenging due to the recalcitrant nature of this biomass. In this study, the recalcitrance of birch was reduced by applying steam-explosion (SE) pretreatment (210 °C and 10 min). Moreover, bioaugmentation with the cellulolytic bacterium Caldicellulosiruptor bescii was applied to possibly enhance the methane production from steam-exploded birch in an anaerobic digestion (AD) process under thermophilic conditions (62 °C). Anaerobic and sequential anaerobic-aerobic treatments of municipal landfill leachate at low temperatures

Abstract Anaerobic treatment of municipal landfill leachate (COD 1–4 g l −1 , NH 4 -N c. 0.16 g l −1 ) was studied at 11 and 24°C by using laboratory-scale UASB and hybrid reactors. Aerobic post-treatment of the anaerobically treated leachate was also studied at 24°C using an activated-sludge process. In the 11°C reactors, a COD removal of up to 60–65% was obtained with 1.5-2 day HRT and 0.7-1.5 kg COD m −3 day −1 organic loading rates. At 24°C, a COD removal of up to 75% was achieved with a 10 h HRT. The highest organic loading rate applied was 10 kg COD m −3 day −1 at 24°C. Use of rumen microorganisms to boost the anaerobic biodegradability of microalgae

[EN] A laboratory bioreactor using rumen microorganisms to treat Scenedesmus spp. biomass was operated for 190 days. At first the bioreactor operated as a Rumen-like Fermenter (RF) with a Sludge Retention Time (SRT) of 7 days. The effects of simulated gastrointestinal digestion upon sialic acid and gangliosides in infant and follow-on formulas and human milk, as well as their bioaccessibility, have been evaluated. The gastric stage is the step that causes a greater decrease in sialic acid and ganglioside contents. The intestinal stage only decreases the total and individual contents of gangliosides. After gastrointestinal digestion, neither sialic acid nor gangliosides were found in the nonbioaccessible fraction. Calcium, Iron, and Zinc Uptake from Digests of Infant Formulas by Caco-2 Cells

Our aim was to estimate the bioavailability of calcium, iron, and zinc from infant formulas using a model that includes in vitro digestion and a Caco-2 cell culture to estimate the uptake. The cell culture conditions were selected, and uptake assays were carried out first with calcium, iron, and zinc standard solutions, and then with the soluble fraction of enzymatic digests of an adapted milk-based and a soy-based infant formula. It was not possible to measure the uptake of calcium, iron, and zinc from standard solutions added to the cell cultures in amounts similar to those present in infant formula digests with our method.
