Guanethidine was intravenously injected into rats and was subsequently determined in the hearts by extracting the tissue with trichloroacetic acid, purifying the extracts on cation exchange columns and by paper chromatography, and coupling the guanethidine with ninhydrin in alkaline solution before fluorimetric estimation. The concentration in the heart reached a maximum of 9 μg/g within 15 min of injecting 15 mg/kg, and was still 6.2 and 5.7 μg/g after 1 and 3 hours, respectively. Determination of Phenylephrine Hydrochloride by Flow Injection Analysis with Chemiluminescence Detection

Abstract A new method is proposed for the determination of phenylephrine hydrochloride by flow injection analysis with direct chemiluminescence detection. The method is based on the oxidation of the drug by potassium permanganate in sulfuric acid medium at 80°C. The calibration graph is linear over the range 0.03–8 ppm phenylephrine hydrochloride, with a relative standard deviation (n = 51, 0.5 ppm) of 1.1% and sample throughput of 134/h. The influence of 38 different foreign compounds was tested, and the method was applied to the determination of phenylephrine hydrochloride in 8 different pharmaceutical formulations.

The stimulatory effect of PGE1 on different functions of isolated guinea-pig hearts (Langendorff method, Tyrode solution) was coupled with an increase in the rate of45Ca uptake from the perfusion medium. The total myocardial Ca content and the amount of exchangeable cellular Ca were not affected. This action of PGE1 on the myocardial Ca metabolism seems to be related to the positive inotropic action of PGE1 and can most probably be explained by an increase in the membrane permeability to Ca ions (similar to the action of epinephrine).

Effects on Arterial Blood Pressure of the Methanol and Dichloromethanol Extracts from Schinus molle L. in Rats

The effects on arterial blood pressure of the methanol and dichloromethanol extracts from Schinus molle L. were analysed in urethane anaesthetized rats. In normotensive rats, the mean arterial blood pressure was significantly reduced by the i.v. administration of both extracts. The dichloromethanol extract inhibited the effects of noradrenaline on arterial blood pressure in the anaesthetized rat and it reduced the maximal contractile effect (Emax) induced by noradrenaline on rat vas deferens in the organ bath. However, the methanol extract did not modify the effects of noradrenaline in the evaluated tests.
