Heterogeneous response to differentiation induction with different polar compounds in a clonal rat rhabdomyosarcoma cell line (BA-HAN-1C)

The clonal rat rhabdomyosarcoma cell line BA-HAN-1C was tested for its susceptibility to differentiation induction with different polar compounds. This cell line is composed of proliferating mononuclear tumour cells, some of which spontaneously fuse to form terminally differentiated postmitotic myotube-like giant cells. Exposure of BA-HAN-1C cells to dimethylsulphoxide (DMSO), hexamethylene bisacetamide (HMBA), sodium butyrate (NaBut) and N-monomethylformamide (NMF) resulted in a significant inhibition of proliferation (P less than 0.001) and in a simultaneous increase in differentiation. Abstract The new cis-dioxomolybdenum (VI) complexes [MoO2(L2)(H2O)] (2) and [MoO2(L3)(H2O)] (3) containing the tridentate hydrazone-based ligands (H2L2 = N'-(3,5-di-tert-butyl-2-hydroxybenzylidene)-4-methylbenzohydrazide and H2L3 = N'-(2-hydroxybenzylidene)-2-(hydroxyimino)propanehydrazide) have been synthesized and characterized via IR, 1H and 13C NMR spectroscopy, mass spectrometry, and single crystal X-ray diffraction analysis. Mutagenic potential of drinking water samples derived from ranneywells was studied. 100-100 l of untreated (rough) and ozone-treated as well as chlorinated-disinfected water were dropped on and adsorbed by macroreticular resin columns (Serdolit PAD-III and Amberlite XAD-2). The adsorbed material was desorbed by methanol and dichloromethane. After elimination of the solvents by vacuum distillation the adsorbed material was dissolved in dimethylsulfoxide. The mutagenic activity was tested in the Ames-Salmonella/rat liver microsome system. The tester strains were TA-98 and TA-100. Liquid nitrogen is the most common medium used by tissue banks for the storage of cryopreserved heart valves. This study evaluates the effect of the length of storage on human cryopreserved heart valves. Human tissues (14 aortic and 13 pulmonary) were frozen in a controlled-rate freezer (1 degrees C/min) and stored in the liquid phase of a nitrogen tank for 9.1+/-1.6 years. The preservative solution was medium M199 containing 5% human serum albumin and 10% Me(2)SO. After thawing in a water bath at 42 degrees C, the cryoprotectant was removed. The in vivo survival rate of rabbit morulae after vitrification in a mixture of dimethyl sulphoxide and ethylene glycol solution without protein supplement (WPS) was compared with two types of protein supplements: rabbit serum (RS) and bovine serum albumin (BSA). Significant dif- ferences were observed in the percentage of transferable embryos (undamaged embryos after devit- rification, 80.4 % versus 93.2 and 92.1 %, WPS, BSA and RS, respectively, P < 0.05) and live born rate (40.9 % versus 56.1 %, WPS and BSA, respectively, P < 0.05).
