Polymeric Selectin Ligands Mimicking Complex Carbohydrates: From Selectin Binders to Modifiers of Macrophage Migration

Novel polymeric cell adhesion inhibitors were developed in which the selectin tetrasaccharide sialyl-LewisX (SLeX ) is multivalently presented on a biocompatible poly(2-hydroxypropyl)methacrylamide (PHPMA) backbone either alone (P1) or in combination with O-sulfated tyramine side chains (P2). Prognostic impact of phosphorylated HER-2 in HER-2+ primary breast cancer.

Abstract Purpose. Tyrosine 1248 is one of the autophosphorylation sites of human epidermal growth factor receptor (HER)-2. We determined the prognostic value of the expression level of tyrosine 1248–phosphorylated HER-2 (pHER-2) in patients with HER-2+ primary breast cancer using a reverse-phase protein array. Patients and Methods. The optimal cutoff value of pHER-2 for segregating disease-free survival (DFS) was determined by receiver operating characteristic (ROC) curve analysis. HER2 amplification in recurrent breast cancer following breast-conserving therapy correlates with distant metastasis and poor survival.

The authors analyzed the HER2 status in early-stage nonrecurrent and recurrent breast cancer groups following breast-conserving treatment. Retrospective analyses of a group of 36 invasive early breast cancer (IBC) patients who developed a local recurrence as a first event and of a random control group of 69 IBC patients were made. Autologous Transplantation of In Vivo Purged PBSC in CML

To determine the effectiveness of different methods for the detection of tumor cell contamination of collected peripheral stem cells, we performed a study on 39 chronic myelogenous leukemia (CML) patients who were consecutively treated at our department. Analyses of tumor cell contamination by fluorescence in situ hybridization (FISH), conventional cytogenetics, and polymerase chain reaction (PCR) showed marked differences in the percentage of evaluable results: Quantitative analysis of tumor cell contamination was feasible in 60 of 105 (57%) samples evaluated with the use of conventional cytogenetic analysis and in 105 of 107 (98%) samples analyzed by FISH. Response to lenalidomide in myelodysplastic syndromes with del(5q): influence of cytogenetics and mutations.

Summary Lenalidomide is an effective drug in low-risk myelodysplastic syndromes (MDS) with isolated del(5q), although not all patients respond. Studies have suggested a role for TP53 mutations and karyotype complexity in disease progression and outcome. In order to assess the impact of complex karyotypes on treatment response and disease progression in 52 lenalidomide-treated patients with del(5q) MDS, conventional G-banding cytogenetics (CC), single nucleotide polymorphism array (SNP-A), and genomic sequencing methods were used. Experimental techniques for testing the sensitivity of bladder tumours to antineoplastic drugs

A number of laboratory tests can be employed to examine the sensitivity of human bladder tumour cells to various chemotherapeutic agents.-Their principles and methods, and some preliminary results, are described with special reference to certain in vitro and in vivo cytotoxicity tests and to heterotransplantation in the hamster. Satisfactory agreement has sometimes been observed between experimental results and clinical responses, but our experience is still very limited.-The employment of several such tests would probably lead to a greater degree of reliability in the laboratory assessment of the sensitivity of bladder tumours to cytotoxic drugs.

High-grade gliomas (HGGs) are the most frequent primary malignant brain tumors in adults, which lead to death within two years of diagnosis. Maximal safe resection of malignant gliomas as the first step of multimodal therapy is an accepted goal in malignant glioma surgery. Gross total resection has an important role in improving overall survival (OS) and progression-free survival (PFS), but identification of tumor borders is particularly difficult in HGGS. Si photomultipliers for bio-sensing applications

In this paper, silicon photomultipliers (SiPM) are proposed as optical detectors for bio sensing. Optical transduction is the most used detection mechanism in many biosensor applications, such as DNA microarray and real-time polymerase chain reaction. The performances of a 25 pixels device used for both applications are studied. The results confirm that the SiPM is more sensitive than the traditionally employed detectors. In fact, it is able to experimentally detect 1 nM and 100 fM of fluorophore concentrations in dried samples and solutions, respectively. We present and discuss in details the detector configuration and its characterization as fluorescence detector for bio sensing.

Very high sensitivity in collinear laser spectroscopy: resonance detection by particle counting techniques

Abstract Fast-beam collinear laser spectroscopy with resonance detection by counting of fluorescence photons provides a versatile tool for precise determinations of nuclear ground-state properties. The intrinsic high sensitivity of this method can be further increased by almost three orders of magnitude if measuring schemes based on ion or atom counting are introduced. The optical pumping from one atomic state to another at an appreciably different excitation energy is detected via state-selective collisional charge exchange processes and charge-state separated atom or ion counting. Applications of these techniques on alkaline earth, noble gas, mercury and thallium beams is examined.
