Effect of addition of Lewis/Brönsted acids in the asymmetric aldol condensation catalyzed by trifluoroacetate salts of proline-based dipeptides

Abstract Proline-based dipeptides catalyze aldol condensations with good yield and stereoselectivity after addition of zinc or sodium acetate to the trifluoroacetate peptide. The chirality of the N-terminal proline in the catalyst determines the absolute configuration of the aldol product, but stereoselectivity depends on the configuration of both amino acids, and is higher for the enantiomeric pair R–S, S–R. Regarding the nature of the second component, optimal results in both yield and stereoselectivity are obtained when neutral unbranched primary amino acids are used.

Abstract The diastereoselective synthesis of hydroxy substituted γ- and δ-lactones was accomplished following two approaches. A 5- or 6-endo cyclization promoted by electrophilic selenium reagents of α- or β-hydroxy acids and a 5- or 6-exo cyclization of hydroxy esters obtained through a diastereoselective hydroxy selenenylation reaction of α- or β-hydroxy esters. Moreover, the diastereoselective methoxy selenenylation of the above compounds was investigated showing a case in which the compound that was unreactive in the hydroxy selenenylation conditions gave, in the methoxy selenenylation conditions, the deprotected diol. A simple synthetic methodology for the preparation of a polystyrene-supported L-proline material is reported, and this material has been used as catalyst in direct asymmetric aldol reactions between several ketones and arylaldehydes to furnish aldol products in high yields and stereoselectivities. Screening of solvents showed that these reactions take place only in the presence of water or methanol, at lower levels of conversion in the latter case. Virazole (Ribavirin) a Cytostatic Agent

Virazole (1-β-D-ribofuranosyl-1,2,4-triazole-3carboxamide,ribavirin) is a synthetic triazol nucleoside with a broad spectrum of antiviral activity. But virazole affects the metabolism not only of virus infected cells. Virazole strongly inhibits the cell proliferation of mouse lymphoma cells (L 5178y), which were not infected with DNA- or RNA-viruses. Starting with 3 × 103 cells/ml and an incubation period of 72 hr, the drug reduces the cell proliferation to 50% (= ED50 concentration) in a concentration of 4,7 μM.

Stereoselective Synthesis of Enantiomerically Pure Piperidine Derivatives byN-Galactosylation of Pyridones

Stereoselective desymmetrization of 4-pyridone has been achieved through selective N-galactosylation, activation of the N-(galactosyl)pyridone by O-silylation and immediate addition of Grignard compounds. Chiral piperidine derivatives, e.g. (S)-(+)-coniine and (5S,9S)-(+)-indolozidine 167B, were synthesised in enantiomerically pure form using these highly regio- and stereoselective reactions. After N-galactosylation of 2-pyridone and O-silylation of the N-galactosyl-2-pyridone, addition of a Grignard compound proceeded with high 1,4-regioselectivity and complete diastereoselectivity, to furnish 4-substituted 5,6-dehydro-2-piperidones. Ultrafast energy transfer in dansylated POPAM–eosin complexes

Abstract Excitation energy transfer (EET) in dendritic host–guest complexes has been studied. Three generations G2, G3 and G4 of dansyl substituted poly(propyleneamine) dendrimers (POPAM) were complexed with a fluorescent dye eosin in chloroform solution. Arrival of excitation from dansyls to eosin was monitored by femtosecond transient absorption spectroscopy. EET rates from the dansyls to eosin(s) are characterised by two time constants 1 ps and 6 ps independent of dendrimer generation. Relaxation processes in eosin were clearly faster when complexed with dendrimer than in solution. Changes in collagen cross-linking of articular cartilage are revealed by spectral reflectance imaging

Excessive cross-linking of collagen during aging may contribute to degeneration of articular cartilage. Traditionally, the amount of cross-links is derived by using destructive high-performance liquid chromatography (HPLC). However, a sensitive, non-destructive method could help to evaluate tissue integrity, including cross-links. We increased collagen cross-linking in bovine articular cartilage by controlled threose incubation. During the incubation, optical spectral reflectance images of the samples were captured and related to HPLC results using regression analysis.
