The TRAPSENSOR facility: an open-ring 7 tesla Penning trap for laserbased precision experiments

APenning-trap facility for high-precision mass spectrometry based on a novel detection method has been built. This method consists in measuring motional frequencies of singly-charged ions trapped in strong magnetic fields through the fluorescence photons from laser-cooled 40Ca+ ions, to overcome limitations faced in electronic single-ion detection techniques. De novo CCND2 mutations leading to stabilization of cyclin D2 cause megalencephaly-polymicrogyria-polydactyly-hydrocephalus syndrome

Activating mutations in genes encoding phosphatidylinositol 3-kinase (PI3K)-AKT pathway components cause megalencephaly-polymicrogyria-polydactyly-hydrocephalus syndrome (MPPH, OMIM 603387)(1-3). Here we report that individuals with MPPH lacking upstream PI3K-AKT pathway mutations carry de novo mutations in CCND2 (encoding cyclin D2) that are clustered around a residue that can be phosphorylated by glycogen synthase kinase 313 (GSK-3 beta)(4). Mutant CCND2 was resistant to proteasomal degradation in vitro compared to wild-type CCND2. In situ analysis of the bacterial communities associated to farmed eel by whole-cell hybridization.

Bacterial communities in water samples and eel slime were investigated by fluorescence in situ hybridization of whole bacterial cells in an eel intensive culture system over 1 year. A newly developed probe, matching 27 Vibrio spp., and a specific probe for Vibrio vulnificus were used. Phylogenetic probes complementary to selected regions of the 16S and 23S ribosomal RNA revealed that Proteobacteria of the alpha and beta subclass were predominant in water and eel slime. Members of the gamma subclass (e.g. vibrios and aeromonads) were more abundant in eel slime, although no V. vulnificus was detected.

We report the basic spectroscopic properties of a home-made holmium-aluminum-germanium co-doped silica fiber, designed for laser applications. We present the ground-state and excitedstate absorption spectra of the fiber along with the upconversion emission spectrum under laser action, which served for making an energy levels’ diagram of Ho3+ ion. We discuss the ways to assess the kinetics of Ho3+ 2-μm fluorescence, focusing on measuring fluorescence lifetime from 5I7 level and nonradiative decay time from 5I6 level. Lielais laimiņš (Sedum maximum) kā modeļsuga smago metālu ietekmes un uzņemšanas fizioloģijas pētījumos

Darbā ir apkopota informācija par augu hiperakumulācijas spējām. Darba mērķis bija pārbaudīt iespēju izmantot lielo laimiņu smago metālu ietekmes un uzņemšanas pētījumos kontrolētos apstākļos un eksperimentāli noteikt biogēno metālu varu, cinku, mangānu un nātriju uzkrāšanos augos, kas audzēti metālu dažādu koncentrāciju ietekmē. Fizioloģiskās atbildes pētījumiem mērīja hlorofila saturu un hlorofila a fluorescenci, beigās noteica metālu saturu dažādās auga daļās. Darba rezultātā secināts, ka lielais laimiņš spēj efektīvi uzkrāt mangānu un cinku virsmezes orgānos, mangāns tika uzkrāts lielos daudzumos. NaCl un smago metālu sāļu ietekme uz piekrastes mitrāja auga krastmalas skābenes Rumex hydrolapathum augšanu un jonu uzkrāšanos

Pētījuma mērķis bija kontrolētos apstākļos pārbaudīt krastmalas skābenes izturību un jonu uzkrāšanos NaCl un smago metālu (kadmija, hroma, niķeļa un svina) sāļu ietekmē. Augus audzēja automātiskajā siltumnīcā, kur tos apstrādāja ar dažādām attiecīgās vielas koncentrācijām un mērīja hlorofila koncentrāciju un hlorofila a fluorescenci. Pēc novākšanas žāvētām auga daļām veica jonu analīzi. Krastmalas skābene izvairījās no jonu uzkrāšanas jaunajās lapās visos variantos. Savukārt uzkrāšanas vieta atšķīrās – Na, Cd un Ni maksimāli uzkrājās vecajās, nokaltušajās lapās, bet Cr un Pb saknēs. Detection of endophytic bacteria in leaves of Vitis vinifera by using fluorescence in situ hybridization

Previous investigation on five cultivars of healthy Sicilian grapevine allowed the isolation of endophytic bacteria belonging to Bacillus genus from different organs (bud, leaf, stalk and shoot). The aim of this work was to use fluorescence in situ hybridization (FISH) experiments in healthy and damaged leaf tissues of Vitis vinifera to visualize and localize bacteria associated with plant materials. The leaves were cleared to minimize the autofluorescence of the plant fragments.
