Prediction of the retention in reversed-phase liquid chromatography using solute-mobile phase-stationary phase polarity parameters.

Abstract A previously reported algorithm, based on the equation: log k=( log k) 0 +p(P N m −P N s ) , that relates the retention in reversed-phase liquid chromatography with solute (p), mobile phase (PmN) and stationary phase (PsN) relative polarity parameters, is improved. The retention data reported by several authors for different sets of compounds, eluted with acetonitrile–water and methanol–water mixtures, are used to test the algorithm and elaborate a database of p values. Abstract The reversed-phase chromatography of proteins by gradient elution with acidic, low-ionic-strength aqueous—organic eluents is often associated with losses of the biological activity of the protein. In this study, the enzymatic activities of catalase, horseradish peroxidase and pepsin were examined under static and dynamic column conditions on non-porous, monodisperse 1.5-μm reversed-phase silicas with various n -alkyl ligands. Abstract Reversed-phase high-performance liquid chromatography on a thermostatted octadecylsilyl column was used to separate mixtures of labelled polyunsaturated fatty acids (as their methyl esters) formed by successive desaturations and elongations of labelled linoleic (18:2 n - 6) or linolenic (18:3 n -3) acid by rat liver microsomes. Acetonitrile-water mixtures were used for elution of the esters. Unsaturated and saturated esters were detected by their refractive indices. The order of elution of fatty acid methyl esters in complex mixtures varies as a function of the chain length and unsaturation, analysis temperature, water concentration and solvent flow-rate. Separation and determination of alkylglycosides by liquid chromatography with electrospray mass spectrometric detection

The separation of alkylpolyglycosides by liquid chromatography with electrospray mass spectrometric detection, using either an alkylamide or a cyanopropyl column, and acetonitrile/water mixtures as mobile phases, was developed. Using the alkylamide column and isocratic elution, the alpha- and beta-epimers and ring isomers (pyranosides and furanosides) of the alkylmonoglycosides were resolved. The ring isomers were also resolved in a much shorter time using the cyanopropyl column with gradient elution. The elution behaviour of charged water-soluble polymers (sodium polystyrene sulphonate) in both salt-free and NaCl-containing aqueous mobile phases has been investigated by size-exclusion chromatography. A comparative analysis has been established between the results as a function of polyion and salt concentrations, which demonstrates that the chromatographic behaviour is similar in both cases. The variation of the slopes of the different concentration-dependent calibration curves is discussed in terms of polyelectrolyte conformational changes. A new high-performance size-exclusion chromatographic strategy is reported for the analysis of the hydrophobic self-associating peptide gramicidin A, incorporated into artificial phospholipid vesicles (liposomes). The method is based on the direct injection of a few microlitres of the gramicidin A-containing liposome suspension into the column, which is eluted with a non-polar solvent, such as tetrahydrofuran. Determination of major carotenoids in vegetables by capillary electrochromatography.

A simple and rapid method for the isocratic separation and determination of carotenoids (carotenes and xanthophylls) in vegetables by CEC is described. The capillary column (100 microm ID, 25 cm effective length) was packed with 3 microm Hypersil ODS particles. The optimized mobile phase contained 60% ACN, 35% THF and 5% of a 5 mM Tris aqueous buffer of pH 8. beta-Carotene, lycopene and lutein were separated with efficiencies of 66 000-128 000 plates/m within a short time (less than 12 min for the last peak eluted, 13/13'-cis-beta-carotene). The effect of pre-heating on monomer elution from bulk-fill resin composites

Background The present study was aimed to evaluate the effect of pre-heating of bulk -fill resin composites on monomer elution from them. Material and Methods Three different types of resin composites were used including Tetric N-Ceram Bulk Fill, X-tra Fill and X-tra Base. 10 cylindrical samples were prepared from each resin composites. Before light curing, 5 samples were pre-heated until reaching 68˚C, then 5 other samples were polymerized at room temperature. After 24 hours, release of UDMA, TEGDMA and BIS-GMA monomers were measured by High-Performance Liquid Chromatography analysis. The fractal calibration method applied to the characterization of polymers in solvent mixtures and in mixed gel packings by SEC.

The size-exclusion chromatographic (SEC) behaviour of different solvent/polymer systems in three packing sets has been analysed from fractal considerations. The three-column sets studied are specifically formed by: (i) 'pure' micro-styragel, (ii) 'mixed' TSK Gel H(HR + XL + HR) and (iii) mixed TSK Gel H(XL + HR + XL). The experimental data reveals that in most of the systems assayed the classical universal calibration (UC) is not fulfilled, denoting the existence of secondary effects accompanying the main SEC mechanism. Abstract We report here, for the first time, quantitative analysis of end-group functionalisation and the extent of end-group modification of polymers with molar mass up to 200,000 g mol −1 , using a combination of isothermal and temperature gradient interaction chromatography. At such high molecular weights, other common analytical techniques such as MALDI-ToF-MS and NMR spectroscopy are simply unable to offer any quantitative insight into the end-group functionality of polymers.
