Organometallic complexes with biological molecules. XV. Effects of tributyltin(IV)chloride on enzyme activity, Ca2+, and biomolecule and synthesis in…
2001
Considerable attention has been given in recent years to the possibility that xenobiotics in the environment may affect reproduction in animals. In this study, the relative impact of tributyltin(IV) (TBT) chloride, one of the most toxic environmental pollutants, was investigated using Ciona intestinalis ovary as a model system. The pleiotropic effects of TBT exposure are concentration dependent and include a decrease of ATP levels, lipid content and nucleic acid content and synthesis. In contrast, a marked increase in calcium (Ca2+) and glucose content is observed. Furthermore, TBT alters enzymatic activity, inhibiting creatine kinase and stimulating alkaline phosphatase and cholinesterase …
Cell wall xyloglucan incorporation by xyloglucan endotransglucosylase/hydrolase in pine hypocotyls
2004
Abstract In the present study, we have examined the incorporation of labelled xyloglucan oligosaccharides into the cell wall xyloglucan of pine hypocotyl sections, and the effect of plant hormones on the incorporation and on XTH enzyme activity (EC 2.4.1.207). Xyloglucan heptasaccharide [ 3 H ]XXXGol was absorbed, transported and incorporated into the cell wall xyloglucan of de-rooted pine hypocotyls. Incorporation of [ 3 H ]XXXGol into soluble and cell wall xyloglucan was induced by IAA, brassinolides and XXXG in pine hypocotyl segments. A relationship between growth induction, by IAA, brassinolides and XXXG, and both soluble and cell wall-bound XTH was found. We suggest that IAA induced-g…
Über den Ureidstoffwechsel beim Laubmoosprotonema von Funaria hydrometrica L. (Sibth.)1)1)Herrn Prof. Dr. Dr. Kurt Mothes zum 75. Geburtstag gewidmet.
1975
Summary The activity of allantoinase (EC 3.5.2.5) is influenced by light and darkness. The extracts of dark-grown moss protonema showed a very higher enzyme activity than the extracts of light-grown plants. The allantoinase activity was increased by light-dark and decreased by dark-light transitions. DMAP mimicked light effects by stabelizing the charakteristic light depending enzyme avctivities in the dark. The inhibitors DCMU, DNP and CCCP increased the enzyme activity of light-grown moss protonema, to some extend they induced a “physiological dark condition”.
All-trans to 11-cis retinol isomerization in nuclear membrane fraction from bovine retinal pigment epithelium
1991
Abstract Isomerization of all-trans to 11-cis retinol has been studied in a membrane preparation from the nuclear fraction of bovine retinal pigment epithelium. When the nuclear membrane preparation deprived of endogenous retinoids is incubated with 4·5 μ m all-trans-retinol, the mean value calculated for the isomerase activity is 1·32 nmol 11-cis retinol formed hr−1 mg protein−1. Simultaneous formation of all-trans and 11-cis retinyl esters is also observed in the nuclear preparation. When assayed under the same experimental condition, RPE 150 000 g post-nuclear sediment shows about 70% of the isomerase activity found in the nuclear membrane fraction. Treatment of the nuclear membrane frac…
Functional stability of the nitrate-reducing community in grassland soils towards high nitrate supply
2006
Abstract To study the effects of short-term fluctuation of nitrate concentrations on the nitrate-reducing community, repacked soil cores were amended with 0, 100 and 300 μ g NO 3 - ‐ N g - 1 soil and incubated for 3, 7 and 14 days, respectively. The nitrate reductase activity was determined in a laboratory-based enzyme assay. In parallel, the community structure of nitrate-reducing microorganisms was characterised by RFLP-PCR using the functional gene narG , which encodes the catalytic site of the membrane-bound nitrate reductase. The community structure remained constant over the experimental period indicating that this functional community is characterised by a high resistance towards flu…
Fruits and vegetables protect against the genotoxicity of heterocyclic aromatic amines activated by human xenobiotic-metabolizing enzymes expressed i…
2010
Heterocyclic aromatic amines (HAAs) can be formed during the cooking of meat and fish at elevated temperatures and are associated with an increased risk for cancer. On the other hand, epidemiological findings suggest that foods rich in fruits and vegetables can protect against cancer. In the present study three teas, two wines, and the juices of 15 fruits and 11 vegetables were investigated for their protective effect against the genotoxic effects of 2-amino-3-methylimidazo[4,5-f]quinoline (IQ) and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP). To closely mimic the enzymatic activation of these HAAs in humans, genetically engineered V79 Chinese hamster fibroblasts were employed tha…
Structural diversity and enzyme activity of volcanic soils at different stages of development and response to experimental disturbance
2008
We investigated the phospholipid fatty acid (PLFA) diversity and enzyme activities in soils from the volcano, Mt. Etna (Sicily). The soils were at sites which have been developing for different periods of time and have formed in volcanic lava of differing ages that have been supplemented with volcanic ejecta from subsequent eruptions. However, the plant communities indicated a marked successional difference between the sites and we have used this as a proxy for developmental stage. We have compared the structural and functional properties of the microbial communities in soils from the two sites and tested experimentally the hypothesis that the more diverse community was more resistant and r…
[35] Determination of oxidized glutathione in blood: High-performance liquid chromatography
1994
Publisher Summary The measurement of glutathione status is important in determining oxidative stress in tissues and biological fluids. The ratio of reduced to oxidized glutathione (GSH/GSSG) is a good indicator of the oxidative stress that may occur under physiological and pathological conditions. Changes in GSSG levels have been considered as intracellular signals able to modulate enzyme activity. Thus, it is important to have accurate methods to determine GSSG in biological fluids and in cells. In many cases, it is possible to use tissues, such as liver, muscle, or brain to determine GSH/GSSG. However, especially in human studies, samples from these tissues are not readily available; the …
Age-dependent changes of nuclear envelope protein phosphokinase and protein phosphatase activities. Significance for altered nucleo-cytoplasmic mRNA …
1984
Nuclear envelopes are associated with a protein phosphokinase and a phosphoprotein phosphatase, whose activities are modulated by poly(A) in an opposite manner. The activities of these enzymes were determined in nuclear ghosts from liver and oviduct of quails of different age and of different hormone status. Under optimal conditions, kinase activity was found to increase in immature animals 8-fold in response to diethylstilbestrol; co-administration of progesterone had no marked effect on enzyme activity. After the initial burst, the activity of the enzyme increased only slightly during ageing. Two proteins present in nuclear ghosts of Mr 64 000 and of Mr 106 000 are phosphorylated during t…
Alterations on AChE Activity of the Fish Anguilla anguilla as Response to Herbicide-Contaminated Water
2000
Abstract The inhibition of both total and specific acetylcholinesterase activities was measured in the whole eyes of the yellow eel Anguilla anguilla after exposure to the carbamate thiobencarb. In vivo assays were conducted under a constant flow-through system of thiobencarb-contaminated water (1/60 LC50 96 h=0.22 ppm for 96 h) followed by a recovery period in clean water (192 h more). The results indicated a measurable level of AChE activity on eyes of control eels, which resulted in a sensitive indicator of the presence of thiobencarb in the water. The pesticide induced significant inhibitory effects on AChE activity ranging from 35% in total AChE activity to 75% in specific AChE activit…
