Synthesis, computational docking and biological evaluation of celastrol derivatives as dual inhibitors of SERCA and P-glycoprotein in cancer therapy.

Abstract A series of eleven celastrol derivatives was designed, synthesized, and evaluated for their in vitro cytotoxic activities against six human cancer cell lines (A549, HepG2, HepAD38, PC3, DLD-1 Bax-Bak WT and DKO) and three human normal cells (LO2, BEAS-2B, CCD19Lu). To our knowledge, six derivatives were the first example of dipeptide celastrol derivatives. Among them, compound 3 was the most promising derivative, as it exhibited a remarkable anti-proliferative activity and improved selectivity in liver cancer HepAD38 versus human normal hepatocytes, LO2. 4-(3H)-quinazolinones N-3 substituted with a five membered heterocycle: A promising scaffold towards bioactive molecules

Abstract The quinazolinone nucleus represents, among the class of fused heterocycles, a very important scaffold to obtain molecules with biological activities. A review of literature revealed how such kind of fused heterocycles, coming from natural or synthetic source, are associated with a wide range of biological activities. This review is mainly directed towards the 4-(3H)-quinazolinones N-3 substituted with a five membered heterocycle in which all the possible combinations of nitrogen, sulfur and oxygen atoms are present.

Load Carrying Capacity of Glass Columns with Rectangular, T and X Transverse Cross-sections

In this study, the load carrying capacities of glass columns with rectangular, T and X cross-sections having different lengths were calculated. The proposed analytical procedure allows to calculate the load carrying capacity associated with strength of transverse cross-section including local and global flexural and/or torsional buckling phenomenon. Moreover, initial imperfections and time dependant effects, the latter associated with the viscoelastic behaviour of interlayer (PVB), were included in the model as suggested in the literature. Settore ICAR/14 - Composizione Architettonica E UrbanaThe essay questions the future's possibilities to operate on the buildings of the second half of the twentieth century according to the Italian Code of Cultural Heritage (2004-2011). When arguing how to encourage the re-use of these artifacts which are often marked by short-life technology cycles obsolescence and abandonment the ambiguities and inconsistencies of current legislative procedures deeply stress the cultural role of research experiences that use project-oriented methodologies. The comparison between some Italian and other European cases highlights the role of a design approach in the construction of a transformative protection perspective for the built matter of the second half of the twentieth century. In a framework aiming to encourage a reconciliation between the recent built environment and the new paradigms of sustainability and circular economy architecture must recognize re-cycle practices as a main theoretical and procedural node. To modernize the late Modern built environment with contemporary construction components is not simply a matter of technical knowledge and expertise: but a theme with theoretical density aesthetic and ethical values. The architectural project in this field today must ask for greater congruence between the bureaucratic and administrative levels and the real consistency of this corpus. It is a matter of identifying strategies to equip it with capabilities meanings and contents remembering that demolition wastes have unsustainable environmental costs and uncertain results concerning the necessary new attribution of political and semantic meaning that these places await.

The definition of effective energy saving strategies capable of satisfying users’ requirements for environmental wellness is a complex task that requires the definition of well-tuned optimization algorithms. Sensory information depends on the environments observed, hence the model adopted to describe it should be adaptive and dynamic. To the Editor: During the early 20th century, at the end of World War I, and during World War II, louseborne relapsing fever (LBRF) caused by Borrelia recurrentis was a major public health problem, especially in eastern Europe and northern Africa (1,2). Currently, poor living conditions, famine, war, and refugee camps are major risk factors for epidemics of LBRF in resource-poor countries, such as those in the Horn of Africa (3,4). Increased migration from resource-poor countries and war/violence create new routes for spread of vectorborne diseases.
