Catechol Acetonide Glycidyl Ether (CAGE): A Functional Epoxide Monomer for Linear and Hyperbranched Multi-Catechol Functional Polyether Architectures

A protected catechol-containing epoxide monomer, catechol acetonide glycidyl ether (CAGE), is introduced. CAGE is conveniently obtained in three steps and enables the incorporation of surface-active catechol moieties into a broad variety of hydrophilic and biocompatible polyether architectures by copolymerization. Via acidic cleavage of the acetal protecting groups, the polymer-attached catechol functionalities are liberated and available for surface attachment or metal complexation. Tritium-labelled (E)- and (Z)-2,3-dimethyl-2-phenyl oxirane 4, (E)- and (Z)-2-methyl-3-phenyl oxirane 7 and 2,2-dimethyl-3-phenyl oxirane 11 have been prepared by reduction of the corresponding bromoketones with sodium borotritide to the corresponding bromohydrins followed by cyclization to the oxiranes. These oxiranes were successfully used as diagnostic substrates to distinguish between different forms of epoxide hydrolase and glutathione transferase.

Rearrangement of 4,5-epoxy-9-trimethylsilyldecalines. Application to the synthesis of the natural eremophilane (-)-aristolochene.

Several 4,5-epoxy-9-trimethylsilyl-eudesmanes and 15-nor-eudesmanes, having different relative stereochemistry and substitution at the oxirane ring, have been prepared starting from (-)-carvone and subjected to acid-promoted rearrangement. The presence of the silicon at C9 favors two different main reaction pathways involving C14-methyl or C1-methylene migration through the stabilization of a C10 carbocation intermediate. Determination of Anticonvulsant Drugs in Pharmaceutical Preparations by Micellar Liquid Chromatography

A micellar liquid chromatographic method for quality control of pharmaceutical preparations (capsules, pills, tablets, injections, drops, and suppositories) containing the anticonvulsant drugs acetazolamide, carbamacepine, chlordiazepoxide, diazepam, ethosuximide, phenytoin, phenobarbital, and zopiclone has been developed. This methodology involves the use of micellar solutions of cetyltrimethylammonium bromide (CTAB) as mobile phases and UV detection. The proposed approach is rapid and reproducible. Sample preparation only requires dissolution with micellar solvent and adequate dilution with the mobile phase before injection into the chromatographic system.

Trans-3,4-dihydroxy-3,4-dihydrochrysene (chrysene-3,4-diol), a major metabolite of chrysene, is further metabolized by rat liver enzymes to products which effectively revert the his- Salmonella typhimurium strain TA98 to histidine prototrophy, but are only weakly mutagenic in strain TA100 and in Chinese hamster V79 cells (acquisition of resistance to 6-thioguanine). The liver enzyme mediated mutagenicity of chrysene-3,4-diol is substantially enhanced in the presence of 1,1,1-trichloropropene 2,3-oxide, an inhibitor of microsomal epoxide hydrolase. Influence of pH on the benzodiazepine-human serum albumin complex. Circular dichroism studies.

The influence of pH on the binding of benzodiazepine derivatives to HSA was studied by circular dichroism measurements and by gel filtration. The binding of nearly all benzodiazepines is increased by rising the pH from 6.60 to 8.20. For flurazepam, clonazepam, and nitrazepam this increase in binding is due to an increase of the affinities, while for the other substances the affinity remains constant and the number of binding sites is increased from one to two. The changes in binding of the benzodiazepines by rising the pH are explained by a cationic amino acid residue near or at the benzodiazepine binding site of the HSA molecule. Abstract Adduct formation of (+)-anti-benzo[a]pyrene 7,8-dihydrodiol 9,10-epoxide [BPDE] and 5′-d(CCTATCGTTATCC) or 5′-d(CCTATm5CGTTATCC) (G = binding target) has been studied. The extent of trans-BPDE-N2-dG adduct formation was higher in the oligonucleotide with 5′-d(m5CG) sequence context in both single- and double stranded form compared to the non-methylated analogue. The stimulating effect of m5dC on adduct formation has previously been demonstrated in other experimental systems.
