Segmented Flow-Based Multistep Synthesis of Cadmium Selenide Quantum Dots with Narrow Particle Size Distribution

2015

A continuous-flow microreactor was used to synthetize II-VI semiconductor quantum dots (CdSe). In order to improve the size distribution of the nanoparticles, the synthesis was carried out in a two-step procedure. A seed solution was obtained in a separate nucleation step, followed by a controllable growth step. Quantum dots that are synthesized with the two-step method show a much narrower size distribution in comparison to those obtained in a conventional batch synthesis.

RAB18 impacts autophagy via lipid droplet-derived lipid transfer and is rescued by ATG9A

2018

AbstractAutophagy is a lysosomal degradation pathway that mediates protein and organelle turnover and maintains cellular homeostasis. Autophagosomes transport cargo to lysosomes and their formation is dependent on an appropriate lipid supply. Here, we show that the knockout of the RAB GTPase RAB18 interferes with lipid droplet (LD) metabolism, resulting in an impaired fatty acid mobilization. The reduced LD-derived lipid availability influences autophagy and provokes adaptive modifications of the autophagy network, which include increased ATG2B expression and ATG12-ATG5 conjugate formation as well as enhanced ATG2B and ATG9A phosphorylation. Phosphorylation of ATG9A directs this transmembra…

Lipid Droplets: A New Player in Colorectal Cancer Stem Cells Unveiled by Spectroscopic Imaging

2015

Abstract The cancer stem cell (CSC) model is describing tumors as a hierarchical organized system and CSCs are suggested to be responsible for cancer recurrence after therapy. The identification of specific markers of CSCs is therefore of paramount importance. Here, we show that high levels of lipid droplets (LDs) are a distinctive mark of CSCs in colorectal (CR) cancer. This increased lipid content was clearly revealed by label-free Raman spectroscopy and it directly correlates with well-accepted CR-CSC markers as CD133 and Wnt pathway activity. By xenotransplantation experiments, we have finally demonstrated that CR-CSCs overexpressing LDs retain most tumorigenic potential. A relevant con…

Comparative evaluation of molecular methods for the quantitative measure of torquetenovirus (TTV) viremia, the new surrogate marker of immune compete…

2019

Background Torquetenovirus (TTV) viremia is emerging as a promising tool to assess functional immune competence, to predict posttransplant immune-related complications, and eventually to customize immunosuppression. Methods In this study, 327 blood samples were tested using two real-time PCR (rtPCR) assays both targeted to the untranslated region of the TTV genome. The first assay was an in-house rtPCR developed by our group, the second one was the recently marketed TTV R-GENE assay. Results In the validation study, the TTV R-GENE showed good performances in precision and reproducibility, and sensitivity as low as 12 TTV DNA copies/mL, like previously reported for the in-house rtPCR. The Bl…

M13_Microfluidics_for_CNT

2018

Compared to pure water, the CNT dispersion has much lower interfacial tension at the dispersion – glass interface due to the presence of SDBS surfactant. Since the behavior of microfluidic system in the confined regime is driven by liquid – solid interfacial tension, the droplet formation and droplet propagation are unstable and unpredictable.

Observed and Simulated Variability of Droplet Spectral Dispersion in Convective Clouds Over the Amazon

2021

In this study, the variability of the spectral dispersion of droplet size distributions (DSDs) in convective clouds is investigated. Analyses are based on aircraft measurements of growing cumuli near the Amazon basin, and on numerical simulations of an idealized ice‐free cumulus. In cleaner clouds, the relative dispersion ϵ, defined as the ratio of the standard deviation to the mean value of the droplet diameter, is negatively correlated with the ratio of the cloud water content (qc) to the adiabatic liquid water content (qa), while no strong correlation between ϵ and qc/qa is seen in polluted clouds. Bin microphysics numerical simulations suggest that these contrasting behaviors are associ…

Cloud droplet number closure for tropical convective clouds during the ACRIDICON–CHUVA campaign

2021

The main objective of the ACRIDICON-CHUVA (Aerosol, Cloud, Precipitation, and Radiation Interactions and Dynamics of Convective Cloud Systems–Cloud Processes of the Main Precipitation Systems in Brazil: A Contribution to Cloud Resolving Modeling and to the Global Precipitation measurements) campaign in September 2014 was the investigation of aerosol-cloud-interactions in the Amazon Basin. Cloud properties near cloud base of growing convective cumuli were characterized by cloud droplet size distribution measurements using a cloud combination probe (CCP) and a cloud and aerosol spectrometer (CAS-DPOL). In the current study, an adiabatic parcel model was used to perform cloud droplet number (N…

Cryopreservation of Limonium serotinum apical meristems from in vitro plantlets using droplet-vitrification

2011

Abstract In this study in vitro shoot tips of a Sicilian genotype of Limonium serotinum were successfully cryopreserved using the droplet-vitrification technique. Growth recovery of cryopreserved shoot tips was possible only when samples were pretreated for 16 h in liquid medium with 0.3 M sucrose, then for 5 h in liquid medium with 0.7 M sucrose before performing the cryopreservation protocol. Optimal conditions included treatment for 20 min in a loading solution containing 1.9 M glycerol + 0.5 M sucrose, treatment with vitrification solution B5 (glycerol 40.0%, sucrose 40.0%, w/v) for 60 and 90 min or vitrification solution A9 (glycerol 30.0%, dimethylsulfoxide 20.0%, ethylene glycol 20.0…
