Antibodies for strain 2117-like vesiviruses (caliciviruses) in humans

The vesivirus strain 2117 has been identified as contaminant of bioreactors used for production of human drugs, due to possible contamination of the reagents used for cell cultivation. Using an ELISA assay, antibodies specific for 2117-like viruses were detected in 32/410 (7.8%) human sera, indicating exposure to these viruses.

The presence in the sera of celiac disease (CD) patients of anti-actin autoantibodies (AAAs) has been suggested as a marker of severe intestinal villus atrophy (1). AAAs have been detected with an immunofluorescence (IF) technique and seem to contribute to villus cytoskeleton damage and to the pathogenesis of intestinal damage in CD (2). The aims of the present study were to evaluate the relationship between the presence of serum IgA AAAs and severity of intestinal mucosa damage in CD patients and to compare the IF assay with a new ELISA for IgA AAA determination. The relevance of the linker tethering site in haptens was investigated for antibody generation and immunoassay development. Three derivatives of the strobilurin fungicide picoxystrobin were synthesized with the same functionalized spacer arm located at three different positions. Protein conjugates of those haptens were employed as immunogens, and novel polyclonal antibodies were produced and characterized. All haptens afforded highly specific antibodies, but different affinities to the free analyte were observed among the obtained antisera. Babesiosis due to Babesia bigemina is a relevant tick‑borne disease, affecting cattle worldwide. Many surface proteins of the pathogen including the Apical Membrane Antigen 1 (AMA‑1) ‑ have been analysed for vaccine and diagnostic purposes. This study focused on B. bigemina AMA‑1 and on its use for the assessment of diagnostic tests. After bioinformatic analyses, AMA‑1 codifying region was amplified and cloned into an expression vector used to induce protein synthesis in Escherichia coli cells. Ievads: Keratokonuss ir ektātiskā radzenes slimība, kura izraisa radzenes slāņu deģenerāciju, progresējošu radzenes biezuma samazināšanos, izveidojot konusveidīgu radzenes formu, provocējot astigmātisma attīstību un redzes pasliktināšanos. Šī slimība jau ir zināma vismaz 270 gadus, bet joprojām nav zināms konkrēts etioloģiskais faktors, tāpēc 20 gadu laikā, pētot keratokonusa patoģenēzi, tika izvirzīti vairāki iespējamie riska faktori un viens no pēdējiem ir pastāvīga acu berzēšana, kas varētu provocēt iekaisuma marķieru pastiprinātu izdalīšanos asaru šķidrumā.
