Activity of O 6 -methylguanine DNA methyltransferase in mononuclear blood cells of formaldehyde-exposed medical students

A recent study reported that exposure of student embalmers in Cincinnati to high concentrations of formaldehyde (2 mg/m3) reduced the activity of the DNA repair protein O6-methylguanine DNA methyltransferase (MGMT). Reduction in a DNA repair enzyme may strongly increase the cancer risk not only with respect to the repair-enzyme causing agent but with respect to all carcinogens causing lesions subject to repair by the enzyme in question. Thus, we examined whether formaldehyde exposure of 57 medical students during their anatomy course at two different Universities in Germany influenced MGMT activity in mononuclear blood cells. Computer assisted morphometric analysis of ram sperm heads: Evaluation of different fixative techniques

The recent development of automated systems for morphometric sperm head analysis has provided a series of objective parameters which have facilitated the standardization of morphological semen evaluation. This current work attempts to establish the optimum fixing conditions for the morphometric characterization of ram spermatozoa. Ejaculates were obtained from 5 Merino rams used for periodic collection of semen and were diluted at 1:50 with TEST medium. Lung fixation for the preservation of air spaces.

The procedures for the fixation of entire lungs of small rodents are presented together with various techniques used to verify the structural integrity of the lung tissue. To achieve this, the lungs were dissected out from rats and mice killed by ether overdose. The specimens were rinsed with isotonic saline and fixation solution under low vacuum conditions. After fixation, they were dried using alcohol and stored in a noncollapsed state (i.e., state of inhalation). Improvement of the butyl methacrylate-paraffin embedment.

The excellent butyl methacrylate-paraffin method as an embedment for light microscopy has been technically improved. More uniform and reproducible polymerization has been obtained by using a vacuum oven to degas the polymerizing mixture and to replace the air with nitrogen at 650 Torr. The amount of benzoyl peroxide required must be determined for each batch of butyl methacrylate. A teflon lined, reusable metal mold and a method of one-step blocking of tissues in preparation for sectioning are also described.

Influence of formalin fixation on the biomechanical properties of human diaphyseal bone.

Owing to the lack of fresh human bones, formalin-fixed specimens are frequently used in biomechanical testing. However, formalin fixation is assumed to affect the biomechanical properties of bone. The aim of this study was to compare axial and torsional stiffness and bone mineral density in fresh and embalmed human bones. The subtrochanteric regions of 12 pairs of fresh human femora were randomised into two groups for paired comparison. After bone mineral density measurement, one group was preserved in 4% formalin. Involvement of protein kinase Cdelta in contact-dependent inhibition of growth in human and murine fibroblasts.

There is evidence that protein kinase C delta (PKCdelta) is a tumor suppressor, although its physiological role has not been elucidated so far. Since important anti-proliferative signals are mediated by cell-cell contacts we studied whether PKCdelta is involved in contact-dependent inhibition of growth in human (FH109) and murine (NIH3T3) fibroblasts. Cell-cell contacts were imitated by the addition of glutardialdehyde-fixed cells to sparsely seeded fibroblasts. The Innervation of Taste Buds in the Soft Palate and Circumvallate Papilla of the Rat as Revealed by the Zinc Iodide-Osmium Tetroxide Technique.

The taste buds in the soft palate and the circumvallate papillae of the rat were investigated by the zinc iodide-osmium tetroxide technique. In addition, electron micrographs of taste buds stained with this method were presented for the first time. Differences in taste bud structures were found between the examined regions. The taste buds of the soft palate showed a complicated plexus of intragemmal nerve fibers. Some fibers exhibited terminal polymorphic swellings. Single branches could be traced close to the space of the taste pore.
