Sexually dimorphic effects of alcohol self-administration on cognitive processes

Chronic alcohol consumption is able to modify cognitive and emotional behaviour in humans. Many studies on gender diversity have identified swingeing differences between man and woman, not only in the propensity for alcohol abuse, but also in their behavioural effects. In this study we investigate in adult male and female rats: i) alcohol drinking behaviour and preference pattern using a 3-bottle choice paradigm with water, 10% ethanol solution and white wine (10%v/v), along a four-week period; ii) Alcohol free access (AFA) effects on locomotion and behavioural reactivity in the Open Field; iii) The influence of AFA on spatial learning and reference memory, employing the Morris Water Maze. …

Alcohol addiction: a role for acetaldehyde

Alcoholism is a chronically relapsing disorder characterized by cycles of repeated high alcohol intake and negative emotional consequences of withdrawal thought to contribute to excessive drinking and susceptibility to relapse. In the past years, the pharmacological and behavioural effects of alcohol, such us sedation, memory and learning impairment, were assigned to the main component of alcoholic drinks, ethanol. Recently acetaldehyde, the first metabolite of ethanol, seems to exert biological activity, besides its adverse effects. The aim of the present review is to elucidate the putative role of acetaldehyde in mediating the neuronal and behavioural features induced by ethanol intake.

Psychodiagnostic Assessment of Pathological Gamblers: A Focus on Personality Disorders, Clinical Syndromes and Alexithymia

Comorbid psychopathological syndromes are common in pathological gamblers (PGs), but the contribution of alexithymia, as a disorder of affect regulation, has not been fully explored yet. This study sought to examine the association between personality disorders, clinical syndromes and alexithymia levels in a group of PGs and to highlight a relationship between gambling behaviour and alexithymia scores, apart from the relationship between other disorders and gambling behaviour. Cytotoxic Activity and Composition of Petroleum Ether Extract from Magydaris tomentosa (Desf.) W. D. J. Koch (Apiaceae)

The petroleum ether extract of Magydaris tomentosa flowers (Desf.) W. D. J. Koch has been analyzed by GC-MS. It is mainly constituted by furanocoumarins such as xanthotoxin, xanthotoxol, isopimpinellin, and bergaptene. Other coumarins such as 7-methoxy-8-(2-formyl-2-methylpropyl) coumarin and osthole also occurred. The antiproliferative activity of Magydaris tomentosa flower extract has been evaluated in vitro on murine monocye/macrophages (J774A.1), human melanoma (A375) and human breast cancer (MCF-7) tumor cell lines, showing a major activity against the latter.

The Alzheimer's disease (AD) is the most common form of senile dementia.1 The most important role in AD is played by the aggregation process of beta-amyloid peptide (Aß), responsible for the cytotoxic effects.2 In this context, the purpose of this study was to synthesize new dicarbonyl compounds 1 structurally related to curcumin3, with anti-aggregation activity against Aß.Parallel studies involve the synthesis of heterocyclic-based curcumin-like molecules that are currently under investigation by means of in silico protocols in order to rationalize the ligand-biological target interactions.

Irreversible Inhibition of Epidermal Growth Factor Receptor Activity by 3-Aminopropanamides

Irreversible epidermal growth factor receptor (EGFR) inhibitors contain a reactive warhead which covalently interacts with a conserved cysteine residue in the kinase domain. The acrylamide fragment, a commonly employed warhead, effectively alkylates Cys797 of EGFR, but its reactivity can cause rapid metabolic deactivation or nonspecific reactions with off-targets. We describe here a new series of irreversible inhibitors containing a 3-aminopropanamide linked in position 6 to 4-anilinoquinazoline or 4-anilinoquinoline-3- carbonitrile driving portions. Synthesis and biological activities of a new class of heat shock protein 90 inhibitors, designed by energy-based pharmacophore virtual screening

The design through energy-based pharmacophore virtual screening has led to aminocyanopyridine derivatives as efficacious new inhibitors of Hsp90. The synthesized compounds showed a good affinity for the Hsp90 ATP binding site in the competitive binding assay. Moreover, they showed an excellent antiproliferative activity against a large number of human tumor cell lines. Further biological studies on the derivative with the higher EC50 confirmed its specific influence on the cellular pathways involving Hsp90.
