RP-HPLC-DAD determination of the differences in the polyphenol content of Fucus vesiculosus extracts with similar antioxidant activity.
2021
Abstract Significant quantities of bioactive compounds have been found in the chemical composition of seaweeds. This source of natural antioxidants such as polyphenols appears to attenuate lipid peroxidation caused by oxidative stress, preventing the harmful effects of a number of injuries including ischemia–reperfusion (I/R). Conventional extraction (CE) has been used for years as a traditional method for obtaining bioactive components from seaweeds. However, recent studies highlight ultrasonic-assisted extraction (UAE) as an alternative and more eco-friendly technique. Therefore, the two methods were optimised and compared to obtain a Fucus vesiculosus extract (FVE) with high antioxidant …
Determination of Caffeine in Analgesic Formulations Using the Apparent Content Curves Method
1994
Abstract The determination of caffeine by UV spectroscopy in pharmaceutical samples, containing different compounds which provide spectral interferences as aspirin, paracetamol, chlorfeniramine or propylphenazone, is carried out. The proposed procedure is based on the apparent content curves method in order to resolve binary, ternary and multicomponent mixtures. Results obtained are, in all cases, in agreement with contents found by a HPLC procedure used as reference method.
Flow Injection Fourier Transform Infrared Determination of Caffeine in Soft Drinks
1997
A fully automated procedure has been developed for FT-IR determination of caffeine in soft drinks. Samples are first degasified by filtration and then directly injected into a flow manifold and passed through a 100 mg C18 SPE cartridge, conditioned with methanol and water. After the cartridge has been cleaned with water, the caffeine is eluted with CHCl3 and stabilized with ethanol. The flow injection (FI) recording is obtained by measuring the absorbance at 1658 cm-1 with a baseline established at 1800 cm-1. Area values for the FI recording obtained between 0.4 and 1.4 min after sample injection are interpolated in the calibration graph of a series of aqueous standards of caffeine, which w…
Simultaneous determination of acetylsalicylic acid and caffeine in pharmaceuticals by flow injection with fourier transform infrared detection.
1993
Abstract A fast quality control methodology has been developed for the simultaneous determination of acetylsalicylic acid (ASA) and caffeine in pharmaceuticals by flow injection—Fourier Transform Infrared Spectrometry. The method is based on the solubilization of ASA and caffeine in CH2Cl2 and the use of a flow system to introduce samples and standards in the spectrometer. Two solutions, containing 90 and 110% of the reported concentration of the two active principles in the sample, were employed in order to control the extreme tolerance levels accepted by the International Pharmacopeia for the composition of formulations. A 300 μl volume of each solution was injected in turn, into a carrie…
Chromatographic determination of caffeine in pharmaceutical formulations using micellar mobile phases
1996
An HPLC procedure is described for the determination of caffeine in pharmaceutical preparations. A Spherisorb octadecylsilane ODS-2 C18 analytical column and spectrophotometric detection at 273 nm were used. The chromatographic behaviour of caffeine with different micellar eluents containing sodium dodecyl sulphate (SDS) is described. The determination of caffeine in pharmaceutical preparations was performed by use of a mobile phase containing 0.05 M sodium dodecylsulphate (SDS) and 1.5% propanol at pH7. At a 6.0 μg mL−1 concentration level the peak area and peak height repeatability were 2.6 and 2.4%, respectively. The application of the proposed method to the analysis of five pharmaceutic…
Evolution of phenolic compounds virgin olive oil during storage
1997
Phenolic compounds are of fundamental importance to the shelf life of virgin olive oils because of their antioxidative properties. In this paper, the evolution of simple and complex olive oil phenols during 18 mon of storage is studied by high-performance liquid chromatography (HPLC) analysis. The olive oils under examination were from various olive cultivars, harvested in two sectors in the same region at different stages of ripeness. The findings indicate that it is not the variety but rather the ripeness of the olives and the soil and climate that influence the phenol composition of virgin olive oil. In addition, a positive correlation was found between the age of the oils and the tyroso…
Enzyme-assisted extraction of polyphenol from edible lotus (Nelumbo nucifera) rhizome knot: Ultra-filtration performance and HPLC-MS2 profile
2018
Rhizome knot is always wasted as useless and inedible part of lotus root, despite its abundance of polyphenols. In this work, enzyme-assisted extraction followed by ultra-filtration was investigated to recover polyphenols from rhizome knot. Cellulase and pectinase treatment enhanced the polyphenols extraction. The 100 kDa membrane resulted in better filtration yield than 50 kDa membrane, 3.84% and 3.37%, respectively. With 100 kDa membrane, the highest filtration yield (4.08%) was achieved with a rotational speed of 600 rpm, TMP of 0.3 MPa and pH of 5. Satisfied permeate turbidity ( 90%) were obtained under these conditions. The main polyphenols identified in both rhizome knot extract and p…
Biotransformation of caffeine and theophylline in mammalian cell lines genetically engineered for expression of single cytochrome P450 isoforms
1992
Primary steps in the metabolism of caffeine and theophylline are cleavage of methyl groups and/or hydroxylation at position 8, mediated by cytochromes P450. V79 Chinese hamster cells genetically engineered for stable expression of single forms of rat cytochromes P450IA1, P450IA2 and P450IIBI and human P450IA2 and rat liver epithelial cells expressing murine P450IA2 were used to overcome problems arising in the proper allocation of metabolic pathways to specific isoforms by conventional techniques. These cell lines were exposed to caffeine and/or theophylline, and concentrations of metabolites formed in the medium were determined by HPLC. Caffeine was metabolized by human, rat and murine P45…
Biotransformation of methylxanthines in mammalian cell lines genetically engineered for expression of single cytochrome P450 isoforms. Allocation of …
1993
V79 Chinese hamster cells genetically engineered for stable expression of single forms of rat cytochromes P450IA1, P450IA2, P450IIB1, human P450IA2, and rat liver epithelial cells expressing murine P450IA2 were used to allocate metabolic pathways of methylxanthines to specific isoforms and to test the suitability of such cell lines for investigations on drug interactions occurring at the cytochrome expressed. The cell lines were exposed to caffeine and/or theophylline and concentrations of metabolites formed in the medium were determined by HPLC. Caffeine was metabolized by human, rat and murine P450IA2, resulting in the formation of four primary demethylated and hydroxylated metabolites. H…

This work deals with the synthesis, the chemical characterization of dibutyltin(IV) complex of caffeic acid (Bu2Sn(IV)HCAF, caf1) and its cytotoxic action on tumor cells. The coordination environment at the tin center was investigated by FTIR, (119)Sn{(1)H} cross polarization magic angle spinning, electrospray ionization mass spectroscopy in the solid state and UV-vis, fluorescence and (1)H, (13)C and (119)Sn NMR spectroscopy in solution phases. Density functional theory study confirmed the proposed structures in solution phase and indicated the most probably stable conformation. The effects on viability of breast cancer MDA-MB231, colorectal cancer HCT116, hepatocellular carcinoma HepG2 an…
