Polymer-doxycycline conjugates as fibril disrupters: an approach towards the treatment of a rare amyloidotic disease.
2014
The term amyloidosis describes neurological diseases where an abnormal protein is misfolded and accumulated as deposits in organs and tissues, known as amyloid, disrupting their normal function. In the most common familial amyloid polyneuropathy (FAP), transthyretin (TTR) displays this role primarily affecting the peripheral nervous system (PNS). Advanced stages of this inherited rare amyloidosis, present as fibril deposits that are responsible for disease progression. In order to stop disease progression, herein we designed an efficient family of nanoconjugates as fibril disrupters. These polymer conjugates are based on doxycycline (doxy), already in phase II trials for Alzheimer's disease…
Electrospinning of α,β-poly(N-2-hydroxyethyl)-dl-aspartamide-graft-polylactic acid to produce a fibrillar scaffold
2010
Abstract α,β-Poly( N -2-hydroxyethyl)- dl -aspartamide grafted with polylactic acid (PHEA- g -PLA) is a biocompatible and biodegradable amphiphilic copolymer that has been already employed to prepare a drug delivery system. In this study we have prepared for the first time a fibrillar scaffold from PHEA- g -PLA by the electrospinning of a solution of this copolymer in a mixture of N , N -dimethyl formamide (DMF) and acetone (80:20 vol/vol). The average diameter and the morphology of electrospun fibers were detected by scanning electron microscopy. Chemical degradation studies in phosphate buffer solution pH 7.4 have been performed until 15 days in order to obtain a preliminary information a…
The role of prognostic stratification on prescription of anticoagulants in older patients with atrial fibrillation : a multicenter, observational, pr…
2022
Background: Literature suggests that different risks of mortality could influence physicians in prescribing or not anticoagulants in older patients with atrial fibrillation (AF). The Multidimensional Prognostic Index (MPI) can be considered a tool for the detection of multidimensional frailty. The aim of this cross-sectional study was to evaluate whether prescription patterns of oral anticoagulants exist, based on MPI values. Methods: Older hospitalised patients (age >= 65 years) with non-valvular AF were included across 24 European centres. MPI was calculated using validated and standardised tools derived from a comprehensive geriatric assessment. Other functional and clinical information …
Predictive value of heart failure with reduced versus preserved ejection fraction for outcome in pulmonary embolism
2020
Abstract Aims This study aimed to investigate whether the risk of short‐term mortality is different in pulmonary embolism (PE) patients who have heart failure with reduced ejection fraction (HFrEF) as compared with those with heart failure with preserved ejection fraction (HFpEF). Methods and results Predictive value of HFrEF or HFpEF for 7‐day (intrahospital) and 30‐day all‐cause mortality was determined in the cohort of 1055 out of 1201 consecutive acute PE patients from the Serbian multicentre PE registry. Patients were classified into either HFrEF or HFpEF group, according to guideline‐proposed criteria. A 7‐day (intrahospital) and 30‐day all‐cause mortality was 18.5% vs. 7.3% vs. 4.5% …
Detection of Ventricular Fibrillation Using the Image from Time-Frequency Representation and Combined Classifiers without Feature Extraction
2018
Due the fact that the required therapy to treat Ventricular Fibrillation (V F) is aggressive (electric shock), the lack of a proper detection and recovering therapy could cause serious injuries to the patient or trigger a ventricular fibrillation, or even death. This work describes the development of an automatic diagnostic system for the detection of the occurrence of V F in real time by means of the time-frequency representation (T F R) image of the ECG. The main novelties are the use of the T F R image as input for a classification process, as well as the use of combined classifiers. The feature extraction stage is eliminated and, together with the use of specialized binary classifiers, …
Assisting Electrophysiological Substrate Quantification in Atrial Fibrillation Ablation
2019
[EN] Catheter ablation (CA) is the most popular treatment of atrial fibrillation (AF) with good results in paroxysmal AF, while its efficiency is significantly reduced in persistent AF. With the equipment used for CA strongly depending on electro-gram (EGM) fractionation quantification, the use of a reliable fractionation estimator is crucial to reduce the high recurrence rates in persistent AF. This work introduces a non-linear EGM fractionation quantification technique, which is based on coarse-grained correlation dimension (CGCD) computed over epochs of 1 second. Recordings were firstly normalized, denoised and lowpass filtered. The final CGCD value was calculated by the median CGCD valu…
X-ray investigations of the superstructure of collagen
2008
When compared with results from humid fibers, characteristic changes of the SAXS-pattern taken from dried collagen fibers have been observed. The average lateral width of the various reflexions increases with the order of reflexion whereby a fluctuation is superimposed, the type of which cannot easily be related to the reflexion order. Moreover, each of the reflexions is obviously laterally split into three components. An explanation of these characteristics can be given with the aid of a distinct model, assuming that the collagen fibrils have definitely varying diameters along their axis, whereby paracrystalline distortions appear, represented by nematic fluctuations. Additionally, there i…
Alteration of Ventricular Fibrillation by Flecainide, Verapamil, and Sotalol
2000
Background—The purpose of this study was to determine whether the myocardial electrophysiological properties are useful for predicting changes in the ventricular fibrillatory pattern. Methods and Results—Thirty-two Langendorff-perfused rabbit hearts were used to record ventricular fibrillatory activity with an epicardial multiple electrode. Under control conditions and after flecainide, verapamil, or d,l-sotalol, the dominant frequency (FrD), type of activation maps, conduction velocity, functional refractory period, and wavelength (WL) of excitation were determined during ventricular fibrillation (VF). Flecainide (1.9±0.3 versus 2.4±0.6 cm, P<0.05) and sotalol (2.1±0.3 versus 2.5±0.5 cm, P…
Interaction between mesenchymal cells and the posterior iris epithelium in chicken embryos
1977
The iris anlage of 3--10 day old chicken embryos was studied by both light and electron microscopy. Serial semithin sections showed that some of the mesenchymal cells overlying the eye cup moved into the primitive eye cavity by the 3rd day of incubation. On the 4th day some of these cells came into close contact with the basement membrane of the dorsal iris epithelium. The bases of the epithelial cells were flat at this stage. Towards the 10th day they formed cytoplasmic processes which did not penetrate the basement membrane. Fine mesenchymal cytoplasmic processes and a large number of extracellular fibrils developed in the epithelial--mesenchymal interface. The fine mesenchymal processes …
Experimental Study of the Effects of EIPA, Losartan, and BQ-123 on Electrophysiological Changes Induced by Myocardial Stretch
2015
